Project description:Cells of the trophoblast lineage constitute the major part of placental tissues in higher mammals. Recent derivation of human trophoblast stem cells (TSC) from placental cytotrophoblasts (CT) and from blastocyst opens new opportunities for studying development and function of human placenta. Here we report that inhibition of TGF pathway leads to direct and robust conversion of primed human pluripotent stem cells into TSC. The resulting cell lines exhibit self-renewal, are able to differentiate into the main trophoblast lineages, and present RNA and epigenetic profiles that are indistinguishable from the TSC lines derived from placenta or blastocyst. Furthermore, activation of YAP pathway is necessary and sufficient for this conversion.

Project description:Accurate annotation of regulatory RNAs is a complex task but nevertheless essential as sRNA molecular and functional studies ensue from it. Several formerly considered small RNAs (sRNA) are now known to be parts of UTR transcripts. In light of experimental data, we review hundreds of Staphylococcus aureus putative regulatory RNAs. We pinpoint those that are likely acting in trans and are not expressed from the opposite strand of a coding gene. We conclude that HG003, a NCTC8325 derivative strain, has about 50 bona fide sRNAs, indicating that these RNAs are less numerous than commonly stated.

Project description:Memory B cells (MBCs) are key cellular components of long-term humoral immunity that dominate recall responses by rapidly differentiating into effector cells. As the biology of MBCs has been mainly studied in the context of model antigen immunizations, the dynamics of these cells during infection remains largely unexplored. Here, we combined influenza and SARS-CoV-2 infection models with fluorescent-reporter mice to identify MBCs regardless of antigen-specificity. scRNA-seq analysis and confocal imaging revealed that two main transcriptionally distinct subsets of MBCs colonize the lung airways after infection. These subsets were class-switched, somatically mutated and preferentially differentiated into plasma rather than germinal center cells upon activation. Combined analysis of antigen-specificity and B cell receptor repertoire unveiled a highly permissive selection process that segregated these subsets into “bona fide” virus-specific MBCs and “bystander” MBCs with no apparent specificity for eliciting viruses. Thus, diverse transcriptional programs in MBCs are not linked to specific effector fates but rather to divergent strategies of the immune system to simultaneously provide rapid protection from re-infection while diversifying the initial B cell repertoire.
