Project description:Germinal center (GC) reaction in the lymphoid organs is a pivotal process for humoral immune response. The miR-17~92 family miRNAs have been demonstrated that regulate TFH cell differentiation, GC reaction and antibody response. However, whether they may also have functions in B cell activation and differentiation in GC reaction remain largely unknown. Here we show that mice with deletion of miR-17~92 in B cells exhibited reduced GC B cell formation and profound impaired plasma cell (PC) differentiation as well as decreased antibody response upon protein antigen immunization or chronic LMCV infection. In an in vitro plasma cell differentiation (iGCB) system, we found that miR-17~92 deficiency results in markedly decreased in vitro plasma cell (iPC) differentiation and defective NFB activation. Moreover, we developed and performed a screen by CRISPR/small guide (sg) RNA-mediated gene editing among the target genes of miR-17~92 in iGCB culture system. Intriguingly, Socs3 deletion substantially restored iPC differentiation and NFB activation in miR-17~92 deficient B cells, suggesting that Socs3 acts as a negative regulator of NFκB pathway involved in the regulation of iPC differentiation by miR-17~92. However, IL-21-induced STAT3 activation was not affected by miR-17~92 deficiency during iPC differentiation. Mechanistically, Socs3 interacts with MAP3K14 (NIK) and that promotes the ubiquitination of NIK and subsequent proteasome-mediated degradation of NIK, thereby leading to inhibition of the processing of p100 in non-canonical NFκB pathway and PC differentiation.

Project description:The microRNAs encoded by the miR-17~92 polycistron are commonly overexpressed in cancer and orchestrate a wide range of oncogenic functions. Here, we identify a novel mechanism for miR-17~92 oncogenic function through the disruption of endogenous miRNA processing. We show that upon oncogenic overexpression of the miR-17~92 primary transcript (pri-miR-17~92), the Microprocessor complex remains associated with partially processed intermediates that aberrantly accumulate. These intermediates reflect a series of hierarchical and conserved steps in the early processing of the pri-miR-17~92 transcript. Encumbrance of the Microprocessor by miR-17~92 intermediates leads to the broad, but selective, downregulation of co-expressed polycistronic miRNAs, including miRNAs derived from tumour suppressive miR-34b/c, and from the Dlk1-Dio3 polycistrons. We propose that the newly identified steps of polycistronic miR-17~92 biogenesis contribute to the oncogenic re-wiring of gene regulation networks. Our results reveal new functional paradigms for polycistronic miRNAs in cancer.

Microarray profiling of cultured oligodendrocytes identifies the miR-17~92 family of miRNA cluster as highly enriched miRNAs in oligodendrocytes.We specifically deleted the miR-17~92 cluster in oligodendrocytes using the 2´3´-cyclic nucleotide 3´-phosphodiesterase (CNP)-Cre mice. Absence of miR-17~92 leads to a reduction of oligodendrocyte number in vivo and we find that the expression of these miRNAs in primary cultures of oligodendrocytes promotes cell proliferation by influencing Akt signalling. Together, these results suggest that the miRNA pathway is essential in determining oligodendroglial cell number and that the miR-17~92 cluster is crucial in this process.

Transcriptome microarray profiling was used for the identification of mRNAs enriched in oligodendrocytes. Total RNA lysates from primary oligodendroctes compared to primary astrocytes were analysed regarding their mRNA levels. Three independent samples for each of the two cell types were used. The study was initially designed with a comparison of oligodendrocytes, astrocytes and microglia. However, only the comparison of oligodendrocytes and astrocytes is discussed in the present study.
