Single-nucleus transcriptomics atlas of middle and late prenatal human cortical development

Project description:Late prenatal development of the human neocortex encompasses a critical period of gliogenesis and cortical expansion. However, systematic single-cell analyses to resolve cellular diversity and gliogenic lineages of the third trimester are lacking. Here, we present a comprehensive single-nucleus RNA sequencing atlas derived from the proliferative germinal matrix and laminating cortical plate of 15 prenatal, non-pathological postmortem samples and 3 adult controls. This dataset captures prenatal gliogenesis with high temporal resolution and is provided as a resource for further interrogation. Our computational analysis resolves greater complexity of glial progenitors, including transient glial intermediate progenitor cell (gIPC) and nascent astrocyte populations in the third trimester of human gestation. We use lineage trajectory and RNA velocity inference to further characterize specific gIPC subpopulations preceding both oligodendrocyte (gIPC-O) and astrocyte (gIPC-A) lineage differentiation. We infer unique transcriptional drivers and biological pathways associated with each developmental state, validate gIPC-A and gIPC-O presence within the human germinal matrix and cortical plate in situ, and demonstrate gIPC states being recapitulated across adult and pediatric glioblastoma tumors.

Definition of cortical somatosensory maps is fine-tuned by prenatal thalamic spontaneous activity

Project description:The brain's sensory representation relies on precise mapping of peripheral inputs onto cortical areas. In the mouse primary somatosensory cortex, prominent mystacial whiskers are represented in large, conspicuous barrels, while the smaller upper lip whiskers are represented by numerous smaller barrels. Traditionally, barrel area size is believed to be determined postnatally by whisker input and receptor density. However, prenatal thalamic spontaneous activity can influence cortical barrel size independently of whisker input. Thus, the developmental mechanisms defining distinct barrel field territories, including size and map definition, remain poorly understood. Here, we show that prenatal ablation of mystacial whiskers enhances the functional and anatomical definition of upper lip whisker barrels without affecting receptors density. These changes result from prenatal alterations in thalamic spontaneous activity patterns involving specific transcriptional modifications, highlighting the significance of prenatal synchronized neuronal activity in shaping cortical barrel size and functional map definition in the developing somatosensory system.

Middle and Late Pleistocene Denisovan subsistence at Baishiya Karst Cave on the Tibetan Plateau

Project description:Genetic and limited palaeoanthropological data suggest that Denisovans, a sister group to Neanderthals, were once widely distributed in eastern Eurasia, likely stretching from high-latitude Siberia, to the high-altitude Tibetan Plateau, to the low-latitude subtropical regions of southeast Asia. This suggests that Denisovans were capable of adapting to a highly diverse range of environments, but archaeological evidence for this is currently limited. As a result, we know little about their behaviours, including subsistence strategies, across the vast areas they likely occupied. Here, we describe the late Middle to Late Pleistocene faunal assemblage from Baishiya Karst Cave on the Tibetan Plateau, where the Xiahe Denisovan mandible and Denisovan sedimentary mtDNA were found, by integrating proteomic screening into traditional zooarchaeological analysis. The results indicate that the faunal assemblage consists of a diverse range of animals, including megafauna, large mammals, small mammals and birds, but is dominated by medium-sized herbivores. Frequent cut marks and percussion traces on bone surfaces throughout the assemblage, even on carnivore bones, indicate that Denisovan activities in Baishiya Karst Cave from at least 190 to 30 thousand years are responsible for the fauna assemblage accumulation. Thorough utilization of acquired animal resources, even perhaps the fur, too, might have helped Denisovans to survive through the last two glacial-interglacial cycles on the cold high-altitude Tibetan Plateau. Our results shed new light on Denisovan behaviours and their adaptations to the diverse and fluctuated environments in the Middle and Late Pleistocene eastern Eurasia.
