Project description:The cerebrovascular system plays pivotal roles in brain development and maintaining its functions. Despite several single-cell transcriptomic studies investigating the molecular atlas of brain vasculature, the comprehensive gene expression profile of the entire spectrum from blood-brain barrier (BBB) formation to maturation remains largely unexplored. In this study, we used transgenic zebrafish with endothelial-specific labeling, Tg(kdrl:eGFP), to perform single-cell RNA sequencing analysis on brain endothelial cells during BBB development. We identified the gene expression profiles of different subtypes of brain endothelial cells and found dynamic molecular programs as cerebrovascular maturation. Moreover, we found that the brain endothelial cells were vigorous in metabolism of energy and lipid at 3 dpf, while more activate in amino and lipid metabolism at 6 and 11 dpf. A comparative analysis of human and zebrafish scRNA-seq data indicated high conservation in brain endothelial cell subtypes and their biological functions during cerebrovascular development between the two species. Additionally, we unveiled the critical roles of zgc:158423 and slc16a1a during cerebrovascular development in zebrafish. Collectively, our findings elucidate gene expression profiles and metabolic characteristics of distinct endothelial subtypes, laying a foundation for understanding the molecular and metabolic landscape of BBB development in zebrafish.

Project description:One pending issue in cardiovascular epigenetics is whether cerebrovascular events, a major complication of atherosclerosis, are associated with any specific DNA methylation changes in the carotid plaque. To clarify that topic, we profiled the DNA methylomes of human symptomatic carotid plaques (SCPs) obtained from patients who suffered cerebrovascular events (n=19) and asymptomatic counterparts (ACP; n=19) with a high-density microarray (~485,000 CpG sites, Illumina), and crossed DNA methylation data with RNAseq-based expression data from an independent SCP set (n=8). Few (30) CpGs showed a significant (p<0.05; absolute Delta-Beta>0.20) differential methylation between the two groups. Within SCPs, methylation correlated significantly with post-cerebrovascular event time (PCET; range: 3-45 days; r-value range -0.926 to 0.857; p<0.05) for ~45,000 CpGs, the vast majority of which became hypomethylated with increasing PCET. Hypomethylation was not due to erasure of the gene-body and CG-poor region hypermethylation that accompany the progression of stable lesions, but rather targeted promoters and CpG islands. Noticeably, promoter hypomethylation and increased expression were observed for genes involved in the inhibition of the inflammatory response, defence against oxidative stress and active DNA demethylation. Our data show that only weak changes in the DNA methylome distinguish symptomatic from asymptomatic carotid plaques, but a widespread demethylation resulting in permissive transcriptional marks at atheroprotective gene promoters is established in plaques after a cerebrovascular event, thus mirroring previous observations that ruptured plaques tend to revert to a more stable structure. The identified loci are candidate targets to accelerate the pace of carotid plaque stabilization. DNA was quantified by Quant-iT PicoGreen dsDNA Reagent (Invitrogen) and the integrity was analyzed in a 1.3% agarose gel. Bisulfite conversion of 600 ng of each sample was perform according to the manufacturer's recommendation for Illumina Infinium Assay. Effective bisulphite conversion was checked for three controls that were converted simultaneously with the samples. 4 ul of bisulfite converted DNA were used to hybridize on Infinium HumanMethylation 450 BeadChip, following Illumina Infinium HD Methylation protocol. Chip analysis was performed using Illumina HiScan SQ fluorescent scanner. The intensities of the images are extracted using GenomeStudio (2011.2) Methylation module (1.8.5) software. Methylation score of each CpG is represented as beta value.

