WT and AUF1 KO satellite cell RNA-sequencing

Project description:Following skeletal muscle injury, muscle stem cells (satellite cells) are activated, proliferate, and differentiate to form myofibers. We show that mRNA decay protein AUF1 regulates satellite cell function through targeted degradation of specific mRNAs. AUF1 targets certain mRNAs containing 3 AU-rich elements (AREs) for rapid decay. Auf1-/- (KO) mice undergo accelerated skeletal muscle wasting with age and impaired muscle repair following injury. Satellite cell mRNA analysis and regeneration studies demonstrate that auf1-/- satellite cell self-renewal is impaired due to increased stability and overexpression of ARE-mRNAs. Control of ARE-mRNA decay by AUF1 and potentially other ARE-binding proteins represents a mechanism for adult stem cell regulation and is implicated in human muscle wasting diseases. Functional microarray analysis of neonatal monocyte-derived macrophages differentiated in the presence or absence of human lactoferrin

Project description:Monocytes were isolated from umbilical cord blood of term born infants and differentiated with M-CSF in the presence or absence of human lactoferrin for 7 days. Afterwards cells were stimulated with LPS or left untreated, RNA isolated using a RNeasy Plus Micro Kit and samples submitted to microarray-analysis. The experiments were performed to investigate the functional impact of human lactoferrin, a transferrin glycoprotein highly prevalent in human breast-milk, on neonatal macrophages. The experimental model serves as a proxy for the proteins' immunoregulatory function within the developing neonatal gut.

High-throughput RNA sequencing analysis of mouse peritoneal cells from wild type and Lactoferrin knockout mouse during Lipopolysaccharide(LPS) stimulation

Project description:Purpose: The goal of this study is using RNA sequencing analysis to evaluate the role of lactoferrin in the LPS-induced acute inflammation in mouse Methods: 4 hs after intraperitoneal injection with LPS(4mg/kg) or equal volume PBS , the peritoneal cells were isolated from the peritoneal lavage fluid from wild type and lactoferrin knockout mice (n=3 for each group) . Then the cell RNA was sequenced by the HiSeq 2500 high-throughput sequencing service (Oebiotech, Shanghai, China). Data were extracted and normalized according to the manufacturer’s standard protocol. Results: Log-fold changes of up- or down-regulated mRNAs between the control and experiment group were selected with a significance threshold of p<0.05. Conclusions: Our study describes the mRNA expression profile changes of peritoneal cells from wild type and lactoferrin knockout mice during LPS-induced acute inflammation

Global transcriptome analysis of paracancerous lung tissues from the B16 metastasis mice model under the influence of lactoferrin knockout

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are using transcriptome profiling (RNA-seq) to evaluate the effects of lactoferrin deficiency on the global transcriptome of the lung microenvironment of the B16 metastasis mice model. Methods: 2×10^5 B16-F10 cells were injected into WT and lactoferrin kncoutout (Lf-/-) mice (each group has 10 eight-weeks-old male mice) through tail vein. At 3 weeks post injection,the paracancerous lung tissue were isolated (not including any metastatic colonies). RNAs were extracted by Trizol and sequenced by Solexa high-throughput sequencing service (Oebiotech, Shanghai, China). Data were extracted and normalized according to the manufacturer’s standard protocol.Each group has two mice paracancerous lung tissues be tested. Results: There are significant gene expression profile changes between WT and Lf-/- tumor-bearing mice. Conclusions: Our study describes the global transciptome changes of paracancerous lung tissues from the B16 metastasis mice model under the influence of lactoferrin kncokout.
