Project description:Nanoscale zero valent iron (nZVI) is used to remediate aquifers polluted by organochlorines or heavy metals and was also suggested to eliminate harmful algal blooms. nZVI can therefore affect microorganisms in the vicinity of the application area, including microalgae. However, studies on early transcriptomic effects of microalgae after exposure to nZVI are rare. Here, we described the early physiological and transcriptomic response of the freshwater ecological indicator green microalga, Raphidocelis subcapitata ATCC 22662, to 100 mg/L of reactive nZVI and non-reactive nano-magnetite (nFe3O4). The combined effect of shading and the release of total iron from nZVI posed a short-term inhibition effect leading to 15 % of deformed cells and cytosol leakage, while cells viability increased after 24 h. nZVI triggered a more pronounced transcriptomic response with (7380 differentially expressed genes [DEGs]) compared to nFe3O4 (4601 DEGs) after 1 h. nZVI, but not nFe3O4 increased the expression of genes function in DNA repair and replication, while deactivated carbohydrate-energy metabolisms, mitochondria signaling, and transmembrane ion transport. This study highlights an early fate assessment of algal cells under nZVI and nFe3O4 exposure using next-generation risk assessment methods and will serve as valuable information for safe and sustainable application of nZVI in water remediation.

Project description:This study assess the biocompatibility of Ti-26Nb compared to Ti-Cp, a reference in implantology, through the combination of conventional toxicological assay, morphological observations and transcriptomic analysis performed on two different cells line : Saos-2 and THP-1. The biocompatibility of Ti-Cp implants was evaluated in comparison with Ti-26Nb implants both having a mirror polished surface, studying cell proliferation (trypan blue and WST-1) and cell morphology/adhesion (SEM) on human THP-1 and Saos-2 cell lines. Finally, an evaluation of a potential toxicity of potassium niobiate powder KNbO3 was carried out by measuring metabolic activity and realizing a transcriptomic study on the THP-1 line (the first one) to deepen the results observed with Ti-26Nb discs. Indeed, niobiate ions are potentially released from implant in cellular acidic environnement. Concerning Saos-2 cells, our results suggest that Ti-26Nb and Ti-Cp discs do not impact proliferation and viability. Concerning THP-1 cell line, a decrease in proliferation and viability is observed in contact of Ti-26Nb discs (hypothesis of a cell-line effect ?, no biocompatibility study on monocyte/macrophage cell lines such as THP-1 actually available in literature). KNbO3 powder does not impact viability of THP-1 cells and does not induce changes at molecular level. Taken together, these data suggest a possible toxicity of Ti-26Nb toward THP-1 cells not directly related to the niobium but perhaps to the manufacturing process of Titane niobium alloy.

Project description:Addition of nanoscale zero valent iron (NZVI) to anaerobic batch reactors to enhance methanogenic activity is described. Two NZVI systems were tested: a commercially available NZVI (cNZVI) slurry and a freshly synthesized NZVI (sNZVI) suspension that was prepared immediately before addition to the reactors. In both systems, the addition of NZVI increased pH and decreased oxidation/reduction potential compared with unamended control reactors. Biodegradation of a model brewery wastewater was enhanced as indicated by an increase in chemical oxygen demand removal with both sNZVI and cNZVI amendments at all concentrations tested (1.25-5.0 g Fe/L). Methane production increased for all NZVI-amended bioreactors, with a maximum increase of 28% achieved on the addition of 2.5 and 5.0 g/L cNZVI. Addition of bulk zero-valent iron resulted in only a 5% increase in methane, indicating the advantage of using the nanoscale particles. NZVI amendments further improved produced biogas by decreasing the amount of CO2 released from the bioreactor by approximately 58%. Overall, addition of cNZVI proved more beneficial than the sNZVI at equal iron concentrations, due to decreased colloidal stability and larger effective particle size of sNZVI. Although some have reported cytotoxicity of NZVI to anaerobic microorganisms, work presented here suggests that NZVI of a certain particle size and reactivity can serve as an amendment to anaerobic digesters to enhance degradation and increase the value of the produced biogas, yielding a more energy-efficient anaerobic method for wastewater treatment.

