Sin1-mTORC2 Augments Lipid Anabolism of Tumor-associated Macrophages to Limit the Type I Interferon-dependent Anti-tumor Immunity

Project description:The biological consequences and mechanisms of metabolic reprogramming of macrophages in the tumor microenvironment (TME) remain largely elusive. Here we showed, in part by myeloid-specific ablation of Sin1, a key component of mammalian target of rapamycin complex (mTORC) 2, that the Sin1-mTORC2-Akt-sterol regulatory element-binding proteins (SREBPs) signaling was augmented in tumor-associated macrophages (TAMs), leading to excessive lipid synthesis and accumulation, which consequently impaired the STING-dependent type I interferon production and CD8+ T cell mediated anti-tumor immunity. The ability of Sin1 deficiency in TAMs to inhibit lipid synthesis and enhance anti-tumor immunity could be blunted by the accumulation of cholesterol/lipids in TAMs. Furthermore, genetic ablation of Sin1 in TAMs or pharmacologic inhibition of lipid anabolism acted synergistically with immune checkpoint blockade (ICB)-based cancer therapy to control tumor growth. Finally, elevated Sin1-mTORC2-Akt activity and lipid anabolism as well as decreased type I IFN responses were all observed in TAMs from human colon cancers and tightly associated with poor patient survival. Together, our study uncovers a novel role of Sin1-regulated lipid anabolic pathway in TAMs which acts as a metabolic checkpoint for anti-tumor immunity via limiting a STING-dependent type I IFN response.

Project description:We herein demonstrate that mammalian target of rapamycin complex 2 (mTORC2), a critical core component of the growth factor signaling system, globally alters histone acetylation through metabolic reprogramming in the highly malignant brain tumor glioblastoma (GBM). Integrated analyses unravel that mTORC2 regulates iron trafficking via histone H3K9 acetylation of the ferritin promoter, facilitating GBM growth and survival. These findings nominate mTORC2 as a critical epigenetic regulator of iron metabolism in cancer.

TMBIM6/BI-1 contributes to cancer progression through assembly with mTORC2 and AKT activation

Project description:Transmembrane B cell lymphoma 2-associated X protein inhibitor motif-containing (TMBIM) 6, a Ca2+ channel-like protein, is highly upregulated in several cancer types. Here, we show that TMBIM6 is closely associated with survival in patients with cervical, breast, lung, and prostate cancer. TMBIM6 deletion or knockdown suppressed primary tumor growth. Further, mTORC2 activation was up-regulated by TMBIM6 and stimulated glycolysis, protein synthesis, and the expression of lipid synthesis genes and glycosylated proteins. Moreover, ER-leaky Ca2+ from TMBIM6, a unique characteristic, was shown to affect mTORC2 assembly and its association with ribosomes. In addition, we identified that BIA compound, a suggestive TMBIM6 antagonist, prevented TMBIM6 binding to mTORC2, decreased mTORC2 activity, and also regulated TMBIM6-leaky Ca2+, further suppressing tumor formation and progression in cancer xenograft models. This previously unknown signaling cascade in which mTORC2 activity is enhanced via the interaction with TMBIM6 provides effective therapeutic targets for various malignancies. We used microarrays to detail the global of gene expression and identified that most of the differentially expressed genes related to apoptotic process, migration, proliferation, and metabolic pathways were decreased in TMBIM6 KO HT1080 cells

mTORC2 regulates amino acid metabolism in cancer by phosphorylation of the cystine-glutamate antiporter xCT

Project description:Mutations in cancer reprogram amino acid metabolism to drive tumor growth, but the molecular mechanisms are not well understood. Using an unbiased proteomic screen, we identified mTORC2 as a critical regulator of amino acid metabolism in cancer via phosphorylation of the cystine-glutamate antiporter xCT. mTORC2 phosphorylates serine 26 at the cytosolic N-terminus of xCT, inhibiting its activity. Genetic inhibition of mTORC2, or pharmacologic mTOR kinase inhibition, promotes glutamate secretion, cystine uptake and incorporation into glutathione linking growth factor receptor signaling with amino acid uptake and utilization. These results identify an unanticipated mechanism of amino acid metabolic reprogramming in cancer, enabling tumor cells to adapt to changing environmental conditions.

Transcriptome changes associated with targeting mTORC2 in lung squamous cell carcinoma

Project description:Lung squamous cell carcinoma (LUSC) represents 30% of NSCLC and is associated with high mortality and a lack of therapies. Targeted therapies for lung adenocarcinoma (LUAD) improve overall survival, but such advances have not been made for treatment of LUSC irrespective of frequent molecular abnormalities. Immune checkpoint inhibitor therapies have achieved some success in LUSC patients. However, most patients do not respond adequately to such treatment, thereby patients are left with fewer specific treatment options. Exploring vulnerabilities in NSCLC, we showed that a high percentage of LUSC patients bear genetic alterations in the PI3K-mTOR-AKT signaling pathway. Loss of function of mTORC2 in LUSC cells inhibits glycolysis and lactate secretion in cultured cells and tumor growth in vivo. The goal of this study was to investigate how targeting mTORC2 impacts LUSC and the tumor microenvironment, which can be harness for therapeutic options.

Opposing tumor promoting and suppressive functions of Rictor/mTORC2 signaling in adult glioma and pediatric SHH medulloblastoma

Project description:Most human cancers arise from stem/progenitor cells by sequential accumulation of genetic/epigenetic alterations, while cancer modeling typically requires simultaneous multiple oncogenic events. Here we show that a single p53 mutation, despite causing no defect in mouse brain, promoted neural stem/progenitor cells to spontaneously accumulate oncogenic alterations, including loss of multiple chromosomal (chr) regions syntenic to human chr10 containing Pten, forming malignant gliomas/glioblastomas with PI3K/Akt activation. Rictor/mTORC2 loss inhibited Akt signaling, greatly delaying and reducing glioma formation by suppressing glioma precursors within the subventricular zone stem-cell niche. Unexpectedly, Rictor/mTORC2 loss delayed timely differentiation of granule cell precursors (GCPs) during cerebellar development, promoting sustained GCP proliferation and medulloblastoma formation, which recapitulated critical features of TP53-mutant Sonic Hedgehog (SHH) medulloblastomas with GLI2 and/or N-MYC amplification. Our study demonstrates that Rictor/mTORC2 has opposing functions in neural stem cells and GCPs in the adult and developing brain, promoting malignant gliomas/glioblastoma and suppressing SHH-medulloblastoma formation, respectively.