Project description:Atherosclerosis is characterized by the pathological accumulation of cholesterol-laden macrophages in the arterial wall. Atherosclerosis is also the main underlying cause of cardiovascular diseases (CVDs), and its development is largely driven by elevated plasma cholesterol. Strong epidemiological data find an inverse association between plasma β-carotene with atherosclerosis, and we recently showed that β-carotene oxygenase 1 (BCO1) activity, responsible for β-carotene cleavage to vitamin A, is associated with reduced plasma cholesterol in humans and mice. In this study, we explore whether intact β-carotene or vitamin A affect atherosclerosis progression in the atheroprone low-density lipoprotein receptor (LDLR) - deficient mice. In comparison to control-fed Ldlr-/- mice, β-carotene-supplemented mice showed reduced atherosclerotic lesion size at the level of the aortic root and plasma cholesterol levels (P = 0.0003). These changes were absent in Ldlr-/-/Bco1-/- mice, despite accumulating β-carotene in plasma and atherosclerotic lesions. We discarded the implication of myeloid BCO1 in the development of atherosclerosis by performing bone marrow transplant experiments. Lipid production assays found that retinoic acid, the active form of vitamin A, reduced the secretion of newly synthetized triglyceride and cholesteryl ester in cell culture and mice. Overall, our findings provide insights into the role of BCO1 activity and vitamin A in atherosclerosis progression through the regulation of hepatic lipid metabolism.

Project description:Atherosclerosis is an autoimmune disease characterized by lipid imbalances and chronic inflammation within blood vessels with limited preventive and treatment options currently available. Previous experiments have demonstrated the atheroprotective potential of collagen 6 subtype alpha6 (COL6A6) in apolipoprotein E-deficient (ApoE-/-) mice with hyperlipidemia. However, the mechanism underlying the anti-atherosclerotic effects of COL6A6 remains elusive. This knowledge gap was addressed in the present study by immunizing ApoE-/- mice with the Pep_A6 vaccine, comprising a COL6A6 peptide-KLH (keyhole limpet hemocyanin) conjugate and aluminum (Alum) hydroxide adjuvant, and conducting a series of experiments. Our objective was to investigate the efficacy of the Pep_A6 vaccine, focusing on immune responses and lipid metabolism. Our finding showed that the Pep_A6 vaccine represents a novel approach to combat atherosclerosis by inducing a large increase in Treg cells, the generation of antigen-specific antibodies and regulating lipid metabolism.

Project description:Diabetes mellitus (DM) is a chronic, noncommunicable disease, mainly due to insufficient insulin secretion or insulin sensitivity. Atherosclerosis is one of the major macrovascular complications of DM, and it is the main cause of death from clinical observation. Mongolian acupuncture therapy has the function of dredging meridians and regulating blood circulation, which has been proved to have unique efficacy and notable advantages in the treatment of DM and its common chronic complications. However, the effects and molecular mechanism of Mongolian acupuncture therapy in T2DM atherosclerosis remain unclear. The aim of this study is to explore the effect and molecular mechanism of streptozotocin and vitamin D3-induced T2DM atherosclerosis in rats, with or without Mongolian acupuncture therapy. The results of serum biochemical and histopathology analysis suggest that acupuncture therapy can improve cellular inflammatory factors, kidney function, serum lipid function and upper carotid artery tissue lesions in rat with T2DM atherosclerosis, which is an effective treatment method. In addition, we applied proteomics and metabolomics to comprehensively demonstrate that Mongolian acupuncture therapy in T2DM atherosclerosis mainly through regulation of lipid metabolism and amino acid metabolism. In general, our findings provide beneficial data support for future clinical studies of Mongolian acupuncture therapy in T2DM atherosclerosis.

Project description:Immune checkpoint inhibitor (ICI) therapies in cancer accelerate atherosclerosis progression. Here, we have charted the landscape of immune checkpoint gene expression and defined the network of disease-relevant interactions with single-cell resolution. We found that signaling through PD-1 and CTLA4 is driven by a population of dendritic cells enriched for FSCN1 that can be derived from peripheral blood cells following anti-PD-1 or -CTLA4 treatment ex vivo. Type 2 diabetes dampened plaque PD-1 and CTLA4 signaling, showing that cardiometabolic comorbidities elicit unique responses to ICIs. Lipid-lowering therapy equalized the intensity and direction of immune checkpoint interactions in human blood, while atherosclerotic mice subjected to a lipid-lowering diet displayed both increased co-inhibitory signaling and a downregulation of inflammatory transcriptional programs in plaques. Our findings underscore the potential of lipid-lowering therapies in stabilizing immune checkpoint interactions and reducing plaque inflammation, offering new insights on atherosclerosis and cardiovascular risks in cancer patients undergoing ICI treatments.
