Project description:Plant response to insect feeding appears to be highly specific with regard to the organisms in the system. Here, we report on the interaction between grapevine Vitis vinifera plants and a phloem-feeding insect pest, the vine mealybug Planococcus ficus. Plants were exposed to P. ficus for periods of 6 hours and 96 hours, after which they were analysed for gene expression levels using microarrays and quantitative real-time PCR (qPCR). Both methods showed that grapevine displayed only a minimal response to mealybug feeding at the transcript level at both time periods. Intermediate grapevine exposure times (24, 48 and 72 hours) to P. ficus feeding were investigated using qPCR analysis of ten additional genes associated with known plant defense responses. Results showed that only a single gene, pathogenesis-related protein 1, was differentially expressed after 48 hours of mealybug feeding. During the course of mealybug feeding, however, a number of other genes were significantly up- or down-regulated at certain time points. Thus, it appears as if grapevine responds minimally to feeding by P. ficus as well as within a very narrow time period. The relative lack of grapevine plant defense mechanisms may be a result of the feeding strategies of mealybugs. Eight samples were analysed. Two replicates each were included for each treatment (6 hour and 96 hour feeding), resulting in four samples. Two control replicates were included for each treatment (6 hour and 96 hour feeding controls), resulting in a further four samples.

Project description:Phenotypic responses to biotic stresses are often studied as the interactions between two species; however, in the phytobiome, these responses frequently result from complex interactions involving several organisms. Here, we show that variation in chlorosis caused by Russian wheat aphid (Diuraphis noxia) feeding is determined, in part, by aphid-associated bacteria. Proteomic analysis of fluids injected into a sterile medium by the aphid during feeding indicate that 99% of the proteins are of bacterial origin. Of these, the greatest proportion are produced by bacteria in the order Enterobacteriales. Bacteria from five genera in four families that have the capacity to produce these proteins were isolated directly from aphids as well as from wheat leaves only after D. noxia feeding. By themselves or in combination, these bacteria were not virulent to wheat, even at high inoculum levels. Metagenomic analysis showed that the same five D. noxia-associated genera dominated the non-Buchnera component of the aphid microbiome, and that representation of these genera was reduced in aphids from colonies established after isolation of newborn nymphs from their mothers prior to feeding (‘isolated’ aphids). Isolation or treatment with antibiotics reduced bacterial numbers, and these aphids caused less feeding damage on wheat than non-isolated or non-antibiotic treated aphids. Our data show that bacterial proteins are a significant component of Russian wheat aphid saliva, that the bacteria producing these proteins are associated with aphids and plants fed upon by aphids, and that these aphid-associated bacteria facilitate aphid virulence to wheat.

Project description:Plants have developed a complicated resistance system, and they exhibit various defense patterns in response to different attackers. However, the determine factors of plant defense patterns are still not clear. Here, we hypothesized that damage patterns of plant attackers play an important role in determining the plant defense patterns. To test this hypothesis, we selected leafminer, which has a special feeding pattern more similar to pathogen damage than chewing insects, as our model insect, and Arabidopsis thaliana as the response plants. The local and systemic responses of Arabidopsis thaliana to leafminer feeding were investigated using the Affymetrix ATH1 genome array.

Project description:The economic importance of wheat and its contribution to human and livestock diets has been already demonstrated. However, wheat production is impacted by pests that induce yield reductions. Among these pests, wheat curl mite (WCM, Aceria tosichella Keifer) impacts wheat all around the world. WCM are tiny pests that feed within the whorl of developing leaves and prevent the leaves from unfurling by causing leaves curling. The curling of the leaves provides a protective niche for the WCM. Additionally, WCM are also the vector of serious viruses in wheat. Little is known regarding the impact of the WCM on wheat transcriptome, and to date, only one article has been published describing the wheat transcriptomic changes after 1 day of WCM feeding. To better understand the wheat transcriptome variation after long-term feeding by WCM (10 days post infestation (dpi)), we used an RNA-seq approach. We collected leaves uninfested and infested with WCR from two wheat cultivars: Byrd (WCM resistant) and Settler CL (WCM susceptible) at 10 dpi. Our transcriptomic analysis revealed the common and specific transcriptomic variations in WCM resistant and susceptible wheat cultivars, chromosome specific location of the differentially expressed genes, and also identified the gene functions and pathways involved in WCM resistance. Collectively, our study provides important insights on wheat defense mechanisms against WCM after long-term feeding.

