Project description:We tested the preclinical efficacy of two small molecule RAGE inhibitors (TTP488 (Azeliragon) and FPS-ZM1) against TNBC metastasis in the orthotopic xenograft MDA-MB-231/4175 - NSG model. Both TTP488 and FPS-ZM1 impaired TNBC metastasis in this lung metastatic breast cancer model, with TTP488 affecting metastasis to a greater degree than FPS-ZM1. Transcriptomic analysis of the primary tumors revealed that TTP488 and FPS-ZM1 displayed high concordance in gene expression changes affecting metastatic pathways.

Project description:Epithelial-Mesenchymal plasticity plays a fundamental role both in embryogenesis and in tumor cell dissemination during tumor progression. The receptor for advanced glycation end products (RAGE) is a driver of cell plasticity in fibrotic diseases; however, its role and molecular mechanism in triple-negative breast cancer (TNBC) remains unclear. Here, we demonstrate that RAGE signaling maintains the mesenchymal phenotype of aggressive TNBC cells by enforcing the expression of SNAIL. We uncover a crosstalk mechanism between the TGF-β and RAGE pathways that is required for the acquisition of mesenchymal traits in TNBC cells. Consistently, RAGE inhibition elicits epithelial features that block migration and invasion capacities. Since RAGE is a sensor of the tumor microenvironment (TME) and the highly glycolytic rate of tumors induces an acidic TME, we modeled acute acidosis in TNBC cells and showed it promotes enhanced production of RAGE ligands and the activation of RAGE-dependent invasive properties. Furthermore, acute acidosis increases SNAIL levels and tumor cell invasion in a RAGE-dependent manner. Finally, we demonstrate that in vivo inhibition of RAGE reduces metastasis incidence and expands survival, consistent with molecular effects that support the relevance of RAGE signaling in EMT plasticity. These results uncover new molecular insights on the regulation of EMT phenotypes in cancer metastasis and provide rationale for pharmacological intervention of this signaling axis.

Project description:Cell surface heparan sulfate (HS) plays an essential role in RAGE signaling by inducing RAGE oligomerization. To understand the physiological significance of HS-induced RAGE oligomerization in vivo, we generated RAGE knock-in mice (RageAHA/AHA) by introducing point mutations to specifically disrupt HS–RAGE interaction. The RAGE variant expressed by RageAHA/AHA mice demonstrated normal ligand-binding but greatly impaired capacity of HS-binding and oligomerization. To grasp the full scale of the alteration in gene expression caused by knocking out Rage, we performed a RNAseq analysis of mature neutrophils and lung from WT, RageAHA/AHA and Rage-/- mice. The overall number of differently regulated genes (DEGs) in Rage-/- neutrophils were almost 2.5 times higher than in RageAHA/AHA neutrophils (603 vs. 247). In contrast, the number of DEGs were much less in lungs compared to neutrophils in both strains (202 DEGs in Rage-/- lungs and merely 31 DEGs in RageAHA/AHA lungs), however the difference between the two genotypes was even more dramatic in lungs (7-fold) than we observed in neutrophils (2.5-fold). By comparing transcriptomes of neutrophils and lung tissues from RageAHA/AHA and Rage-/- mice, we present clear evidence that complete deficiency of RAGE had much broader impact on global gene expression compared to point mutations of RAGE.

