We further found that knocking down expression of the viral tat and/or nef genes was correlated with CD3g transcript and TCR/CD3 surface receptor levels on HIV-1 infected cells. This study was undertaken to investigate the direct effect of HIV-1 Tat expression on the TCR/CD3 machinery. Progressive downregulation from TCR/CD3hi to TCR/CD3lo to TCR/CD3â was observed on Tat expressing cells in a manner that emulated HIV-1 infection, with a lack of CD3g transcripts again responsible for the defect. Thus, the progression TCR/CD3hi to TCR/CD3lo to TCR/CD3â is an active, reversible process with receptor levels fluctuating in response to intracellular dynamics. Examination of tat mutants found that the regions involved in Tat-mediated transactivation and TAR binding are required for TCR/CD3 downregulation while the lysine at position 28 and Tat exon 2 are dispensable. Global gene expression, assessed in association with TCR/CD3 downregulation in HIV-1 infected and Tat expressing cells, detected broad suppression of TCR/CD3 signaling, co-stimulation and negative regulatory genes along with target transcription factors, ligands and receptors. A significant subset of the genes altered in HIV-1 infected cells was specifically targeted by Tat in association with TCR/CD3 loss. Our finding that Tat negatively regulates many facets of the TCR/CD3 machinery has important implications for disease pathogenesis. We used microarrays to investigate changes in CD4+ T cell gene expression induced by HIV-1 infection for comparison with the Tat expressing cells. Project description:The persistence of HIV infection under ART is due to a reservoir of latently infected cells that harbor replication-competent virus and evade immune recognition. Defining the mechanisms responsible for the establishment and maintenance of HIV latency is crucial to achieve HIV eradication or functional cure. Previous studies demonstrated a non-cytotoxic CD8+ T-cells mediated inhibition of virus replication during untreated HIV/SIV infection and inhibition of virus production under ART; however, the mechanisms responsible for this antiviral effect remained poorly understood. In our primary cell-based in vitro latency model we demonstrated that co-culture with CD8+ T-cells promotes changes in metabolic and cell survival pathways in HIV-infected memory CD4+ T-cells that may negatively regulate HIV expression and ultimately promote the establishment of latency. Modulation of this CD8-mediated activity may represent a tool to disrupt HIV latency and reservoir persistence in ART-treated individuals.

Project description:HIV-1 latency is a critical hurdle to a cure for HIV-1 infection, but our understanding of the molecular biology of latency control is still incomplete. We provide experimental evidence that HIV-1 infection of primary T cells and T cell lines generates a substantial amount of TCR/CD3 activation-inert latently infected T cells. Proteomic and genome-wide RNA-level analysis comparing CD3-responsive and CD3-inert latently HIV-1 infected T cells, followed by software-based data integration suggested two phenomena to govern CD3-inertness: (i) the presence of extensive transcriptomic noise that affected the efficacy of CD3 signaling and (ii) defined changes to specific signaling pathways. Validation experiments demonstrated that compounds known to increase transcriptomic noise further diminished the ability of TCR/CD3 stimulation to trigger HIV-1 reactivation. Conversely, targeting specific central nodes in the generated PINs such as STAT3 improved the ability of TCR/CD3 activation to trigger HIV-1 reactivation in T cell lines and primary T cells. The data emphasize that latent HIV-1 infection is largely the result of extensive, stable biomolecular changes to the signaling network of the host T cells harboring latent HIV-1 infection events. We discuss the implications of these findings for the idea of using single drug therapies to trigger HIV-1 reactivation in the latent T cell reservoir in patients.

Project description:The main obstacle in curing an established HIV-1 infection is the long-lived reservoir of latently infected CD4+ T cells. This reservoir is maintained by T cell expansion that can be activated by several mechanisms including antigen-driven proliferation and homeostatic proliferation (HSP). Antigen-driven proliferation triggered by T cell receptor (TCR) signaling is a strong physiological inducer of CD4+ T cell expansion. This also reactivates latent HIV-1 and thus, can be the source of viral rebound. Unlike antigen-driven proliferation, HSP allows the expansion of HIV-1-infected CD4+ T cells without activating HIV-1 expression. While this condition strongly activates STAT5 signaling, the mechanisms for HIV-1 containment are unknown. Our previous work using HIV-infected primarily CD4+ T cells maintained under HSP conditions suggested a post-transcriptional block as a cause of the containment (Tsunetsugu-Yokota et al. 2016 Front Microbiol). To decipher the mechanisms that contribute to the HIV-1 refractory state in homeostatic proliferating CD4+ T cells, we analyzed differentially expressed genes in primary CD4 + cells that were cultured either under HSP conditions (culture with IL-7 and IL-15) or after TCR-stimulation (culture with IL-2 after anti-CD3/CD28 activation).

Project description:Cell activation is a vital step for T cell memory/effector differentiation as well as for productive HIV infection. To identify novel regulators of this process, we used next generation sequencing to profile changes in microRNA expression occurring in purified human naive CD4 T cells in response to TCR stimulation and HIV infection. HIV infection had no significant impact on global miR expression in quiescent nave CD4 T cells. We identified miR-34c-5p as a novel miR strongly induced by TCR stimulation of nave CD4 T cells, and found that it was consistently down-regulated in response to viral infection. Over-expression of miR-34c-5p had a positive effect on HIV-1 replication. Finally, we demonstrated that miR-34c-5p alters the expression of several genes involved in TCR signaling and cell activation, identifying it as a novel regulator of nave CD4 T cell activation potentially targeted by HIV infection.
