Identification of miR-151a as a universal endogenous control for exosome cargo normalization in human cancer

Project description:The exosomal particles in the blood of cancer patients possess many times more tumor markers than free circulating. Among these elements, miRNAs have great biomedical relevance due to their stability and feasible detection. However, there is not available in the market any reliable endogenous control for the exosomal compartment, nor specific for the miRNA content, preventing the obtainment of standardization measures in cancer liquid-biopsy. In this study, we firstly identified three miRNAs out of a panel of nine potential normalizers that arised from an integrative analysis comparing the global miRNA exosomal profile by miRNA-seq of six lung and ovarian human cancer cell lines under different chemotherapy conditions. Their value as normalizers were also tested in 14 additional human cancer cell lines from different tumor types and after radiotherapy and an alternative chemotherapy treatment. Its translational validation comprised 124 prospective samples, 70 plasma from NSCLC patients, 12 from glioblastoma patients, 10 healthy donors and 16 paired samples from plasma and ascites fluid from ovarian cancer patients. The variability and normalizing properties were tested in comparison with the tissue-origin gold standard miR-16. Our results indicate that miR-151a is constantly represented in the exosomal content with minimal variability compared with miR16 independently of many conditions tested, providing for the first time a universal normalizer for the exosomal compartment that will impact the use of liquid biopsy.

Exosome RNA cargo in activated liver stellate cells

Project description:Previously we have described the role of liver stellate cell exosomes in triggering macrophage migration and immune activation, we wanted to study what cargo in this exosomes were responsible for such activation. We found a few interesting mRNAs and siRNAs involved in the immune response pathway and cytokine production. We have found that Ectodysplasin A, a hepatokine, has an important role in stimulating the macrophage response and to be highly expressed in liver cirrhosis.

Pericardial fluid exosome miRNA profiling

Project description:Pericardial sac surrounding the heart contains pericardial fluid (PF), which is rich in exosomes. PF exosomes increase angiogenesis in hypoxic endothelial cells and in animal model of hindlimb ischemia by passing the proangiogenic miRNAs to recipient cells. However, under pathological conditions such as diabetes, exosome cargo composition changes and harmful miRNAs can be transferred to the recipient cells and induce more deleterious effects in target tissues. In order to check cargo composition of different PF exosomes, we used PF exosomes from non-diabetic aortic valve replacement (AVR), mitral valve replacement (MVR), coronary artery bypass grafting (CABG) patients and CABG patients with diabetes.

ECM deposition is driven by caveolin1-dependent regulation of exosomal biogenesis and cargo sorting

Project description:The composition and physical properties of the extracellular matrix (ECM) critically influence tumor progression, but the molecular mechanisms underlying ECM layering are poorly understood. Tumor-stroma interaction is critically dependent on cell-cell communication mediated by exosomes, small vesicles generated within multivesicular bodies (MVBs). Here, we show that caveolin-1 (Cav1) is a central modulator of both exosome biogenesis and exosomal protein cargo sorting through the regulation of cholesterol content at the endosomal compartment/MVBs. Quantitative proteomics profiling revealed that Cav1 is required for exosomal sorting of ECM protein cargo subsets including tenascin-C (TnC), and for fibroblast-derived exosomes to efficiently depose ECM and promote tumor cell invasiveness. Cav1-driven exosomal ECM deposition not only promotes local stromal remodeling, but also the generation of distant ECM-enriched stromal niches. These results support a model by which Cav1 is a central regulatory hub for tumor-stroma interactions through a novel exosome-dependent ECM deposition mechanism. Neuron-derived exosome from plasma show altered miRNA cargo as a function of antidepressant drug response.

Project description:We investigated NDEs from depressed and psychiatrically healthy controls, before and after, eight weeks of antidepressant treatment (ADT). Neuronal-derived exosomes contained miRNA that showed enrichment for brain functions. Changes in NDE cargo, specifically miR-21-5p, miR-30d-5p, and miR-486-5p expression, were significantly associated with ADT response. Furthermore, we found these targets to be altered in brain tissue from depressed individuals. Together, our study indicates that changes in peripherally isolated NDE can act as a biomarker of ADT response specifically through size and cargo.
