Project description:Here we report the reprogramming of human fibroblasts to produce chemically-induced osteogenic cells (ciOG), and explore the potential uses of ciOG in bone repair and disease treatment. A chemical cocktail of RepSox, forskolin, and phenamil was used for osteogenic induction of fibroblasts by activation of RUNX2 expression. Following a maturation, the cells differentiated toward an osteoblast phenotype that produced mineralized tissue. When generated on a nanofiber substrate ciOG accelerated osteogenic gene expressions and mineralization compared to PCL film followed by bone matrix formation in a calvarial defect, indicating that the engineered biomaterial promotes the osteogenic capacity of ciOG in vivo.

Project description:We developed new isosorbide derivatives (propoxylated or ethoxylated isosorbide) which have more reactivity in PCL-PU copolymer synthesis than the original isosorbide. PCL-PU copolymer incorporated propoxylated isosorbide (ISB-P) exhibited notable mechanical performance (50 MPa tensile strength and 1150% elongation with hyperelasticity under cyclic load), ultra-cell-adhesive property, and enhanced osteogenic differentiation. There was no significant difference in surface characteristics between ISB-P and PCL), however, mRNA profiles showed higher expressions of heat shock proteins which have been reported to promote osteogenic differentiation at 4 h after cell seeding in rMSC cultured on ISB-P compared to PCL. In conclusion, the engagement of heat shock protein to the ISB-P surface during cell attachment may promote osteogenic differentiation in ISB-P.

Plasma functionalization of PCL nanofibers changes protein interactions with cells resulting in increased cell viability

Project description:The surface properties of electrospun scaffolds can greatly influence protein adsorption and thus strongly dictate cell-material interactions. In this study, we aim to investigate possible correlations between the surface properties of argon, nitrogen and ammonia/helium plasma-functionalized polycaprolactone (PCL) nanofibers and their cellular interactions by examining the protein corona patterns of the plasma-treated nanofibers as well as the cell membrane proteins involved in cell proliferation. As a result of the performed plasma treatments, PCL nanofiber morphology was preserved while wettability was improved profoundly after all treatments because of the incorporation of polar surface groups. Depending on the discharge gas, different types of groups are incorporated which influenced the resultant cell-material interactions. Argon plasma-functionalized PCL nanofibers, only enriched by oxygen-containing functional groups, were found to show the best cell-material interactions, followed by N2 and He/NH3 plasma-treated samples. SDS-PAGE and LC-MS clearly indicated an increased protein retention compared to non-treated PCL NFs. The most differential proteins surface a distinct molecular machinery mediating the increased proliferation and viability on plasma treated NF. Finally, differential protein analysis between the different plasma’s gives a first hint into the molecular underpinning of the superior performance of Argon NF.

A chemical cocktail of RepSox, forskolin, and phenamil was used for osteogenic induction of fibroblasts by activation of RUNX2 expression. Following a maturation, the cells differentiated toward an osteoblast phenotype that produced mineralized tissue. Single-cell RNA sequencing analysis of ciOG identified unique clusters of cells, among which the cluster of active cells related to osteogenic cells was ~49% of the cells, or ~80% of active cells; another cluster of active cells (~12% of the cells) was related to fibroblasts. The reprogramming trajectory over time revealed the transition from fibroblasts to mainly osteogenic cells or to an inactive state.

Project description:Microarray analysis was used to evaluate expression differences from a single donor human bone marrow stromal cells (hBMSCs) as a function of varied polymer-based tissue engineering scaffolds. These scaffolds include polycaprolactone (PCL) nanofibers (PCL_NF), films (PCL_SC), poly D,L-lactic acid (PDLLA) nanofibers (PDLLA_NF), films (PDLLA_SC), tissue culture polystyrene (TCPS) and TCPS with osteogenic supplements (TCPS_OS). The results revealed that scaffold structure was able to significantly affect gene expression, with nanofiber scaffolds inducing similar gene expression patterns to hBMCSs cultured with osteogenic media. A library of scaffolds prepared from polycaprolactone or poly D,L-lactic acid was sythesized and cultured with hBMSCs for 14 days with RNA extracted from cells on Day 1 and Day 14. Gene expression analysis was performed using BRB ArrayTools. SC = spun coat, BNF = big nanofiber, TCPS = tissue culture polystyrene, TCPS+OS = tissue culture polystyrene with osteogenic supplements. This data forms is part of a pending publication: Baker et al. Ontology Analysis of Global Gene Expression Differences of Human Bone Marrow Stromal Cells Cultured on 3D Scaffolds or 2D Films and is a subset of the 72 array data referenced in ( Kumar et al. The determination of stem cell fate by 3D scaffold structures through the control of cell shape, Biomaterials (2011) 32, 9188-9196.) The 72 array data set is submitted separately to GEO as GSE50743.
