Project description:We developed a multi-paratopic VEGF decoy receptor (Ate-Grab) by fusing the scFv of atezolizumab to VEGF-Grab to target PD-L1 expressing cells in tumor microenvironment. We compared the single-cell transcriptomes of gemcitabine-treated and Ate-Grab with Gemcitabine-treated Pan02 tumors. We confirmed the subtype of cancer-associated fibroblasts(CAFs) that modulates the collagen inside the tumor microenvrionment. In the Ate-Grab+Gemcitabine group, it was confirmed that new CAFs were regulated.

Oleuropein reprograms the immunosuppressive footprint of tumor-associated myeloid cells and enhances PD-L1/PD-1 blockade immunotherapy in cancer

Project description:Therapies based on PD-1/PD-L1 blockade fail in most cancer patients. Here we evaluated the capacities of oleuropein to reprogram tumor-associated immunosuppressive myeloid cells to increase the potency of immunotherapies. Oleuropein caused major global reprogramming of monocytic and granulocytic myeloid-derived suppressor cells and tumor-associated macrophages towards immunostimulatory subsets. Differential quantitative proteomics uncovered activated and down-modulated pathways at high resolution for each subset which regulated major differentiation programs. Oleuropein significantly potentiated the capacities of myeloid cells to activate T-cells and enhanced antitumor properties of PD-1 blockade, either by systemic anti-PD-1 antibody administration, or locally by intratumor antibody delivery with a self-amplifying RNA vector based on Semliki Forest virus. Combination therapies decreased tumor infiltration by immunosuppressive myeloid cells and increased dendritic cell recruitment within draining lymph nodes, leading to systemic antitumor T-cell responses. Potent therapeutic activities were evident in lung cancer models resistant to immunotherapies and in colon cancer models.

Expression data in PD-L1 positive and PD-L1 negative human melanoma samples

Project description:Blocking the PD-1/PD-L1 immunosuppressive pathway has shown promise in the treatment of certain cancers including melanoma. This study investigates differences in the gene expression profiles of human melanomas that do or do not display the immunosuppressive protein PD-L1. Further understanding of genes expressed within the tumor microenvironment of PD-L1+ tumors may lead to improved rationally designed treatments. Gene expression profiling was performed on total RNA extracted by laser capture microdissection from 11 archived formalin-fixed paraffin-embedded (FFPE) melanoma specimens, 5 of which were PD-L1 positive and 6 PD-L1 negative. Gene-expression profiles of PD-L1-affected CD8+ T cells

Project description:Programmed cell death 1 ligand 1 (PD-L1) is known to suppress immune system and to be an unfavorable prognostic factor in ovarian cancer. The purpose of this study was to elucidate the function of PD-L1 in peritoneal dissemination. Tumor cell lysis by CTLs was attenuated when PD-L1 on tumor cells was overexpressed and promoted when it was silenced. PD-L1 overexpression also inhibited gathering and degranulation of CTLs. Gene expression profile of mouse CTLs caused by PD-L1-overexpressing ovarian cancer was related to human CTLs exhaustion. In mouse ovarian cancer dissemination models, depleting PD-L1 expression on tumor cells resulted in inhibited tumor growth in the peritoneal cavity and prolonged survival. Restoring immune function by inhibiting immune-suppressive factors such as PD-L1 may be a promising therapeutic strategy for peritoneal dissemination. Genome-wide transcriptional changes in OT-1 mouse CD8+ T cells that were co-incubated with OVA peptide-loaded ID8 mouse ovarian cancer cell lines. CTLs from 4 mice were devided into 2 groups, and co-incubated with PD-L1-overexpressed ID8 or PD-L1-depleted ID8.

Dual Impacts of Serine/Glycine Free Diet in Enhancing Antitumor Immunity and Promoting Evasion via PD-L1 Lactylation [scTCR]

Project description:The effect of Serine/glycine free diet (-SG diet) on colorectal cancer (CRC) remains unclear, meanwhile PD-1 inhibitors are less effective for most CRC patients with proficient mismatch repair/microsatellite stable (pMMR/MSS) characteristics. Here, we demonstrate that -SG diet inhibits CRC growth and promotes the accumulation of cytotoxic T cells to enhance anti-tumor immunity. Additionally, we also identified the lactylation of PD-L1 in tumor cells as a mechanism of immune evasion during cytotocix-T-cell-mediated anti-tumor responses, and blocking PD-1/PD-L1 signaling pathway is able to rejuvenate the function of CD8+ T cells recruited by -SG diet, indicating the clinical potential of combining -SG diet with immunotherapy. Finally, we conducted a single-arm, phase I study, which suggested -SG diet could be a feasible and safe strategy to regulate systemic immunology. Collectively, our findings highlight the promising therapeutic future of -SG diet for treating solid tumors.

ERK and USP5 govern PD-1 homeostasis via deubiquitination to modulate tumor immunotherapy

Project description:The programmed cell death protein 1 (PD-1) is an inhibitory receptor on T cells and plays an important role in promoting cancer immune evasion. While ubiquitin E3 ligases for regulating PD-1 stability have been reported, deubiquitinase (DUB) governing PD-1 homeostasis to modulate tumor immunotherapy remains unknown. Here, we identify the ubiquitin-specific protease 5 (USP5) as a bona fide deubiquitinase for PD-1. Mechanistically, USP5 interacts with PD-1, leading to deubiquitination and stabilization of PD-1. Moreover, extracellular signal-regulated kinase (ERK) phosphorylates PD-1 at Thr234 and stabilizes PD-1 through promoting PD-1/USP5 interaction. Conditional knockout (cKO) of Usp5 in T cells significantly suppresses tumor growth in mice largely through decreasing PD-1 and increasing tumor-infiltrating cytotoxic CD8+ T cells. Furthermore, co-targeting USP5 and ERK signaling has an additive effect on reducing PD-1 and suppressing tumor growth in vivo. Together, this study uncovers the molecular mechanism for ERK/USP5 axis in regulating PD-1 stability and identifies potential combined therapeutic strategies for enhancing anti-tumor efficacy.
