Project description:Myelofibrosis (MF) is a progressive, bone marrow (BM) malignancy associated with monocytosis, and is believed to promote the pathological remodelling of the extracellular matrix. Here, we show that the mechanical properties of MF, namely the liquid-to-solid properties (viscoelasticity) of BM, contribute to aberrant differentiation of monocytes. Human monocytes cultured in stiff, elastic hydrogels show pro-inflammatory polarization and differentiation towards dendritic cells, as opposed to those cultured in a viscoelastic matrix.

A comprehensive characterization of the viscoelastic properties of Bovine Submaxillary Mucin (BSM) and the effect of additives

Project description:This study presents a comprehensive characterization of the viscoelastic and structural properties of Bovine Submaxillary Mucin (BSM), a good biomaterial model system for mucins and mucus in general. We conducted concentration studies of BSM and examined the effects of various additives - sodium chloride, calcium chloride, lysozyme, and DNA - on its rheological behavior. Through oscillatory and shear flow macrorheological experiments, we established a detailed correlation between the concentration of BSM and its viscoelastic properties, uncovering changes in the rheological moduli with the addition of sodium and calcium chloride, particularly at higher concentrations. This highlights the mucins responsiveness to ionic strength and composition variations. The presence of lysozyme and DNA further influenced the rheological characteristics of BSM, adding a new dimension to our understanding of mucin behavior. The rheological spectra could be well-described by a Fractional Kelvin-Voigt Model (FKVM), which allows to describe the complete behavior with a minimum of model parameters. A detailed proteomics analysis provided insight into the molecular interactions within BSM, enhancing our understanding of its structural intricacies. Complementing this, cryo-scanning electron microscopy (cryo-SEM) was employed to visualize the microstructural alterations and network properties in relation to the rheological data. By elucidating the complex interplay between mucin concentration, environmental conditions, and viscoelastic properties, this research significantly contributes to the field of mucus research and lays an important basis for future studies and advanced applications of numerous mucin-based biomaterials.

Distinct Combinatorial Events Generated by ECM Degradation Dictate Cell Behavior

Project description:It is well established that the expression profiles of multiple and possibly redundant matrix remodeling proteases (e.g. collagenases) strongly differ in health, disease and development. Although enzymatic redundancy might be inferred from their close similarity in structure, their in-vivo activity can lead to extremely diverse tissue-remodeling outcomes. We observed that proteolysis of collagen-rich natural extracellular matrix (ECM), generated uniquely by individual homologous proteases, leads to specific combinatorial events, which eventually affects overall ECM topography, visco-elastic properties and composition. We reveal striking differences in the movement and signaling patterns, morphology, and gene expression profiles of cells interacting with natural collagen-rich ECM degraded by different collagenases. Thus, unlike envisioned before matrix-remodeling systems are not redundant and give rise to precise ECM-cell crosstalk. As ECM proteolysis is an abundant biochemical process critical to tissue homoeostasis, these results improve our fundamental understanding of combinatorial factors dictating cell behavior.

ECM deposition is driven by caveolin1-dependent regulation of exosomal biogenesis and cargo sorting

Project description:The composition and physical properties of the extracellular matrix (ECM) critically influence tumor progression, but the molecular mechanisms underlying ECM layering are poorly understood. Tumor-stroma interaction is critically dependent on cell-cell communication mediated by exosomes, small vesicles generated within multivesicular bodies (MVBs). Here, we show that caveolin-1 (Cav1) is a central modulator of both exosome biogenesis and exosomal protein cargo sorting through the regulation of cholesterol content at the endosomal compartment/MVBs. Quantitative proteomics profiling revealed that Cav1 is required for exosomal sorting of ECM protein cargo subsets including tenascin-C (TnC), and for fibroblast-derived exosomes to efficiently depose ECM and promote tumor cell invasiveness. Cav1-driven exosomal ECM deposition not only promotes local stromal remodeling, but also the generation of distant ECM-enriched stromal niches. These results support a model by which Cav1 is a central regulatory hub for tumor-stroma interactions through a novel exosome-dependent ECM deposition mechanism.
