Project description:We present data of the grassland restoration experiment performed in the Bílé Karpaty Mts. (White Carpathians, Czech Republic) in dry species-rich meadows. First we harvested seed material in a preserved source meadow (donor site hereafter) by brush harvesting the vegetation once (B1 hereafter), brush harvesting three times during the season (B3 hereafter), and by cutting green hay (GH hereafter). Then we determined the species composition and seed quantity of the harvested material. Furthermore, we transferred the seeds to an experimental site on ex-arable land (receptor site hereafter), and monitored the development of the meadow communities in the following five years. Project description:Sowing is widely used for the restoration of species-rich grasslands but still there are knowledge gaps regarding the most suitable application of different seed mixtures. We tested the effect of seed mixtures application timing on the establishment of sown forbs and weed control. 36 experimental plots with nine sowing treatments were established in an abandoned cropland in Hungary. Grass-seeds, diverse forb seed mixture and the combination of the two were applied: diverse forb mixture was sown simultaneously or 1, 2 or 3 years after grass sowing, in plots sown previously with grass or in empty plots (fallows). All sowing treatments supported the rapid establishment of the sown species in large cover and hampered weed encroachment. Forbs performed better when sown into fallows than in grass-matrix and forbs establishment was worse in older fallows than in younger ones. Grasses expressed a strong priority effect, especially when forbs were sown at least two years later than grasses. We also investigated the relation between seed germinability, weather parameters and establishment success. Germination rate in the greenhouse could not predict the establishment success of forbs in the field and showed great differences between years, hence we recommend sowing target forbs in multiple years.

Project description:The colonization of land by plants relies on numerous evolutionary innovations crucial for terrestrial adaptation. These include advances in sexual reproduction and the ability to properly respond to various environmental stresses, which involve precise control of their regulatory genes. A notable genetic innovation in land plants is the emergence of histone lysine methyltransferases ATXR5/6, which specifically catalyse the repressive histone H3 lysine 27 monomethylation (H3K27me1). However, the evolutionary reason for land plants to develop these enzymes remains unclear. In this study, we investigate the importance of ATXR5/6 by generating strong atxr5;axtr6 double mutants in Arabidopsis. Our results show that ATXR5/6 are essential for plant reproductive development and play a critical role in supporting normal plant growth by repressing the transcription of stress responsive genes. In addition, ATXR5/6 are necessary for maintaining H3K27 trimethylation (H3K27me3), likely by providing H3K27me1 as a substrate for further methylation. We also demonstrate that the function of ATXR5/6 in regulating reproductive development and responsive genes is conserved in the monocot rice. We propose that land plants may have evolved ATXR5/6 to assist in the transcriptional regulation necessary for terrestrial adaptation

Project description:This experiment probed for the presence of known Arabidopsis and rice microRNAs in total RNA samples derived from species representative of the major groups of land plants: Eudicots (Arabidopsis thaliana, Nicotiana benthamiana), monocots (Oryza satica, Triticum aestivum), magnoliids (Liriodendron tulipifera), gymnosperms (Pinus resinosa), ferns (Ceratopteris thalictroides), lycopods (Selaginella uncinata), and mosses (Polytrichum juniperinum). In most cases two technical or biological replicates were performed.

Project description:A common-garden experiment was carried out to compare two genetically distinct strains of Atlantic salmon (Salmo salar) fed diets formulated with either high (CHO) or low (NoCHO) carbohydrate (starch). Twenty salmon from either a commercial farmed strain or a land-locked population were placed in two tanks (10 fish of each population in each tank) and fed either CHO or NoCHO feeds for 32 days. At the end of the experimental period fish were fasted for 8 h, euthanized and samples of blood and liver collected. Both diet and population had an effect on circulating glucose levels with land-locked salmon showing hypoglycaemia and dietary starch increasing this parameter. In contrast, land-locked salmon showed increased plasma triacylglycerol levels regardless of dietary treatment. This enhanced ability to metabolise dietary starch in land-locked compared to farmed salmon stock was also reflected at a molecular (gene) level as most of the metabolic pathways evaluated in the present study were mainly affected by the factor population rather than by diet. In particular, lower expression of genes for mitochondrial metabolism in land-locked salmon reflects drastic differences in energy metabolism between the populations. The liver transcriptome analysis highlighted some new gene candidates such as elovl6 to evaluate in future studies assessing the capacity of salmonids to cope with feeds containing higher levels of dietary starch.
