Project description:Hepatic Natural Killer (he-NK) cells are innate immune effectors that contribute to immune tolerance and eliminate dangerous “non-self” antigens. He-NK cells fall into two groups, liver resident (rNK) cells, mainly cytokine secretors, and infiltrating conventional NK (cNK) cells, which are cytotoxic. He-NK cell dysfunction and contribution to end-stage liver disease are not fully understood. The present study evaluates the transcriptomes of he-NK subpopulations in chronic hepatic disease of different etiologies, including sterile inflammation (non-alcoholic steatohepatitis - NASH), autoimmunity (primary sclerosing cholangitis - PSC), and viral infection (hepatitis C - HCV).

Molecular crosstalk between hepatocytes and hepatic stellate cells.

Project description:Aim of the study was to characterize at a molecular level (changes in transcriptomes) the crosstalk between tumor hepatocytes and activated hepatic stellate cells (HSC) in liver cancer. This was adressed by using a coculture model system of HepaRG cell line (tumor hepatocytes, human), and LX2 cell line (HSC, human). By using genome-wide expression profiling, we demonstrated that hepatocyte-HSC crosstalk is bidirectional and results in the deregulation of functionally relevant gene networks. HepaRG and LX2 cells were cultured alone in serum- and DMSO-free William's E medium or together using 1 M-BM-5m pore size transwell inserts which allow diffusion of media components but prevent cell migration (BD Biosciences, San Jose, CA). Triplicate experiments were performed: HepaRG (culture versus coculture), LX2 (culture versus coculture).

Regulation of hepatic microRNAs in response to early stage Echinococcus multilocularis egg enfection in C57BL/6 mice

Project description:We report a comprehensive analysis of the hepatic miRNA transcriptome at one-month post-infection of experimental primary alveolar echinococcosis (AE), a parasitic infection a parasitic infection caused upon ingestion of E. multilocularis eggs. In infected mice, the expression levels of 28 miRNAs were significantly altered. Of these, 9 were up-regulated and 19 were down-regulated as compared to the non-infected controls. We also studied the issue related to functionally mature arm selection preference (5p and/or 3p) from the miRNA precursor and showed that 9 pre-miRNAs exhibited different arm selection preferences in normal versus infected liver tissues. In conclusion, this study provides first evidence that miRNAs are regulated early in primary murine AE. Thus, further research on the regulation of miRNAs during AE is needed, since miRNAs constitute an attractive potential option for development of novel therapeutic approaches against AE.
