Project description:Strategies that promote functional organ growth with minimal adverse effects are the ultimate goal of regenerative medicine but no single approach is currently available for organ level repair. Here, using an evolutionary adapted in vivo infection model - Mycobacterium leprae, with host cell reprogramming ability and its natural animal host, the nine-banded armadillo (Dasypus novemcinctus) that harbor bacteria in the highly regenerative liver - we present an in vivo model for promoting adult liver growth at organ level without adverse effects. Experimentally infected armadillos harboring bacteria in the liver, but not infection-resistant or drug-treated animals, showed a significantly increased total liver: body weight ratio, indicative of bacterial-driven liver organ growth in living animals. The machine-learning approach revealed an increase in healthy liver lobule number with a proportionate expansion of the hepatocyte mass with integrating vasculature and biliary networks responsible for functional liver growth. Intriguingly, infected enlarged livers show intact microarchitecture but without evidence of hepatocellular damage, fibrosis/scarring or tumorigenesis. Reactivation of armadillo liver progenitor and developmental genes/proteins, as well as upregulation of growth-, metabolism- and differentiation-associated markers with minimal change in oncogenes or tumor suppressor genes, suggests that bacteria have adapted dynamic regenerative, homeostasis and reprogramming mechanisms to promote de novo organogenesis while maintaining tissue integrity and tumor preventive strategies for host-dependent bacterial propagation. Thus, our model may facilitate the unravelling of in vivo endogenous regenerative pathways that effectively re-engage liver organ growth, with broad implications.

Project description:Fibrosis, characterized by the abnormal deposition of extracellular matrix, can cause organ dysfunction and mortality. However, research on the mechanisms of fibrosis, especially that focusing on the identification of drug targets, is currently still lacking. This study is the first to reveal that a small molecule (YTB-8) derived from the natural β-carboline alkaloid flazin has potent protective effects against renal fibrosis in vitro and in vivo. The cellular target of YTB-8, a nuclear and endoplasmic reticulum membrane protein known as transmembrane protein 43 (TMEM43), was identified by activity-based protein profiling (ABPP) strategy in a renal fibrosis model. YTB-8 was found to interact with amino acids Arg312 and Leu382 of TMEM43 and exert anti-renal fibrosis effects by suppressing the NF-κB and TGF-β/Smad signaling pathways. Utilization of simulated TMEM43 and its pocket structure for virtual screening, followed by in vitro and in vivo detection, led to us identifying two FDA-approved drugs, drospirenone and adapalene, with anti-renal fibrosis efficacy. TMEM43 was also found to be the target of YTB-8 in its alleviation of lung, liver, and skin fibrosis in vitro, verifying the value of TMEM43 as a general target for new-generation drug design relating to organ fibrosis. Collectively, these findings revealed TMEM43 to be a promising universal cellular target for organ fibrosis intervention.

Although fibroblasts are typically made responsible, it was recently suggested that endothelial cells contribute to fibrosis, although the functional importance of this phenomenon remained unclear. In this study, we addressed this question by genetic manipulation of the fibrogenic HMG-box transcription factor SOX9 specifically in vascular endothelial cells. Induced transgenic overexpression of SOX9 in these cells in mice triggered extensive fibrosis, organ growth and dysfunction in the heart, lung, liver and spleen. We found an upregulation of endogenous Sox9 in endothelial cells during cardiac, lung and liver fibrosis, and endothelial specific Sox9 deletion prevented fibrosis and organ dysfunction in mouse models of systolic and diastolic heart failure, as well as in mouse bleomycin induced pulmonary and non-alcoholic steato-hepatitis (NASH) triggered liver fibrosis. Mechanistically, a combined approach of bulk and single cell RNA sequencing of endothelial cells across multiple vascular beds revealed that Sox9 promoted the expression of extracellular matrix and matrix remodelling genes, growth factors, and inflammatory mediators, which triggered the expression of extracellular matrix directly by endothelial cells, but also activated fibroblasts. As we found the upregulation of endothelial Sox9 also in failing human hearts, we propose that endothelial Sox9 could be a target for anti-fibrotic therapy.

Project description:Fibrosis is a hallmark of chronic disease. Although fibroblasts are known to be involved, it is unclear to what extent endothelial cells might contribute. We detected increased expression of the transcription factor Sox9 in endothelial cells of fibrotic mouse organs in models of systolic heart failure by pressure overload, diastolic heart failure by high-fat diet and L-Name administration, pulmonary fibrosis after bleomycin treatment, and liver fibrosis as consequence of CDAA diet. We also observed endothelial SOX9 upregulation in human heart tissue in patients with heart failure. To test whether this SOX9 induction was sufficient to cause disease, we generated mice with endothelial cell-specific overexpression of Sox9, which indeed triggered extensive fibrosis in multiple organs and promoted signs of heart failure. Endothelial Sox9 deletion, in turn, prevented fibrosis and organ dysfunction in pre-clinical surgical and pharmacological mouse models of heart failure as well as of lung, and liver injury. Bulk and single cell RNA sequencing of endothelial cells across multiple vascular beds revealed that SOX9 induced extracellular matrix, growth factor, and inflammatory genes leading to matrix deposition by endothelial cells. Moreover, in part through triggering an increased expression of the secreted growth factor Ccn2 as direct SOX9 target, endothelial cells activated neighboring fibroblasts to migrate and deposit matrix in response to SOX9. Endothelial Sox9 deletion reversed these changes, suggesting a role for endothelial SOX9 as fibrosis-promoting transcription factor across organs in response to disease stimuli. Therefore, endothelial cells could be a promising target to counteract fibrotic heart, liver and lung disease
