Project description:In field conditions, tea plants are often exposed to drought stress, which has profound effects on the growth and development of tea plants. However, most studies on tea plants in response to drought stress focused on single gene or protein expression, and transcriptome or proteome profiles, the impact of drought stress on ubiquitination in proteins remains unearthed. We performed a global profile of ubiquitinated (Kub) proteins in tea leaves under drought stress. In total, 1,409 lysine Kub sites in 781 proteins were identified, of which 14 sites in 12 proteins were up-regulated and 123 sites in 91 proteins were down-regulated compared with drought and control. Furthermore, we analyzed the Kub proteins related to ubiquitin-mediated proteolysis, catechins biosynthesis, and carbohydrate and amino acid metabolism in tea leaves under drought stress. The results indicated that many Kub proteins involved in ubiquitin-mediated proteolysis played important roles in protein degradation. Several Kub proteins related to catechins biosynthesis were positively correlated with each other because of their co-expression and co-localization. Our study preliminarily revealed the global profiling of Kub proteins in metabolic pathways and provided an important resource for further study on the functions of Kub proteins in tea plants under drought stress.

Project description:Drought is considered as a threat to the sustainability of Mediterranean forests and, of the agro-silvo-pastoral “dehesa” system. Holm oak (Quercus ilex L.) is one of the most representative species of these ecosystems and this abiotic stress causes a high tree mortality rate, being more accused at the seedling stage. In this study, an integrative analysis of transcriptome and proteome in leaves of Q. ilex seedlings under drought conditions was carried out to identify gene products involved in the metabolic pathways related to drought tolerant mechanisms.

Project description:affy_popsec_orleans_poplar - xyleme - This project aims to identify candidate genes for water deficit acclimation and/or adaptation in a tree species: poplar. Due to compelling evidence that transcriptional regulation plays a major role in regulating many biological processes, we will look for genes and gene expression networks related to drought stress. We intend to analyse the transcriptome in two poplar genotypes of contrasted tolerance to water deficit, at various stages and intensities of stress and simultaneously in whole xylem and cambial zone from young trees. The co-analysis of two genotypes of contrasted tolerance to water deficit should allow to better discriminating genes presenting a potential adaptative character from genes responding passively to the constraint. The consideration of both whole xylem and cambial zone will enable us to discriminate between regulations events originating from very young xylem cells undergoing their first steps of xylem differentiation (cambial zone) compared to whole xylem sample enriched in more differentiated xylem cells and parenchyma ray cells.-Two poplar clones, named Soligo (S) and Carpaccio (C) were submitted to 4 treatments: non-stressed control, severe-drought stress, mild-drought stress and early-drought stress. Two pools of 2 or 3 trees were considered as biological replicates. Two different samples were collected on each individual tree: whole stem xylem (X) and cambial zone plus very young expanding xylem (Y). Total RNAs were extracted from each tree and assemble in pool of 3 or 2 individuals using equimolar ratio for each X and for Y. One affymetrix slide corresponds to one pooled RNA sample (X or Y). A total number of slides : 4 treatments x 2 clones x 2 tissues x 2 biological replicates (pool) = 32 slides, will be done. Examination at the molecular level how this tree species responds to drought and salt stresses by regulating the expression of genes involved in signal transduction, transcriptional regulation, and stress responses.

We look for genes and gene expression networks related to drought stress. We intend to analyse the transcriptome in laser-microdissected stomata, in two genotypes, Carpaccio and Soligo, at various stages and intensities of stress. In response to water deficit, water loss is adjusted through stomatal conductance. Transcriptome of pure stomatal material should bring valuable information hardly reachable in whole leaf transcriptome analysis. The comparison between medium and severe stress intensities and between early and long term stresses will power the selection of genes of interest. The co-analysis of two genotypes of contrasted tolerance to water deficit should help to discriminate genes presenting a potential adaptative character from genes responding passively to the constraint.-Two poplar clones, Soligo (S) and Carpaccio (C) were submitted to 4 treatments: control, mild water deficit, moderate water deficit (12-day long for both) and early-drought stress (about 36-h long). Growth and physiology was characterised on a batch of plants and samples collected on another batch of plants. Five hundred stomata (one thousand five hundreds guard cells) were laser microdissected in mature leaves of each tree. Stomata from 3 trees (1500) were pooled for total RNAs extraction. A pool is considered as one biological replicate and corresponds to one Affymetrix slide. The two biological replicates originate from the same experiment. Project description:Tea (Camellia sinensis (L.) O. Kuntze) is an important non-alcoholic commercial beverage crop. Tea tree is a perennial plant, and winter dormancy is its part of biological adaptation to environmental changes. We recently discovered a novel tea tree cultivar that can generate tender shoots in winter, but the regulatory mechanism of this ever-growing tender shoot development in winter is not clear. In this study, we conducted a proteomic analysis for identification of key genes and proteins differentially expressed between the winter and spring tender shoots, to explore the putative regulatory mechanisms and physiological basis of its ever-growing character during winter.
