Perioperative Testosterone Replacement Therapy in Sarcopenic Male Colorectal Cancer Patients

Project description:With increasing age and the additional impact from the bowel cancer and the chemotherapy and/ or radiotherapy it has been described that testosterone (a male hormone produced naturally in the body) levels are reduced. Testosterone has an impact on numerous body functions including the muscle mass and quality. Previous studies have identified that muscle mass is reduced as a result of ageing but also because of the deleterious effect of cancer and chemotherapy and/or radiotherapy. There is growing evidence from published studies that patients with better muscle mass and quality, do better after surgery. Mr Jenkins and his team are therefore looking at ways, the investigators can try and prevent or reduce this muscle loss and therefore improve patient outcomes. The aim of this study is to assess whether using testosterone replacement therapy in the form of a topically applied gel daily for a total of 12 weeks, is feasible and acceptable by the patients who are diagnosed with colorectal cancer and are waiting to undergo surgery. The investigators will also collect information related to the testosterone replacement therapy such as questionnaires on the quality of life, fatigue and muscle mass, and blood biomarker changes in the blood.

Project description:We generated a mouse model (ARNoC/Y) with a mutation in the FQNLF motif located on the aminoterminal domain (NTD) that disrupts the N/C interactions of the androgen receptor (AR). These mice exhibit a normal male phenotype. To be able to compare WT males and ARNoC/Y mice more directly, we performed a castration experiment with testosterone (T) supplementation for 4 days. Since slightly differences in prostate weight were observed, we performed RNA-seq on these prostate to evaluate the biological relevance.

Project description:We generated a mouse model (ARLmon/Y) with disrupted dimerization of the ligand-binding domain 25 (LBD) of the androgen receptor (AR). These mice exhibit an external female phenotype with undescended testes. Histological analysis of the testes of ARLmon/Y mice revealed some residual spermatogenesis. Hormone measurements in serum of the ARLmon/Y mice showed significantly high levels of androgens. To study if there is any remaining AR activity, we orchidectomized WT and ARLmon/Y mice and gave them supraphysiological testosterone replacement for 4 days. RNA-seq was performed on kindeys.

Transcription profiling of mouse prostate after castration and hormone replacement

Project description:In this study, we used microarray analysis to determine gene expression profile changes in the mouse prostate following castration and hormone replacement. We first identified genes with significant expression changes in each of these two processes and then generated a list of androgen responsive genes and a list of genes whose expression were inversely correlated with the presence of androgen. The analysis of this data set is described in Wang et al., Differentiation, 2006 Experiment Overall Design: C57/B6 mice of 10-12 weeks old (Charles River, Hollister, CA) were used. Mice were castrated, and at 3 or 14 days after surgery (C3 or C14), mice from these groups were sacrificed and whole prostates were collected. Prostates were also collected from sham-treated mice for control (N3). To obtain re-growing or regenerating prostate, mice that had been castrated 14 days before were implanted with testosterone (T) pellet (15 mg/pellet/mouse, Innovative Research, Sarasota, FL). Three days after pellet implantation, mice were sacrificed and prostates were collected (C14+T3). Five mice were used for each treatment/time point. Total RNA was purified with RNeasy kit.

Renal transcriptome changes in testosterone deficient-pigs fed a high-fat diet

Project description:Purpose: Obesity and dyslipidemia are associated with increased risk of renal disease.Testosterone deficiency aggravated high-fat diet-induced obesity and hypercholeterolemia. However,whether testosterone deficiency or testosterone deficiency-induced dyslipidemia aggravate the progression of renal disease is not clear. To gain insight into the role of testosterone in modulating renal lipid metabolism, we profiled renal gene expression by RNA-Seq in HFC-fed intact male pigs (IM), castrated male pigs (CM), and castrated male pigs with testosterone replacement (CMT). Methods: Sexually mature male miniature pigs were either surgical castrated or sham-operated, and castrated with testosterone replacement. We administrated to pigs a high-fat and high-cholesterol (HFC) diet for twelve weeks. RNA-Seq was employed to profile renal gene expression in pigs with different testosterone levels. Conclusions: This study demonstrated that testosterone deficiency aggravated renal lipid accumulation in pigs fed an HFC diet and that these effects could be reversed by testosterone replacement therapy. Impaired metabolic processes, bile acid secretion,estrogen signaling pathway and enhanced triglyceride synthesis may contribute to the increased renal lipid accumulation induced by testosterone deficiency and an HFC diet.

RNA-Seq was employed to profile hepatic gene expression in pigs with different testosterone levels. Liver mRNA profiles of intact male pigs fed a HFC diet, castrated male pigs fed a HFC diet, and castrated male pigs treated with testosterone fed a HFC diet were generated by deep sequencing, using Illumina HiSeq 2000.

Testosterone deficiency induces changes of the transcriptomes of visceral adipose tissue in miniature pigs fed a high-fat and high-cholesterol diet

Project description:Testosterone deficiency causes fat deposit, particularly in the visceral region, and its replacement might reverse fat accumulation, but the underlying mechanisms of such processes are largely unclear. To gain insights into the genome-wide role of testosterone on visceral adipose tissue (VAT), RNA-Seq was used to investigate testosterone deficiency induced changes of VAT in miniature pigs fed a high-fat and high-cholesterol (HFC) diet among intact male pigs (IM), castrated male pigs (CM), and castrated male pigs with testosterone replacement (CMT) treatments. The results showed that testosterone deficiency induced VAT deposit and increased serum leptin level. Moreover, a total of 1,732 differentially expressed genes (DEGs) were identified between different two groups. Compared with gene expression profiles in IM and CMT pigs, upregulated genes in CM pigs, i.e., LOC100520753 (CD68), LCN2, EMR1, NCF1 (p47phox), and NOX2 (GP91-PHOX), were mainly enriched in inflammatory response, oxidation-reduction process, and response to oxidative stress, while downregulated genes in CM pigs, i.e., DIO3, PCK1, and ABHD5, mainly focused on small molecule metabolic process. Taken together, our study provides a novel genome-wide view on the role of testosterone on VAT deposit under HFC diet, thus improving our understanding of the molecular mechanisms involved in VAT changes induced by testosterone deficiency.
