Project description:Heterosis refers to the superior performance of the hybrid compared with its parental lines. Despite several genetic models and different molecular pathways proposed to explain heterosis, it remains unclear how cells integrate complementary gene expression, metabolic accumulation and/or hormone signaling to drive heterotic growth. In this work, using multiple omics combined with biochemical analyses, we showed an enhanced TORC1 signaling of the elite hybrid rice Shanyou 63 (SY63) relative to the parental lines, which was associated with accumulation of growth-promoting (translation, cell division) and energy metabolic (glycolysis and TCA) proteins energy metabolic activities, and superior growth of panicle meristem. Metabolism of nuclear-cytosolic acetyl-Coenzyme A was also enhanced in the hybrid, which paralleled with increases of histone H3 acetylation to selectively target growth-promoting and metabolic genes expression. Lysine acetylation of cellular proteins including TORC1, ribosomal proteins, and energy metabolic enzymes was also augmented and/or remodeled, potentially modulating their activities. The data revealed that the positive feedback loop between TOR-signaling, energy-producing metabolic activity, lysine acetylation, and growth-promoting gene expression is a mechanism underling the superior growth rate of the hybrid. The results implied that an enhanced activity in hybrids would drive and sustain the TOR signaling feedback loop to promote superior growth, which may represent a general mechanistic model for heterosis.

Project description:Although utilization of heterosis has largely improved the yield of many crops worldwide, the underlying molecular mechanism of heterosis, particularly for allopolyploids, remains unclear. Here, we compared epigenome and transcriptome data of an elite hybrid and its parental lines in three assessed tissues (seedling, flower bud, and silique) to explore their contribution to heterosis in allopolyploid B. napus. Transcriptome analysis illustrated that a small proportion of non-additive genes in the hybrid compared with its parents, as well as parental expression level dominance, might have a significant effect on heterosis. We identified histone modification (H3K4me3 and H3K27me3) variation between the parents and hybrid, most of which resulted from the differences between parents. H3K4me3 variations were positively correlated with gene expression differences among the hybrid and its parents. Furthermore, H3K4me3 and H3K27me3 were rather stable in hybridization and were mainly inherited additively in the B. napus hybrid. Together, our data revealed that transcriptome reprogramming and histone modification remodeling in the hybrid could serve as valuable resources for better understanding heterosis in allopolyploid crops.

Digital analysis of gene expression using high throughput sequencing technology revealed several differential gene expression patterns in the hybrid, including additivity, nonadditivity and overdominance, etc. Alteration of OsHDT1 levels affected many genes specifically in the hybrid. In addition, we show that increased OsHDT1 could suppress overdominance gene expression, providing evidence of overdominance gene action in heterosis. These results not only support the overdominance hypothesis to explain heterosis, but also provide evidence that variation in the levels of single trans-acting regulatory proteins such as chromatin factors is important to establish differential gene expression pattern in the hybrid.

Project description:Although heterosis has significantly contributed to increases in worldwide crop production, the molecular mechanisms regulating this phenomenon are still unknown. In the present study, we used a comparative proteomic approach to explore hybrid vigor via the proteome of both the popcorn L54 ♀ and P8 ♂ genotypes and the resultant UENF/UEM01 hybrid cross. To analyze the differentially abundant proteins involved in heterosis, we used the primary roots of these genotypes to analyze growth parameters and extract proteins. The results of the growth parameter analysis showed that the mid- and best-parent heterosis were positive for root length and root dry matter but negative for root fresh matter, seedling fresh matter, and protein content. The comparative proteomic analysis identified 1343 proteins in the primary roots of hybrid UENF/UEM01 and its parental lines; 220 proteins were differentially regulated in terms of protein abundance. A total of 62 regulated proteins were classified as non-additive, of which 53.2% were classified as high-parent abundance (+), 17.8% as above high-parent abundance (+ +), 16.1% as below low-parent abundance (- -), and 12.9% as low-parent abundance (-). A total of 22 biological processes were associated with non-additive proteins; processes involving translation, ribosome biogenesis, and energy-related metabolism represented 45.2% of the non-additive proteins. Our results suggest that heterosis in the popcorn hybrid UENF/UEM01 at an early stage of plant development is associated with an up-regulation of proteins related to synthesis and energy metabolism

Project description:Heterosis has long been exploited for crop breeding; however, the genetic mechanisms, particularly the initial establishment of heterosis during the early vegetative growth phase, remain elusive. The biggest challenge for that is to exclude noise genes from the identified heterosis-related candidates. Herein, we use nutrient-deficient hybrid with no measurable growth heterosis as control to filter potential background noise differentially expressed genes In this dataset, we compared the differentially expressed genes in both heterotic and non-heterotic hybrid. After filtering these irrelevant genes, only 336 differentially expressed genes (DEGs), which is significantly lower than those of previous reports, were identified as heterosis-related genes in a super hybrid rice of Liang-You-Pei-Jiu (LYP9) at early-tillering stage

Project description:Hybrid breeding is of economic importance in agriculture for increasing yield, yet the basis of the heterosis is not well understood. In Arabidopsis, crosses between different accessions produce hybrids with varied levels of heterosis relative to parental phenotypes in biomass. In all hybrids the advantages of the F1 hybrid is lost in the F2 for both phenotypic uniformity and yield gain. Success in generating F5/F6 Hybrid Mimic from the cross between C24 and Landsberg erecta (Ler) demonstrated that the large plant phenotype of the F1 hybrids can be stabilized. Hybrid Mimics selection was applied to Wassilewskija (Ws)/Ler and Col/Ler hybrids. The two hybrids showing different levels of heterosis. At 30 DAS, the Col/Ler hybrid generated Hybrid Mimics with rosette diameter and fresh weight equivalent to the F1 hybrid; Ws/Ler Hybrid Mimics outperformed the F1 hybrids in both the rosette size and biomass. Transcriptome analysis revealed up-regulation of cell wall biosynthesis and expansion genes could be a common pathway in increased size in Arabidopsis hybrids and Hybrid Mimics. Intercross of two independent Hybrid Mimic lines can further increase the biomass gain. Our results encourage the use of Hybrid Mimics for breeding and for investigating the molecular basis of heterosis.
