Project description:Mouse, also known as a good model animals, is widely uaed in the research of hair follicle development. Unraveling the dynamic accessibility of regulatory chromatin at single-cell resolution is key to understanding stem/progenitor cell fate choices. and it's therefore of significance to investigate the chromatin accessibility and transcriptional regulation mechanism during mouse hair follicle development. To provides comprehensively understanding on the transcriptional regulation mechanism and cell lineage cell fate decisions, we performed single-cell ATAC sequencing on 23,716 single cells from induction (embryonic day 13.5), organogenesis (embryonic day 16.5) and cytodifferentiation (postnatal day 0) stage fetus mouse dorsal skin. Based on unsupervised clustering analysis, cell lineage inference and etc, the single cell map of chromatin open area was drawn from each cell type and the mechanism of cell transcription regulation was explored. Collectively, our data here provide a reference for deeply exploring the epigenetic regulation mechanism of mouse hair follicles development.

A single-cell transcriptome atlas during Cashmere goat hair follicle morphogenesis

Project description:Cashmere, also known as soft gold, is produced from secondary hair follicles in Cashmere goats and it’s therefore of significance to investigate the molecular profiles during Cashmere goat hair follicle development. However, our current understanding of the machinery underlying Cashmere goat hair follicle remains largely unexplored and researches regarding hair follicle development mainly used the mouse as a research model. To provides comprehensively understanding on the cellular heterogeneity and cell lineage cell fate decisions, we performed single-cell RNA sequencing on 19,705 single cells from induction (embryonic day 60), organogenesis (embryonic day 90) and cytodifferentiation (embryonic day 120) stage fetus Cashmere goat dorsal skin. Unsupervised clustering analysis identified 16 cell clusters and their corresponding cell types were also successfully characterized. Based on cell lineage inference, we revealed detailed transcriptional gene expression profiles during dermal and epidermal lineage cell fate decisions. These works together delineate unparalleled molecular profiles of different cell populations during Cashmere goat hair follicle morphogenesis and provide a valuable resource for identifying biomarkers during Cashmere goat hair follicle development.

Project description:Hair follicles of the yak are in anagen and catagen , the hair follicles of healthy female yaks around 2 years old are collected for the preparation of single-cell suspension, the cells were sequenced by scRNA-seq on the 10x genome platform. A total of about 12000 single-cell transcriptome information were obtained. According to the reported marker genes, the main cell groups in anagen and catagen of yak were identified. Based on the analysis of pseudotime trajectory, the differentiation trajectory of epidermal cell lineage and dermal cell lineage during hair follicle development and the dynamic changes of genes during differentiation were described.

SKPs derive from hair follicle precursors and exhibit properties of adult dermal stem cells

Project description:Global expression analysis of neural crest-like skin-derived precursors (SKPs) and Sox2-positive follicle dermal cells that SKPs originate from. In spite of the remarkable regenerative capacity of mammalian skin, an adult dermal stem cell has not yet been identified. Here, we provide evidence that SKPs, multipotent neural crest-like skin-derived precursors, represent an adult dermal stem cell. When transplanted into adult skin, SKPs can reconstitute the adult dermis, contribute to dermal wound-healing, home to a hair follicle niche, and instruct epidermal cells to make hair follicles. Hair follicle-derived SKPs self-renew, maintain their multipotency, and serially reconstitute hair follicles. The endogenous origin of SKPs are Sox2-positive follicle dermal cells that share a similar global gene expression profile with SKPs. These endogenous cells home back to their follicle niche, induce hair follicle morphogenesis, and differentiate into neural and dermal progeny. Hair follicle-associated dermal cells will move out of their niche to contribute to dermal maintenance and wound-healing. These studies therefore identify a dermal stem cell, and provide a biological rationale for the presence of a multipotent precursor within adult dermis, findings with important therapeutic implications.
