Project description:Chinese Shitou goose is a type of large goose with high meat yield. Understanding the genetic regulation of muscle development in Shitou goose would be beneficial to improve the meat production traits of geese. Muscle development is regulated by genes related to myoblast proliferation and differentiation. In this study, the RNA-seq method was used to construct the mRNA and lncRNA expression profiles of Shitou goose myoblasts and myotubes. A total of 1664 differentially expressed (DE) mRNAs and 244 DE-lncRNAs were identified. The alternative mRNA splicing in proliferation and differentiation stages was also analyzed. Notably, pathways enriched in DE-mRNAs, DE-splicing transcripts, and DE-lncRNAs all point to the Wnt signaling pathway, indicating that the Wnt signaling is a key regulatory pathway of muscle development in Shitou goose. We also constructed the interactive network of DE-lncRNAs and DE-mRNAs and revealed some key genes of lncRNAs regulating the proliferation and differentiation of myoblasts. These results provide new insights for the study of the muscle development of the Shitou goose.

Project description:TNFα is a cytokine with multiple biological effects. It activates cascading signal pathways by binding to receptors on the cell membrane, regulating processes such as cell growth, differentiation, apoptosis, and inflammatory responses. The formation of skeletal muscle primarily relies on processes such as myoblasts proliferation, migration, differentiation, cytoskeletal rearrangement, and myotube fusion, which constitute a complex and coordinated process regulated by multiple genes. While research on TNFα has primarily focused on immunology, some studies suggest that TNFα also plays a physiological role in myogenesis, muscle repair, diseases, and atrophy.We isolated the nuclei (N) and cytoplasm (C) of flox and TNFα-CKO primary myoblasts in the early stages of differentiation and attempted to identify new targets in the TNFα signaling pathway.

Project description:The branched-chain amino acids (BCAA) are essential to animal growth and intramuscular fat deposition, and also metabolic health (circulating level elevated in obesity and diabetes). However, the molecular role and effect of valine on muscle growth and fat deposition were less explored. Valine supplementation significantly affected mouse muscle growth, increasing the abdominal fat but decreasing the back fat. In the leg muscle, the expression levels of genes related to autophagy and fat deposition were significantly disturbed. Furthermore, valine promotes lipid deposition in myoblasts in a dose-dependent manner, and co-treatment of valine and palmitic acid can act in concert to enhance lipid deposition in myoblast. Moreover, transcriptome sequencing on myoblasts revealed that the signaling pathways such as mTOR, PI3K-AKT and fatty acid metabolism were activated after valine treatment, and palmitic acid additionally stimulated signaling pathways related to lipid metabolism in myoblasts. Therefore, valine could independently activate the autophagy pathway, and the fatty acid metabolism pathway to enhance intramuscular fat deposition

Project description:We identified the differentially expressed miRNAs in Landes goose liver after overfeeding for 21 days using high-throughput sequencing. We obtained 21453493 and 21525819 clean reads in normal liver and fatty liver by high-throughput sequencing, respectively. Of these clean reads, we respectively gained 9244896 and 9847086 miRNAs sequences in two groups by filtering the known non-miRNA reads, such as rRNA, tRNA, snRNA, and snoRNA by screening against ncRNA deposited in the GenBank and Rfam databases. These findings provided insights into the expression profiles of miRNAs in goose liver, and deepened our understanding of miRNAs in hepatic steatosis of geese.

Project description:We previously identified C/EBPβ as an inhibitor of myogenic differentiation and a regulator of muscle satellite cell self-renewal. We found that C/EBPβ is regulated during the transition from proliferation to growth arrest in myoblasts and overexpression of C/EBPβ in myoblasts promotes cell growth arrest in vitro and improves engraftment of satellite cells into the stem cell niche in vivo. To identify the molecular mechanism by which C/EBPβ regulates myoblast proliferation and growth arrest, we performed RNA-seq on C/EBPβ-overexpressing myoblasts.

Project description:Polycystic Kidney Disease is characterized by the formation of large fluid-filled cysts that eventually destroy the renal parenchyma leading to end-stage renal failure. Although remarkable progress has been made in understanding the pathologic mechanism of the disease, the precise orchestration of the early events leading to cyst formation is still unclear. Abnormal cellular proliferation was traditionally considered to be one of the primary irregularities leading to cyst initiation and growth. Consequently, many therapeutic interventions have focused on targeting this abnormal proliferation, and some have even progressed to clinical trials. However, the role of proliferation in cyst development was primarily examined at stages where cysts are already visible in the kidneys and therefore at later stages of disease development. In this study we focused on the cystic phenotype since birth in an attempt to clarify the temporal contribution of cellular proliferation in cyst development. Using a PKD2 transgenic rat model (PKD2 (1-703)) of different ages (0-60 days after birth) we performed gene expression profiling and phenotype analysis by measuring various kidney parameters. Phenotype analysis demonstrated that renal cysts appear immediately after birth in the PKD2 transgenic rat model (PKD2 (1-703)). On the other hand, abnormal proliferation occurs at later stages of the disease as identified by gene expression profiling. Interestingly, other pathways appear to be deregulated at early stages of the disease in this PKD model. Our data suggest that cystogenesis precedes deregulation of proliferation-related pathways, suggesting that proliferation abnormalities may contribute in cyst growth rather than cyst formation. In this study we focused on the cystic phenotype since birth in an attempt to clarify the temporal contribution of cellular proliferation in cyst development. Project description:Regeneration is one of the key factors affecting downy feather production. However, the signals and molecular controlling the progression of feather regeneration was poorly understood. We used a high density microarray to profile the temporal transcriptome dynamics during the regeneration periods. A total of 3540 genes expressed differentially with significant fold changes during feather regeneration. Cluster analysis revealed event specific dynamic expression of genes related to cell cycling, cell migration, cell proliferation, follicle re-growth and fiber elongation. These clusters involved functional clusters like regulation of actin cytoskeleton, focal adhesion and sphingolipid metabolism, and enriched signaling pathways like Wnt signaling pathway, MAPK pathway and TGFβ signaling pathway. In cell cycling, mainly involved genes were cyclin B3, cyclin Y. In TGFβ signaling pathway, mainly involved genes were TGFβ3, BMP7, NOG and BMP2. While Wnt5a, Wnt10a, FZD2, and FZD10 were involved in canonical Wnt/β-catenin pathway. Our study provides a comprehensive genetic blue print of diverse cellular responses during regeneraton of goose feather. The data provide an initial step towards a better understanding of molecular mechanisms underlying feather regeneration process and also suggest that signals or molecular participated in feather regeneration was quite different from the generation of hair in mammals.