Project description:Hand-to-mouth activity is considered to be the main way for children to come into contact with contaminated soil, and bioavailability is an important factor affecting their health risk. To reduce soil As risk to humans by oral exposure, nanoscale zero-valent iron (nZVI) has been extensively studied for immobilizing As-contaminated soil, but its efficiency has not been investigated using in vitro assay and its influence on As-RBA. In this study, two contaminated soil samples (A and B) were amended with 1% and 2% (w/w) nZVI for 56 days to study its effect on As fraction by sequence extraction, As bioaccessibility by SBRC assay, and As relative bioavailability (RBA) by the mouse liver and kidney model. Based on the sequence extraction, the As associated with the E1 (exchangeable fraction) and C2 (carbonate fraction) fractions were decreased from 3.00% to 1.68% for soil A and from 21.6% to 7.86% for soil B after being treated with 2% nZVI for 56 days. When assessing As bioaccessibility in all soils treated with nZVI by SBRC assay, it was found that As bioaccessibility was significantly higher in the gastric phase (GP) and lower in the intestinal phase (IP) (p < 0.05), and the bioaccessible Fe concentration decreased significantly from the gastric to intestinal phase at the same time. Based on the mouse liver-kidney model, the As-RBA in soil A increased from 21.6% to 22.3% and 39.9%, but in soil B decreased from 73.0% to 55.3% and 68.9%, respectively. In addition, there was a significant difference between As bioaccessibility based on GP or IP of SBRC assay and As-RBA in two soils after being treated with nZVI for 56 days. To more accurately assess the effects of nZVI human arsenic exposure, As-RBA should be considered in concert with secondary evidence provided through fraction and bioaccessibility assessments. In addition, it is necessary to develop a suitable in vitro assay to predict As-RBA in nZVI-amended soils.

Project description:The overuse of chromium (Cr) has significantly negatively impacted human life and environmental sustainability. Recently, the employment of nano zero-valent iron (nZVI) for Cr(VI) removal is becoming an emerging approach. In this study, carbonized melamine foam-supported nZVI composites, prepared by a simple impregnation-carbonization-reduction method, were assessed for efficient Cr(VI) removal. The prepared composites were characterized by XPS, SEM, TEM, BET and XRD. Batch experiments at different conditions revealed that the amount of iron added, the temperature of carbonization and the initial Cr(VI) concentration were critical factors. Fe@MF-12.5-800 exhibited the highest removal efficiency of 99% Cr(VI) (10 mg/L) at neutral pH among the carbonized melamine foam-supported nZVI composites. Its iron particles were effectively soldered onto the carbonaceous surfaces within the pore networks. Moreover, Fe@MF-12.5-800 demonstrated remarkable stability (60%, 7 days) in an open environment compared with nZVI particles.

Project description:IntroductionMyocardial injury is a serious complication in sepsis with high mortality. Zero-valent iron nanoparticles (nanoFe) displayed novel roles in cecal ligation and puncture (CLP)-induced septic mouse model. Nonetheless, its high reactivity makes it difficult for long-term storage.ObjectivesTo overcome the obstacle and improve therapeutic efficiency, a surface passivation of nanoFe was designed using sodium sulfide.MethodsWe prepared iron sulfide nanoclusters and constructed CLP mouse models. Then the effect of sulfide-modified nanoscale zero-valent iron (S-nanoFe) on the survival rate, blood routine parameters, blood biochemical parameters, cardiac function, and pathological indicators of myocardium was observed. RNA-seq was used to further explore the comprehensive protective mechanisms of S-nanoFe. Finally, the stability of S-nanoFe-1d and S-nanoFe-30 d, together with the therapeutic efficacy of sepsis between S-nanoFe and nanoFe was compared.ResultsThe results revealed that S-nanoFe significantly inhibited the growth of bacteria and exerted a protective role against septic myocardial injury. S-nanoFe treatment activated AMPK signaling and ameliorated several CLP-induced pathological processes including myocardial inflammation, oxidative stress, mitochondrial dysfunction. RNA-seq analysis further clarified the comprehensive myocardial protective mechanisms of S-nanoFe against septic injury. Importantly, S-nanoFe had a good stability and a comparable protective efficacy to nanoFe.ConclusionsThe surface vulcanization strategy for nanoFe has a significant protective role against sepsis and septic myocardial injury. This study provides an alternative strategy for overcoming sepsis and septic myocardial injury and opens up possibilities for the development of nanoparticle in infectious diseases.
