Project description:In this manucript, we investigate the effect of different crosslinking density of the thiolated hyalonric acid hydrogel as the supporting matrix for monolayer expanded chondrocyte's chondrogenic redifferentiation.A wide ragne of gene expression level was found to be correlated with teh level of crosslinking, and the biological processes and KEGG pathways associated with this panel of genes are related to cell death, immune response, and regulation of various foundamental developmental processes.

Transcription profiling of Bos taurus hypertrophic growth plate chondrocytes vs. reserve zone chondrocytes

Project description:Growth plate chondrocytes were isolated from the distal metacarpus of young dairy cattle (all under 10 mo of age), the chondrocytes were released from the extracellular matrix by digestion with Collagenase P for 4 hours, and the various zones of the growth plate were separated by density centrifugation. The least-dense Hypertrophic Zone (HZ) cells were compared to the most-dense Reserve Zone (RZ) cells. 6 pairs of HZ vs RZ were compared by microarray. Expression profiles from chondrocytes in culture - time course

Project description:To characterize the process of chondrocyte differentiation and maturation, we used Affymetrix GeneChip analysis to profile the dynamic changes in mRNA expression in high density cultures of primary rib chondrocytes from newborn mice. Temporal expression profiles were obtained from RNA harvested from chondrocytes cultured for 2, 6, 9, 12 and 15 days. Progressive maturation of cells into cartilage-producing hypertrophic chondrocytes was accompanied by changes of expression for many genes, particularly genes encoding extracellular matrix components and transcription factors.

Real-time quantitative PCR analysis of UPR genes in human osteoarthritic chondrocytes specimens treated with thapsigargin compared to the untreated (control) chondrocytes

Project description:Human osteoarthritic (OA) chondrocytes were obtained from the cartilage of femoral condyles and tibial plateaus from three patients undergoing total knee arthroplasty. The chondrocytes were released from the cartilage, seeded at high density, and cultured in Dulbecco’s modified Eagle’s medium supplemented with 10% heat-inactivated fetal bovine serum and an antibiotic mixture (100 units/ml penicillin base and 100 μg/ml streptomycin base) at 37°C in a humidified atmosphere of 5% CO2/95% air. The chondrocytes from each of the three donors were left untreated (control) and treated with thapsigargin (50nM for 20hours).Total RNA was extracted from the chondrocytes and processed for qPCR according to the manufacturer's instructions (SA Biosciences RT2 Profiler PCR Array PAHS-089Z).

Interplay between calcium cycling and sarcomeres directs cardiomyocyte redifferentiation and maturation during regeneration

Project description:Zebrafish hearts can regenerate by replacing damaged tissue with new cardiomyocytes. Although the steps leading up to the proliferation of surviving cardiomyocytes have been extensively studied, little is known about the mechanisms that control proliferation and redifferentiation to a mature state. We found that the cardiac dyad, a structure that regulates calcium handling and excitation-contraction coupling, played a key role in the redifferentiation process. A component of the cardiac dyad called leucine-rich repeat–containing 10 (Lrrc10) acted as a negative regulator of proliferation, prevented cardiomegaly, and induced redifferentiation. We found that its function was conserved in mammalian cardiomyocytes. This study highlights the importance of the underlying mechanisms required for heart regeneration and their application to the generation of fully functional cardiomyocytes.
