Project description:Key regulatory decisions during cleavage divisions in mammalian embryogenesis determine the fate of preimplantation embryonic cells. Single-cell RNA-sequencing of early stage—2-cell, 4-cell, and 8-cell—blastomeres show that the aryl hydrocarbon receptor (AHR), traditionally considered as an environmental sensor, directs the segregation of 4-cell blastomeres. Disruption of endogenous AHR functions in Ahr knockout embryos or in embryos from dams exposed to dioxin, the prototypic xenobiotic AHR agonist, significantly impairs blastocyst formation by causing repression of OCT4 expression, decline of CDX2 expression, loss of OCT4 and CDX2 transcriptional heterogeneity, and incidence of nonspecific pluripotency downregulation. Trajectory and gene variability analyses further confirm that deregulation of OCT4 functions and changes of transcriptional heterogeneity resulting from disruption of AHR functions restrict the emergence of differentiating blastomeres within these embryos. It appears that AHR governs the differentiation of progenitor blastomeres and that interference with AHR functions significantly perturbs embryogenesis.

An in vitro model of human hematopoiesis identifies a regulatory role for the aryl hydrocarbon receptor

Project description:An in vitro model of human hematopoietic stem cell differentiation is found to allow development of multiple immune lineages through use of single-cell methods. Using this model, we demonstrate that activation of aryl hydrocarbon receptor (AHR) signaling drives human hematopoietic stem and progenitor cells (HSPCs) to differentiate towards myeloid lineages at the expense of lymphoid lineages. Gene perturbations in progenitor cells and lineage-specified immune cells by AHR ligand, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) are identified.

Project description:As part of a study of the role of the aryl hydrocarbon receptor (Ahr) in maintenance and senescence of hematopoietic stem cells (HSC), global gene expression profiling was done with HSC isolated from Ahr-knockout and wild-type mice. HSC from young-adult (8 wk old) AhR-KO mice had changes in expression of many genes related to HSC maintenance, consistent with the phenotype observed in aging Ahr-KO mice: decreased survival rate, splenomegaly, increased circulating white blood cells, hematopoietic cell accumulation in tissues, anemia, increased numbers of stem/progenitor and lineage-committed cells in bone marrow, decreased erythroid progenitor cells in bone marrow, and decreased self-renewal capacity of HSC. 7 samples: 3 Ahr knockout, 4 wild-type

Project description:[original Title] Comparison of expression data of primary murine melanocytes from aryl hydrocarbon deficient mice and corresponding wild-type C57BL/6 mice Melanin is produced exclusively by melanocytes and melanogenesis is the vital response to protect skin cells against Ultraviolet B (UVB)-induced DNA damage. The aryl hydrocarbon receptor (AhR) is a transcription factor, which may be involved in the physiological tanning response. Normal murine melanocytes express functional AhR. We tested gene expression in WT versus AhR-deficient mice primary murine melanocytes, isolated from the skin and cultivated for several passages. Skin epidermal cells from 2 individual C57BL/6 mice and 2 individual AhR-deficient mice (deletion of exon2, AhRtm1Bra) were grown for 6-8 weeks in selection medium to propagate melanocytes.
