Project description:Our goal was to identify UTP-regulated genes in WT cADSC and P2Y₂ KO cADSC in the frame of the potential therapeutic transplantation of these cells into the mouse ischemic heart after a myocardial infarction. We identified UTP-regulated genes involved in inflammation and the immune response, UTP target genes linked to extracellular matrix organization such as matrix metallopeptidases and various collagen types, UTP-regulated genes related to macrophage chemotaxis and differentiation into pro-fibrotic foam cells and UTP target genes linked to angiogenesis regulation and the response to hypoxia.

Project description:Hematopoietic stem cell (HSC) untreated and treated with UTP 10uM at 1, 6 and 24 h Experiment Overall Design: Highly purified CD34+ cells from 6 healthy donors were seeded at 1000000 cells/ml in serum free medium (EX vivo 15) w/o cytokines and treated with 10 uM UTP for 1, 6 and 24 hours. As a control, CD34+ untreated cells were maintened in the same culture conditions for the same times. RNAs originating from the 6 donors were pooled in order to obtain at least 2 mg per sample (untreated and UTP-treated respectively).

Project description:Purpose: Detect transcriptome-wide mapping of m6A modifications of mouse macrophage RAW264.7 cells by MeRIP-seq after 24 h-treatment with 100 μM 2,4,6-trichlorophenol (TCP), 100 μM 2,4,6-tribromophenol (TBP) and 100 μM 2,4,6-triiodophenol (TIP). Methods: Total RNA was extracted using TRIzol Reagent. Poly (A) RNA was purified from 50 μg total RNA using Dynabeads. Then the poly(A) RNA was fragmented into small pieces using Magnesium RNA Fragmentation Module.The fragmented RNA was incubated with m6A-specific antibody. The desired m6A-enriched RNA eluate was used to construct cDNA library in parallel with input control. Conclusion: Our study revealed the difference of gene expression at transcriptional level and m6A-methylated RNA among TCP, TBP and TIP treated RAW264.7 cells by RNA-seq and MeRIP-seq technologies.
