Comparative analysis of piRNA profiles reveals cryoinjury or freezability between boar (Sus scrofa) and giant panda (Ailuropoda melanoleuca) sperm during cryopreservation

Project description:In this study, differentially expressed (DE)piRNAs of fresh and frozen-thawed sperm with different freeze tolerance compacity from giant panda and boar were evaluated. The results showed 1160 (22 down-regulated and 1138 up-regulated) and 384 (110 up-regulated and 274 down-regulated) differentially expressed (DE) piRNAs were identified in giant panda and boar sperm, respectively. Gene ontology (GO) enrichment analysis revealed that the target DE mRNAs of DE piRNAs were mainly enriched in biological regulation, cellular process and metabolic process in giant panda and boar sperm. Moreover, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that the target DE mRNAs of DE piRNAs were only distributed on DNA replication and cAMP signaling pathway in giant panda, but cAMP, cGMP and MAPK signaling pathway in boar sperm which were considered as part of olfactory transduction pathway.Conclusion:Olfactory transduction related pathways maybe contributed to different freeze tolerance compacity between giant panda and boar sperm, which will benefit to further understand the molecular mechanism of sperm cryoinjury and freezability.

Additionally, we speculated that maternal fibre intake might lead to significant changes in cardiac gene expression in the offspring by promoting the production of short-chain fatty acids (SCFAs). To examine the impact of maternal high fibre intake on the cellular composition of the heart, we conducted single-cell RNA sequencing (scRNA-seq) analysis on cardiac tissue from 6-week-old offspring who had not been subjected to any external challenges.

RNA-seq of Senegalese sole testis: comparison between fish fed a control and an algae-supplemented diet

Project description:Senegalese sole males born and raised in captivity (F1 males) do not spawn naturally and typically display lower gamete quality when compared with wild individuals. Broodstock nutrition is an important aspect when dealing with reproduction because it influences not only fish health, but also gamete and progeny quality. The usage of dietary algae antioxidants to improve fish reproduction is under-explored, especially in terms of the male reproductive system. In this experiment, 6 % of a blended meal of Phaeodactylum tricornutum and Gracilaria gracilis was incorporated in Senegalese sole broodstock feed, to evaluate the effects on sperm quality of F1 males throughout the breeding season. RNA-seq was employed to assess differences in the gonadal tissue of fish fed the algae supplemented diets.
