Effect anti-TNF-alpha treatment on CD4+ T cell activation

Project description:The experiment was designed to test how autocrine TNF-alpha impacts human CD4+ T cell activation. Naïve CD4+ T cells from healthy human blood donors were either left unactivated or activated with anti-CD3/anti-CD28 in presence of an isotype control antibody or anti-TNF-alpha antibody. RNA was extracter at 48 hours. Analysis revealed that anti-TNF-alpha treatment prevented upregulation of activation-induced T cell metabolic, signaling and effector molecules.

Proteomic analysis of pemphigus autoantibodies indicates a much larger, more diverse, and dynamic repertoire than determined by B-cell genetics

Project description:Genetic techniques such as antibody phage display have indicated an oligoclonal autoantibody response in various autoimmune diseases including pemphigus. These techniques have limited sampling of B cell clones. Characterization by mass spectometry of pemphigus serum autoantibodies affinity-purified on the autoantigen desmoglein indicates a much more polyclonal response. Conversely, many genetically detectable anti-desmoglein B cells do not contribute detectably to the serum antibody response. There is no convergence of the autoantibody response among patients, as determined by CDR3 sequence or heavy chain variable gene usage, implying targeting of these genes will not be a useful therapeutic strategy. Longitudinal analysis of autoantibodies over years indicates that, although many antibody clones persist, the proportion of each clonal antibody changes. These studies indicate a dynamic and very diverse autoantibody response not revealed by genetic studies, and explain why similar overall anti-desmoglein titers may give variable disease activity.

Enhanced Staphylococcus aureus vaccine protection by uncoupling of the alpha-toxin-ADAM10 interaction

Project description:Staphylococcus aureus is the leading global cause of bacterial infection-associated mortality, as vaccine development has been elusive. S. aureus alpha-toxin (Hla) is an essential virulence factor in disease. In children, the anti-Hla neutralizing antibody response is the only known correlate of human protective immunity. We demonstrate that the anti-Hla response is limited in the first 2 years of life, illuminating a vaccine-targetable population. We assessed whether Hla antigenic variants would enhance vaccine-mediated immunity in neonatal mice. We identified a variant (HlaH35L/R66C/E70C, HlaHRE) that elicits a nearly 100-fold increase in the neutralizing anti-Hla antibody response compared to other candidate Hla antigens. HlaHRE immunization durably protects against skin and soft tissue infection, enhancing the T follicular helper response and germinal center B cell response. Protective immunity was also conferred to offspring following maternal immunization with HlaHRE. These findings thereby define a novel path for universal S. aureus vaccine development at the maternal-infant interface.

Mucosal gene signatures to predict response to infliximab in patients with ulcerative colitis

Project description:Infliximab, an anti-TNF-alpha monoclonal antibody, is an effective treatment for ulcerative colitis (UC) with over 60% of patients responding to treatment and up to 30% reaching remission. The mechanism of resistance to anti-TNF-alpha is unknown. This study used colonic mucosal gene expression to provide a predictive response signature for infliximab treatment in UC. Keywords: drug response Twenty-four patients with active UC, refractory to corticosteroids and/or immunosuppression, underwent colonoscopy with biopsies from diseased colon within a week prior to the first intravenous infusion of 5 mg infliximab per kg body weight. Response to infliximab was defined as endoscopic and histologic healing at 4-6 weeks after first infliximab treatment. Six control patients with normal colonoscopy were included. Total RNA was isolated from colonic mucosal biopsies, labelled and hybridized to Affymetrix Human Genome U133 Plus 2.0 Arrays.

Project description:RNA was isolated from material that had been subjected to immunoprecipitation (IP) from RKO cells that were either left untreated or irradiated with 20 J/m2 UVC and collected 1h later; IP assays were carried out using either an antibody recognizing RNA-binding protein TIAR, or using a control IgG1 antibody. RNA was reverse-transcribed in the presence of [alpha-33P]dCTP and the radiolabeled product used to hybridize human cDNA arrays. The experiment was repeated using three independent sample sets. Expression data from oral squamous cell carcinomas, 3A and HO1-N-1

Project description:Co-culture with human oral squamous cell carcinoma cells, 3A or NEM, or culture with each of their conditioned medium induced many osteoclasts from osteoclast precursor cells, which were generated by a 24-h pretreatment of RANKL or TNF-α. However, HO1-N-1 cells did not induce any osteoclasts. Osteoprotegerin, a decoy RANKL receptor, denosumab, an anti-RANKL antibody drug, and infliximab, an anti-TNF-α antibody drug, did not prevent this tumor-associated osteoclastogenesis. We compared the expression of molecules associated with osteoclastogenesis between 3A and HO1-N-1 cells.