Project description:Purpose: The BET family protein BRD4 is an important anti-tumor target and is highly expressed in breast cancer. However, BET inhibitors are susceptible to drug resistance. The aim of this study is to explore the mechanism of BET inhibitor JQ-1 combined with cardamonin in the anti-invasion and metastasis of triple-negative breast cancer. Methods: triple-negative breast cancer cells mRNA of DMSO (Control), JQ-1,cardamonin and combined groups were progressed with deep sequencing, in triplicate through Illumina sequencing platform (HiSeqTM 2500). Genes with adjusted P-value < 0.01 and |foldChange| > 2 were defined as differentially expressed genes (DEGs) and subjected to the following GO and KEGG enrichment. qRT-PCR was performed to validate several essential genes using QuantStudio 5 (Thermo Fisher) and SYBR Green assays. Results: As an inhibitor of mTOR, Cardamonin(CAR) can inhibit the proliferation of tumors. Research have found that BET inhibitor can affect the function of a large number of genes by regulating the epigenome, which plays an important role in a variety of diseases. In this study we explored the JQ1 which is a BRD4 protein inhibitor can inhibit the proliferation and metastasis of MDA-MB-231 cells in triple-negative breast cancer cell lines, and this phenomenon can be significantly enhanced when CAR is combined with JQ-1, and the same results are shown in mouse tumor formation experiments and triple-negative breast cancer organoids. At the same time, RNA-seq was performed in MD1-MB-231 cells to explore the changes of specific molecular signaling pathways. We have mapped over 40 million sequence reads to the MDA-MB-231 cell line genome in each specimen.Compared with the control group, CAR and JQ1, the combination group has significantly enriched gene expression. KEGG and GO enrichment analysis indicate that the combination group showed a series of biological processes such as macroautophagy, negative regulation of cell growth, lipid catabolic process, which led to decreased cell viability and decreased proliferation and metastasis ability. Conclusions: In this study, we demonstrated that JQ-1 combined with cardamomin could inhibit the invasion and metastasis of triple-negative breast cancer MDA-MB-231 cells, induce cell cycle arrest and promote cell apoptosis. The results of RNA-Seq were consistent with our in vitro and in vivo experiments, which demonstrated that the combination of JQ-1 and cardamonin effectively inhibited the proliferation, invasion and metastasis of MDA-MB-231 cells.

Our published study showed that PAI-1 could induce the migration and metastasis of TNBC cells. However, the underlying mechanism by which PAI-1 regulates TNBC metastasis has not been addressed. Using microarray analysis of lncRNA expression profiles, we identified a lncRNA SOX2-OT, which is by induced by PAI-1 and could function as an oncogenic lncRNA in TNBC.

Project description:Tumor-initiating cells with reprogramming plasticity and/or de-differentiation attributes have been thought to initiate primary tumor development as well as to regenerate secondary tumors in metastatic organs; however, the molecular mechanisms are not fully understood. We previously found that breast tumor-initiating cell marker, CD44, directs multicellular aggregation and cluster formation of circulating tumor cells (CTCs), which further enhance stemness and survival of such cells, enabling metastatic colonization to the lungs. To further elucidate the molecular network underlying CTC cluster formation, we performed global proteomic profiling and discovered that the tetraspanin protein CD81, which is normally enriched in exosomes (small extracellular vesicles), is a new driver of cancer initiation and metastasis as a facilitator and target of CD44. Loss of CD81 compromises tumorigenicity and mammosphere formation of triple negative breast cancer (TNBC) cells. Assisted by machine learning-based algorithms and mutagenesis approach, we found that CD81 interacts with CD44 on the cellular membrane through their extracellular regions. Notably, genetic knockout of CD44 or CD81 results in loss of both CD81 and CD44 in secreted exosomes, a state which abolishes exosome-induced self-renewal of recipient cells, such as mammosphere formation. In addition, RNA sequencing analysis showed that CD81 knockdown up-regulates expression of a cell differentiation marker, SEMA7a, whose down-regulation partially rescues mammosphere formation inhibition by CD81 depletion. Clinically, CD81 expression was observed in >80% of CTCs and specifically enriched and co-expressed along with CD44 in clustered CTCs of breast cancer patients. Mimicing the phenotypes of CD44 deficiency, loss of CD81 also inhibited tumor cell aggregation and lung metastasis of TNBC in both human and mouse tumor models, supporting the clinical significance of CD81 in association with patient outcomes. Our study highlights a new driving role of CD81 in cancer exosome-induced stemness, clustered CTCs, and metastasis initiation of TNBC, reported for the first time to our knowledge.
