Project description:Purpose: To identify abiotic stress responsive and tissue specific miRNAs at genome wide level in wheat (Triticum aestivum) Results: Small RNA libraries were constructed from four tissues (root, shoot, mature leaf and spikelets) and three stress treatments of wheat seedlings (control, high temperature, salinity and water-deficit). A total of 59.5 million reads were obtained by high throughput sequencing of eight wheat libraries, of which 32.5 million reads were found to be unique. Using UEA sRNA workbench we identified 47 conserved miRNAs belonging to 20 families, 1030 candidate novel and 51 true novel miRNAs. Several of these miRNAs displayed tissue specific expression whereas few were found to be responsive to abiotic stress treatments. Target genes were predicted for miRNAs identified in this study and their grouping into functional categories revealed that the putative targets were involved in diverse biological processes. RLM-RACE of predicted targets of three conserved miRNAs (miR156, miR160 and miR164) confirmed their mRNA cleavage, thus indicating their regulation at post-transcriptional level by corresponding miRNAs. Expression profiling of confirmed target genes of these miRNAs was also performed. Conclusions: This is the first comprehensive study on profiling of miRNAs in a variety of tissues and in response to several abiotic stresses in wheat. Our findings provide valuable resource for better understanding on the role of miRNAs in stress tolerance as well as plant development. Additionally, this information could be utilized for designing wheat plants for enhanced abiotic stress tolerance and higher productivity.

Project description:Work-related stress in police officers has been recognized as a risk factor for mental and physical illness, compromising work performance and public safety. While genetic, individual and environmental factors may contribute to stress, basic pathophysiological mechanisms are still not completely understood. Previous studies have shown the potential of microRNAs (miRNAs) as biomarkers and/or mediator for stress responses. In this study, we aimed at identifying possible dysregulation of salivary miRNAs associated with work-related stress in police officers. The study was conducted in a cohort composed of police officers in service at the mobile unit of Genoa in 2009, whose response to occupational stress has been continuously monitored since 2009 using the Demand-Control and the Effort-Reward Imbalance models. 113 male police officers in service in 2022 were recruited and divided in three groups according to their response to stress (low, intermediate and high stress response). miRNAs were extracted from saliva samples and analyzed by small RNA sequencing followed by an integrated bioinformatics/biostatistics analysis. The high stress response group was characterized by 18 differentially expressed miRNAs compared with the low stress response group, with four miRNAs (miR-10400-5p, miR-1290, miR-6074-5p, and miR-9902) at higher levels and fourteen miRNAs (miR-203a-3p, miR-7-5p, miR-143-3p, miR-181a-5p, miR-142-5p, miR-223-5p, let-7f-5p, let-7g-5p, miR-26b-5p, miR-27a-3p, miR-30e-5p, miR-26a-5p, miR-142-3p, and miR-21-5p) at lower levels. We also discovered sets of miRNAs that exhibited dynamic expression changes in response to intermediate and low stress. Functional enrichment analysis identified processes related to apoptosis, cellular response to stress, negative regulation of gene expression, responses to abiotic stimuli, positive regulation of catabolism as commonly regulated by the modulated miRNAs. Work-related stress response was associated with specific salivary miRNA expression patterns, with a general decrease of miRNA levels in the highly distressed police officers. These results suggest salivary miRNA profiling as a non-invasive and useful biomarker of stress and further characterize the link between chronic psychosocial stress and cellular and behavioral response mechanisms.

