Project description:Background: The disruption of the circadian clock is associated with inflammatory and immunological disorders. BMAL2, a critical circadian protein, forms a dimer with CLOCK, activating transcription. Extracellular cold-inducible RNA-binding protein (eCIRP), released during sepsis, can induce macrophage endotoxin tolerance. We hypothesized that eCIRP modulates BMAL2 expression and promotes macrophage endotoxin tolerance through triggering receptor expressed on myeloid cells-1 (TREM-1). Methods: C57BL/6 wild-type (WT) male mice were subjected to sepsis by cecal ligation and puncture (CLP). Serum was collected 20 hours post-CLP to assess eCIRP levels. Peritoneal macrophages (PerM) and immortalized bone marrow-derived macrophages (iBMDMs) were seeded into plates and treated with recombinant mouse (rm) CIRP (eCIRP) at various doses. Induction of BMAL2 activation in iBMDM was performed by transfection of BMAL2 CRISPR activation plasmid. Circadian gene expression profiles in eCIRP-treated macrophages were determined by PCR array and confirmed by qPCR. Cytokine levels in the culture supernatants were assessed by ELISA. The interaction of BMAL2 in the PD-L1 promoter was determined by computational modeling and confirmed by the BIAcore assay. Results: PCR array screening of circadian clock genes in peritoneal macrophages treated with eCIRP revealed the elevated expression of BMAL2, CRY1, and PER2. eCIRP serum levels were increased in septic mice. In eCIRP-treated macrophages, TREM-1 deficiency prevented the upregulation of these circadian genes. Macrophages pre-treated with eCIRP exhibited reduced TNFá and IL-6 release upon LPS challenge, indicating macrophage endotoxin tolerance. In addition, eCIRP increased the expression of PD-L1, IL-10, and STAT3, markers of immune tolerance. Interestingly, TREM-1 deficiency reversed eCIRP-induced macrophage endotoxin tolerance. In macrophages, inducible BMAL2 expression correlated with increased PD-L1 expression. In septic human patients, blood monocytes exhibited increased expression of BMAL2 and PD-L1 in comparison to healthy subjects. Computational modeling and BIAcore assay identified a putative binding region of BMAL2 in the PD-L1 promoter, suggesting BMAL2 positively regulates PD-L1 expression in macrophages. Conclusion: eCIRP upregulates BMAL2 expression via TREM-1, leading to macrophage endotoxin tolerance in sepsis. Targeting eCIRP to maintain circadian rhythm may correct endotoxin tolerance and enhance host resistance to bacterial infection.

Project description:In vitro models are often used to study the functions of macrophages, including the process of phagocytosis. The use of primary macrophages has limitations associated with the individual characteristics of animals, which can lead to insufficient standardization of the results obtained. This disadvantage is lacking in immortalized cell lines. The PMJ2-R cell line was derived from in vivo immortalization and retains many of the primary peritoneal macrophages functions, but little is known about its differences from normal cells. In this article, we carried out a comparative analysis of the proteomes of PMJ2-R cells and primary peritoneal macrophages isolated from the C57BL/J6 mice. Particular attention was paid to the analysis of proteins involved in the process of phagocytosis. A total of 4005 proteins were identified, of which 797 were quantified. Our results indicate that there are significant differences in the abundance of a large number of proteins, including important proteins associated with the process of phagocytosis, such as Elmo1, Gsn, Hspa8, Itgb1, Ncf2, Rac2, Rack1, Sirpa, Sod1, C3, Msr1. Thus, when using PMJ2R cells as model cells in the study of the peritoneal macrophages functions, features revealed in this study should be taken into account.

Project description:To compare up-regulation of genes following CpG activation, we performed microarray analysis of activated macrophages from B6 and F1(B6xMOLF) mouse strains. Cells were activated for 0, 2 and 4 hrs with 200nM of type B CpG. Levels of mRNA for many genes differened dramatically between the strains Peritoneal macrophages were elicited from pertoneal cavity of mice 3 days after thioglycollate injection. The cells were plated overnight at a density of 5 million cells per 100 mm dish and activated the next day with 200 nM CpG. Project description:Alveolar macrophages are the first line of defense against pathogens in the lungs of all mammalian species and therefore may constitute an appropriate therapeutic target cell in the treatment and prevention of opportunistic airway infections. Analysis of alveolar macrophages from several species has revealed a unique cellular phenotype and transcriptome, presumably linked to their distinct airway environment and function in host defense. The current study extends these findings to the horse. Firstly, the transcriptome of equine alveolar macrophages was compared to that of equine peritoneal macrophages. This comparison revealed the differential basal expression of 451 genes. Gene expression analysis revealed an alternative macrophage polarisation profile in alveolar macrophages and a hybrid macrophage activation profile in peritoneal macrophages, a phenomenon potentially attributable to a degree endotoxin tolerance. Secondly, the change in gene expression profile of equine AMs following lipopolysaccharide stimulation was investigated. This revealed a significant change in the expression of 240 genes, including well known upregulated inflammatory genes.
