Cold Storage Effects on Fitness of the Whitefly Parasitoids Encarsia sophia and Eretmocerus hayati.

Project description:Successful biological control of the whitefly Bemisia tabaci involves the mass rearing of biocontrol agents in large numbers for field release. Cold storage of the biocontrol agents is often necessary to provide a sufficient number of biocontrol agents during an eventual pest outbreak. In this study, the fitness of two whitefly parasitoids Encarsia sophia Girault and Dodd (Hymenoptera: Aphelinidae) and Eretmocerus hayati Zolnerowich and Rose (Hymenoptera: Aphelinidae) was evaluated under fluctuating cold storage temperatures. The emergence rate of old pupae of either species was not affected when stored at 12, 10, 8 and 6 °C for 1 week. Cold storage had no effect on the longevity of the emerging adult En. sophia except young pupae stored at 4 °C, while Er. hayati was negatively affected after 2 weeks of storage time at all temperatures. Parasitism by adults emerging from older pupae stored at 12 °C for 1 week was equivalent to the control. Combined with the results for the emergence time, we suggest that the old pupal stage of En. sophia and Er. hayati could be stored at 12 and 10 °C, respectively (transferred every 22 h to 26 ± 1 °C for 2 h), for 1 week, with no or little adverse effect.

Isolation and characterization of nine microsatellite loci for a parasitoid wasp, Encarsia smithi (Silvestri) (Hymenoptera: Aphelinidae).

Project description:The parasitoid wasp Encarsia smithi is an important agent in the classical biological control of two species of invasive spiny whiteflies, Aleurocanthus spiniferus and Aleurocanthus camelliae. To evaluate the performance of parasitism indexed by genetic diversity, a highly polymorphic genetic marker is required. In this report, nine microsatellite loci are described for E. smithi. The microsatellite loci were obtained through the construction of an enriched library and exhibited polymorphisms (2-6 alleles per locus) and high levels of expected heterozygosities (0.203-0.780, average 0.537). Linkage disequilibrium and null alleles were not detected in these microsatellite loci. The isolated microsatellite markers may be useful to estimate the genetic diversity of E. smithi.

Effects of plant virus and its insect vector on Encarsia formosa, a biocontrol agent of whiteflies.

Project description:In this study, we investigated the tritrophic interactions among a persistently transmitted plant virus, Tomato yellow leaf curl virus (TYLCV), its insect vector, the sweetpotato whitefly Bemisia tabaci, and a parasitoid, Encarsia formosa Gahan, one of the most extensively used biological control agents. As an emerging invasive pest worldwide, the two most damaging whiteflies are B. tabaci B and Q cryptic species. On healthy tomato plants, parasitoid-induced mortality was significantly higher in B. tabaci B than in Q. In contrast, similar mortality levels of B and Q were observed on TYLCV-infected plants. A higher rate of parasitism was consistently observed in B, independent of the TYLCV infection. Similarly, the life history traits of E. formosa were influenced by both TYLCV and the two cryptic species of B. tabaci. Specifically, E. formosa parasitizing B had a greater adult longevity and shorter developmental time on healthy plants, whereas the parasitoids developing from Q has a greater adult longevity on TYLCV-infected plants. The emergence rate of E. formosa was unaffected by either B. tabaci cryptic species or the virus. These results suggest that the vector-borne pathogen can manipulate the host suitability of a parasitoid and hence the parasitoid-host interactions.