Approaches such as array-based transcript profiling allow assessing the expression of thousands of genes in control and stress tissues. The variety Aragonês, used in wine production, was subjected to controlled individual abiotic stresses, WS and HS. Project description:Abiotic stress exposure of plants induces metabolic reprogramming which is tightly regulated by signalling cascades connecting transcriptional with translational and metabolic regulation. Complexity of such interconnected metabolic networks impedes the functional understanding of molecular plant stress response compromising the design of breeding strategies and biotechnological processes. Thus, defining a molecular network to enable the prediction of a plant’s stress mode promises to promote the understanding of stress responsive biochemical regulation and its technological application. Arabidopsis wild type plants and two mutant lines with deficiency in sucrose or starch metabolism were grown under ambient and cold/high light stress conditions. Stress-induced dynamics of the primary metabolome and the proteome were quantified in a mass spectrometry-based high-throughput experiment. Wild type data were used to train a machine learning algorithm to classify mutant lines under control and stress conditions. Multivariate analysis and classification identified a module consisting of 23 proteins enabling the reliable prediction of coupled temperature and light stress conditions. 18 of these 23 proteins displayed putative protein-protein interactions connecting transcriptional regulation with regulation of primary and secondary metabolism under stress. The identified stress-responsive core module provides evidence for predictability of complex biochemical regulation during environmental fluctuation.

Project description:Sunflower is an important source of vegetable oil worlwide. A differential organ-specific sunflower ESTs was previously generated by a subtractive hybridization method, including a considerable number of abiotic stress associated sequences. The objective of this work is to analyze the sunflower gene expression of previously identified candidate genes under a comprehensive microarray analysis of the leaf transcriptoma under cold and salinity stresses, considering the impacts of these abiotic stresses on sunflower yield in many productive areas. The aimed of this work is to perform genome analysis of sunflower based on its functional regions and the characterization of the sunflower transcriptoma profiles for different organ-specific genes. Abiotic-related expressed genes were the target of this characterization through a gene expression analysis of the local EST bank (annotated according to Gene Ontology Annotation) using a cDNA organ-specific microarray chip approach. We analyzed 287 differentially expressed genes derived from leaf, stem, R1 and R4 flower developmental stages. Transcriptional analysis allowed the detection of three different groups of genes according to their expression patterns. Group 1 contained 112 up-regulated genes under abiotic stress conditions (cold or salinity), whereas Group 2 (126 genes) did not show changes in their expression levels. On the other hand, 49 genes were classified as Group 3 included were down-regulated genes under both stresses. Eighty genes exhibited a significative fold change under abiotic stress conditions, being six of them validated by qRT-PCR. Microrarray profiling of cold and NaCl-treated sunflower leaves revealed dynamic changes in transcript abundance, including transcription factors, defense/stress related proteins, and effectors of homeostasis, all of which highlight the complexity of both stress responses. Project description:Subfamily 2 of SNF1-related protein kinase (SnRK2) plays important roles in plant abiotic stress responses as a global positive regulator of abscisic acid signaling. In the genome of the model tree Populus trichocarpa, 12 SnRK2 genes have been identified, and some are upregulated by abiotic stresses. In this study, we heterologously overexpressed the PtSnRK2 genes in Arabidopsis thaliana and found that overexpression of PtSnRK2.5 and PtSnRK2.7 genes enhanced stress tolerance. In the PtSnRK2.5 and PtSnRK2.7 overexpressors, chlorophyll content and root elongation were maintained under salt stress conditions, leading to higher survival rates under salt stress compared with those in the wild type. Transcriptomic analysis revealed that PtSnRK2.7 overexpression affected stress-related metabolic genes, including lipid metabolism and flavonoid metabolism, even under normal growth conditions. However, the stress response genes reported to be upregulated in Arabidopsis SRK2C/SnRK2.6 and wheat SnRK2.8 overexpressors were not changed by PtSnRK2.7 overexpression. Instead, PtSnRK2.7 overexpression widely and largely influenced the transcriptome in response to salt stress; genes related to transport activity, including anion transport-related genes, were characteristically upregulated, and a variety of metabolic genes were specifically downregulated. We also found that the salt stress response genes were greatly upregulated in the PtSnRK2.7 overexpressor. Taken together, poplar subclass 2 PtSnRK2 genes can modulate salt stress tolerance in Arabidopsis, through the activation of cellular signaling pathways in a different manner from that by herbal subclass 2 SnRK2 genes. Total RNAs of the transgenic plants expressing 35S:PtSnRK2.7 treated with or without 200 mM NaCl. As a control experiment, the wild-type plants were treated similarly. The total RNAs were used for microarray analysis to reveal genes affected by the PtSnRK2.7 overexpression.