Project description:While pathogen-induced immunity is comparatively well characterized, far less is known about plant defense responses to arthropod herbivores. To date, most molecular-genetic studies of plant-arthropod interactions have focused on insects. However, plant-feeding (phytophagous) mites are also pests of diverse plants, and mites induce different patterns of damage to plant tissues than do well-studied insects (e.g., Lepidopteran larvae or aphids). The two-spotted spider mite, Tetranychus urticae, is among the most significant mite pests in agriculture. T. urticae is an extreme generalist that has been documented on a staggering number of plant hosts (more than 1,100), and is renowned for the rapid evolution of pesticide resistance. To understand reciprocal interactions between T. urticae and a plant host at the molecular level, we examined mite herbivory using Arabidopsis thaliana. Despite differences in feeding guilds, we found that transcriptional responses of A. thaliana to mite herbivory generally resembled those observed for insect herbivores. In particular, defense to mites was mediated by jasmonic acid (JA) biosynthesis and signaling. Further, indole glucosinolates dramatically increased mite mortality and development times. Variation in both basal and activated levels of these defense pathways might also explain differences in mite damage and feeding success between A. thaliana accessions. On the herbivore side, a diverse set of genes associated with detoxification of xenobiotics was induced upon exposure to increasing levels of in planta indole glucosinolates. Our findings provide molecular insights into the nature of, and response to, herbivory for a representative of a major class of arthropod herbivores. We used microarray to assess global gene expresion in Arabidopsis thaliana upon Tetranychus urticae attack in two A. thaliana accessions: Bla-2, resistant to spider mite herbivory and Kon, susceptible to spider mite herbivory. 3 week old Arabidopsis thaliana plants were subjected to Tetranychus urticae attack through application of 10 mites for various periods of time (timecourse scenario) or hundreds of mites for 1 hour (feeding site scenario).

Project description:The greenbug aphid, Schizaphis graminum (Rondani) is an important cereal pest periodically threatening wheat yields in the United States and around the world. The wheat breeding program at Texas A&M University has introgressed the greenbug resistance gene Gb3 from synthetic wheat ‘Largo’ into the widely grown cultivars TAM 110 and TAM 112; however, the molecular mechanisms of Gb3-induced defense responses remain unknown. Using Affymetrix GeneChip Wheat Genome Arrays, we investigated the temporal dynamics of Gb3-mediated transcriptional responses upon greenbug feeding on resistant and susceptible bulks (RB and SB respectively) derived from two near-isogenic lines. Following statistical analysis, the MapMan functional classification on 692 differentially expressed transcripts from the interaction group identified 122 transcripts that were putatively associated with biotic stress responses. In RB, Gb3 induced transmembrane receptor kinases and downstream early defense signal transduction pathways. However, non-Gb3 mediated pathways in SB prompted induction of transcripts mediating callose decomposition and transcripts regulating biosynthesis of stress hormones jasmonic acid, ethylene, and abscisic acid. Additionally, the SB showed elevated levels of transcripts mediating redox homeostasis, peroxidases, glutathione S-transferases, and defense-related secondary metabolites. Overall, the results indicated that greenbugs inflicted more damage in the SB compared to RB and absence of Gb3-mediated signaling resulted in cell wall remodeling and strong accumulation of toxic compounds warranting production of antioxidant and defense-related compounds. The study reinforced our knowledge about preferential feeding on S genotypes and plausible antixenosis or tolerance-related defense signaling in Gb3-associated R genotypes and generated several new hypotheses.

Project description:Brown planthopper (BPH) is one of the most destructive pests in rice production. The pyramiding application of BPH-resistance genes BPH14 and BPH15 can effectively improve rice resistance to BPH, however, the molecular mechanisms underlying BPH14/BPH15 pyramiding lines are poorly understood. Here, a mRNA expression profiling analysis was performed on the near isogenic lines (NILs) containing the BPH14, BPH15 or BPH14/BPH15 and their recurrent parent (RP) Wushansimiao. A total of 14492 differentially expressed mRNAs (DEGs) were identified from 12 mRNA profiles of resistant NILs and RP at different feeding stages. In the transcriptome analysis, 531 DEGs appeared to be common among the resistant NILs compared to RP before and after BPH feeding, which were enriched in defense response, phosphorylation and salt stress response. In addition, 258 DEGs shared only in resistant NILs were obtained among the different feeding stages, which were enriched in oxidative stress response, karrikin response and chloroplast organization. 21 DEGs were further selected as candidates for BPH resistance. OsPOX8.1, a potential candidate DEG related to BPH resistance, increased reactive oxygen species levels in rice protoplast. Our results provide valuable information to further explore the defense mechanism of insect-resistant gene pyramiding lines and develop robust strategies for insect control.
