Transcripts enriched in microsome fraction in ribosome-independent manner

Project description:To identify the transcripts fractionated into microsome fraction in ribosome-independent manner, we isolate rough microsome fraction by sucrose density gradient ultracengrifugation, then the rough microsome fraction is centrifugated following treatment with puromycine and EDTA in high-salt buffer to remove ribosomes. The pellet and surpernatant are named naked microsome fraction (NM) and stripped ribosome fraction (SR), respectively. By calculating the ratio of the level of each mRNA in NM and SR, we identify the enriched transcripts in NM.

Transcript sequencing of mononuclear fractions from dissociated murine skeletal muscle

Project description:The important role of mesenchymal progenitors is quickly emerging in various biological processes. Thus far an appropriate marker has been lacking. Here the fractionation of dissociated skeletal muscle and subsequent enrichment of MPs by an established method was employed. Endothelial and blood derived cells comprised the lineage positive fraction and the remaining cell suspension was fractionated using the surface marker Sca1(Ly6a) to enrich for MPs. RNA sequencing was performed on all fractions and subsequent transcriptomic data was interrogated for MP specific transcription factors. Hic1 was found to be highly enriched in the MP fraction.

Project description:Transcriptome analysis of dentritic cells from the spleen (CD11c+) or MLN (CD11c+CD103+) of mice with DC-specific deletion of TGFBR2 gene, or their control littermates. TRGFR2 gene was deleted in dendritic cells using Cre/lox approach. Mice with this deletion develop spontaneous multi-organ autoimmune inflammation and die by 15 weeks of age. Splenic CD11c+ Dc were isolated by magetic cell sorting. MLN CD11c+CD103+ DC were flow sorted.
