Project description:Specific markers and in-depth analysis of adventitial fibroblast heterogeneity are underexplored. Using single-cell RNA-sequencing of enriched fraction of adventitial mesenchymal cells, we defined and quantified novel markers in healthy murine adventitia. PHATE dimensionality reduction and subsequent clustering characterized heterogeneity, predicting three novel differentiation trajectories. Gene ontology (GO) analysis supported divergent functional profiles, related to vascular development, antigen presentation and collagen fibril organization. Vascular pathologies, including ageing and hypercholesterolemia, differentially affected trajectory presence in vivo. We validated increased presence of the trajectory with collagen gene ontology prior to plaque development, and its association with increased adventitial collagen accumulation, while trajectory presence diminished in advanced atherosclerosis. Fibroblast trajectory gene-sets were also observed in human advanced atherosclerosis, where aforementioned trajectory negatively correlated to detrimental plaque traits. We have therefore unveiled the basis for fibroblast heterogeneity in health and vascular pathologies, and our data suggest that trajectory regulation will be instrumental for trajectory-specific interventions.

Captured fragments are sequenced using Illumina paired-end approach and mapped to human genome (hg19). Results. Hypomethylation of chromosomal DNA predominates in atherosclerotic plaques. Project description:Abnormal epigenetic gene regulation may play a pivotal role in atherogenesis. In particular, global DNA hypomethylation potentially leading to proatherogenic gene expression, occurs in atherosclerotic lesions in humans and animal models. In order to identify genomic sequences targeted for DNA hypomethylation in atherosclerosis, we analysed the methylation status of CpG islands (CGIs) in 45 human arteries with advanced atherosclerotic lesions and 16 normal counterparts by a microarray approach. Methylation data for 10,367 CGIs revealed that a subset (1.5%) of such sequences was hypermethylated in normal arteries, in accordance with data previously obtained in peripheral blood cells. Ninety-four per cent of this CGI subset was demethylated in atherosclerotic tissue, while only 17 of normally hypomethylated CGIs was hypermethylated in diseased tissue. A functional classification of genes physically associated with differentially methylated CGIs revealed a bias towards transcription factors (42%). The latter include HOX members, NOTCH1 and FOXP1, which are known to regulate angiogenesis, dedifferentiation, cell migration and macrophage function. The methylation status of selected CGIs was validated in further 10 subjects for either group. Expression patterns of these factors were compatible with the observed differential methylation. Our data suggest that one of the molecular changes associated with aberrant DNA methylation in advanced atherosclerosis is the regulation of critical transcription factor genes for the induction of a proatherogenic cellular phenotype. Two-condition experiment, i.e. normal vs. atherosclerotic arteries. 16 and 45 normal and atherosclerotic samples, respectively, were pooled and used to interrogate CpG island arrays in triplicate, for a total of six arrays. Each array was co-hybridized with untreated DNA (reference) and hypermethylated DNA obtained by biochemical filtration.

Project description:The rupture of unstable atherosclerotic plaques, leading to debilitating or fatal thrombotic events, is a major health burden worldwide. Limited understanding as to the molecular drivers of plaque instability and rupture hinders efforts in diagnosis and treatment prior to thrombotic events. Utilising an advanced pre-clinical mouse model (Tandem stenosis (TS) model), which presents human-like unstable atherosclerotic disease, we apply high-end omic methods to characterize the molecular signatures associated with plaque instability in atherosclerotic arteries. Through quantitative proteomic profiling, we depict unique proteome signatures of unstable plaques compared to stable plaques and healthy arteries. Coupled with single-cell RNA-sequencing of leukocytes, we describe the heterodimer complex S100a8/S100a9 as unique to unstable plaque, with neutrophils implicated as the transcriptional drivers of S100a8/a9 expression. We confirm S100a9 expression in human carotid atherosclerotic plaques and we further utilise the TS pre-clinical model to pharmacologically inhibit S100a8/S100a9, resulting in plaque stabilisation. Thus, we establish the TS model as a sophisticated translational tool for the profiling of unstable atherosclerotic plaques and demonstrate that unstable and stable atherosclerosis are highly different disease entities.
