Project description:We have shown that Chrnb4-cre; Dicerflox/flox retinas (Dicer CKO) display a cone dystrophy by postnatal day P21, independent of rod degeneration. To elucidate which gene pathways were affected by Dicer loss, RNA sequencing was performed in whole neural retinas of P21 control and Dicer CKO mice. This experiment aims to identify which genes were differentially expressed between Dicer conditional knockout and control mice.

Project description:β-cell specific Mettl14 knock-out mice display reduced N6-methyladenosine (m6A) levels and recapitulate human Type II diabetes (T2D) islet phenotype with early diabetes onset and mortality secondary to decreased β-cell proliferation and insulin degranulation. To gain insights into the role of m6A in regulating the IGF1/insulin -> AKT - > PDX1 pathway and to dissect the signaling networks modulating AKT phosphorylation, we subjected freshly isolated islets from control and Mettl14 knock-out mice to phospho-antibody microarrays.

Transcriptome analysis of tendon in Mkx conditional knock out mice

Project description:Tendons are crucial connective tissues that link muscle to bone. We have demonstrated that the transcription factor Mkx plays a pivotal role in tendon development and homeostasis by regulating collagen fibril organization through tendon cell fate determination from progenitor cells and promoting the expression of type I collagen and proteoglycans. However, the precise role of Mkx in tendon homeostasis after birth is not well understood. Therefore, we generated conditional Mkx knockout mice by crossing Mkx-CreER mice with Mkx floxed mice. Following tamoxifen administration, we performed transcriptome analysis of tendons from control mice (Mkx-CreER/+ and Mkx flox/+) and Mkx conditional knockout mice (Mkx-CreER/flox)
