Project description:In order to identify gene-expression patterns in mesenchymal stem cells associated with different birth weights and intrauterine growth parameters, we conducted comparative microarray gene expression experiments prior and post acute insulin stimulation Mesenchymal stem cells (MSC) were extracted from the Wharton's jelly of the umbilical cord of normal and SGA babies. The MSCs were expended in culture. The MSCs were subjected to acute insulin treatment for various time points. The RNA of these cells were collected for gene-expression study.

Proteomic Analysis of Estrogen Mediated Enhancement of MSC-Induced Angiogenesis in vivo

Project description:Therapeutic use of mesangial stem cells (MSCs) for tissue repair, including heart, has great potential. MSCs from multiple sources, including those derived from human umbilical matrix, namely Wharton's jelly, may serve as a resourceful candidate. Even after more than a decade, low engraftment efficacy of MSCs, in vivo, remains a limitation which requires in-depth understanding of the mechanisms and factors involved to facilitate a better and clinically relevant outcome. We recently hypothesized and demonstrated that the sex hormone – 17-beta estradiol (E2) – facilitates protective processes within the cardiovascular system by modulating MSC function, in vitro. Here, using a proteomic approach, we investigated the angiogenic potential of MSCs in vivo and the modulatory actions of E2 on mechanisms involved in tissue repair (potentially post-ischemic injury). Employing an in vivo approach with an ovariectomized (OVX) mice model, we evaluated the effects of E2 on MSC-induced angiogenesis using Matrigel plug assay.

MSCs found in the Wharton's jelly of the human umbilical cord are easily obtained and are capable of transplantation without rejection. We isolated MSCs from Wharton’s jelly and bone marrow (WJ-MSCs and BM-MSCs, respectively) and compared their transcriptional profiles.

The cells were cultured under a hypoxic condition (1%O2). Results: 1169 miRNAs were identified in original BCC and WJ-EV treated BCC (pBCC), respectively.

In this study, BM-MSCs and WJ-MSCs were isolated from 4 and 3 independent donors, respectively.
