Project description:The gastrointestinal (GI) microbiome contributes significantly to host nutrition and health. However, relationships involving GI microbes, their hosts and host macrohabitats remain to be established. Here, we define clear patterns of variation in the GI microbiomes of six groups of Mexican black howler monkeys (Alouatta pigra) occupying a gradation of habitats including a continuous evergreen rainforest, an evergreen rainforest fragment, a continuous semi-deciduous forest and captivity. High throughput microbial 16S ribosomal RNA gene sequencing indicated that diversity, richness and composition of howler GI microbiomes varied with host habitat in relation to diet. Howlers occupying suboptimal habitats consumed less diverse diets and correspondingly had less diverse gut microbiomes. Quantitative real-time PCR also revealed a reduction in the number of genes related to butyrate production and hydrogen metabolism in the microbiomes of howlers occupying suboptimal habitats, which may impact host health.

Project description:The availability and spatial distribution of food resources affect animal behavior and survival. Black howler monkeys (Alouatta pigra) have a foraging strategy to balance their nutrient intake that involves mixing their consumption of leaves and fruits. The spatial aggregation of food items should impact this strategy, but how it does so is largely unknown. We quantified how leaf and fruit intake combined (here termed food set selection) was spatially aggregated in patches and how food aggregation varied across seasons. Using variograms we estimated patch diameter and with Generalized Least Square models determined the effect of food spatial aggregation on food selection. Only fruits were structured in patches in the season of highest availability (dry-season). The patches of food set selection had a diameter between 6.9 and 14 m and were explained by those of mature fruit availability which were between 18 and 19 m in diameter. Our results suggest that the spatial pattern of food selection is influenced by patches of large fruit-bearing trees, not by particular species. Fruit also occur along spatial gradients, but these do not explain food selection, suggesting that howlers maximize food intake in response to local aggregation of fruit that are limiting during certain seasons. We demonstrate how the independent spatial modelling of resources and behavior enables the definition of patches and testing their spatial relationship.

Project description:We present the first description of a diurnal live birth of a wild black howler monkey (Alouatta pigra). The mother formed part of a group of five individuals inhabiting an anthropogenic setting in the tropical lowlands of southeast Mexico. A total of 7 h and 50 min passed from the rupture of the amniotic sac early in the day to the crowning of the infant from the birth canal. The delivery of the infant lasted ~ 3 min. We describe the event while referencing images and time points in a supplementary video recording. We place our findings in the context of the available reports of live births in the Alouatta genus, time of day, birth duration, and group activity budget on the day of the birth. While primates tend to give birth at night to reduce complications from group interactions, the observed birth took place during the day, which may have been possible due to an alteration in group time allocation. Our report provides in-depth details of the events of a birth and important information regarding the natural history of the black howler monkey.

Project description:Mammals rely on the metabolic functions of their gut microbiota to meet their energetic needs and digest potentially toxic components in their diet. The gut microbiome plastically responds to shifts in host diet and may buffer variation in energy and nutrient availability. However, it is unclear how seasonal differences in the gut microbiome influence microbial metabolism and nutrients available to hosts. In this study, we examine seasonal variation in the gut metabolome of black howler monkeys (Alouatta pigra) to determine whether those variations are associated with differences in gut microbiome composition and nutrient intake, and if plasticity in the gut microbiome buffers shortfalls in energy or nutrient intake. We integrated data on the metabolome of 81 faecal samples from 16 individuals collected across three distinct seasons with gut microbiome, nutrient intake and plant metabolite consumption data from the same period. Faecal metabolite profiles differed significantly between seasons and were strongly associated with changes in plant metabolite consumption. However, microbial community composition and faecal metabolite composition were not strongly associated. Additionally, the connectivity and stability of faecal metabolome networks varied seasonally, with network connectivity being highest during the dry, fruit-dominated season when black howler monkey diets were calorically and nutritionally constrained. Network stability was highest during the dry, leaf-dominated season when most nutrients were being consumed at intermediate rates. Our results suggest that the gut microbiome buffers seasonal variation in dietary intake, and that the buffering effect is most limited when host diet becomes calorically or nutritionally restricted.

Project description:In addition to macronutrients, foods consist of a complex set of chemical compounds that can influence dietary selectivity and consumer physiology. Metabolomics allow us to describe this complexity by quantifying all small molecules, or metabolites, in a food item. In this study we use GC-MS based metabolomics to describe the metabolite profiles of foods consumed by one population of Mexican black howler monkeys (Alouatta pigra) over a 10-month period. Our data indicate that each food exhibited a distinct metabolite profile, and the average weekly intake of metabolites such as neochlorogenic acid and serotonin (5-hydroxytryptamine) was correlated with the consumption of certain plant parts. We speculate that these patterns result in temporal changes in howler monkey physiology such as food retention time. In contrast, variation in the weekly intake of metabolites such as oxalic acid was 70% less than variation in the concentration of the same metabolites across food items, suggesting that howler monkeys regulated the intake of these metabolites, possibly to avoid physiological consequences such as kidney stone formation. Finally, seasonal variation in the consumption of individual nutrient and non-nutrient metabolites were correlated with changes in the relative abundances of associated gut microbial taxa, implying indirect effects of food item metabolites on howler monkey nutritional ecology that likely drive foraging decisions. While additional research is needed to validate these findings, the patterns we report serve as important baseline data for understanding the effects of plant metabolites on the food choice in primates.
