Project description:To cope with limiting phosphorus (P) availability, plants have evolved a series of mechanisms to recycle internal P sources and to acquire P from the soil. One of these mechanisms is the release of low-molecular weight carboxylates, such as malate, which helps to liberate phosphate desorbed to aluminum and iron oxides. As malate release into the rhizosphere and root apopplast also increases iron availability. To identity genes involved in this interaction, we investigated time-dependent changes in the transcriptome of Arabidopsis thaliana roots exposed to sufficient and deficient phoshate levels .

Gene expression data from arbuscular mycorrhizal and phosphate-treated Medicago truncatula roots.

Project description:Arbuscular mycorrhiza (AM) interactions between plants and Glomeromycota fungi primarily support phosphate aquisition of most terrestrial plant species. To unravel gene expression in Medicago truncatula root colonization by AM fungi, we used genome-wide transcriptome profiling based on whole mycorrhizal roots. We used GeneChips to detail the global programme of gene expression in response to colonization by arbuscular mycorrhizal fungi and in response to a treatment with phosphate and identified genes differentially expressed during this process. Medicago truncatula roots were harvested at 28 days post inoculation with the two different arbuscular mycorrhizal fungi Glomus intraradices (Gi-Myc) and Glomus mosseae (Gm-Myc) under low phosphate conditions (20 µM phosphate) or after a 28 days treatment with 2 mM phosphate in the absence of arbuscular mycorrhizal fungi (2mM-P). As a control, uninfected roots grown under low phosphate conditions (20 µM phosphate) were used (20miM-P). Three biological replicates consisting of pools of five roots were used for RNA extraction and hybridization on Affymetrix GeneChips.

Cellular Patterning of <i>Arabidopsis</i> Roots Under Low Phosphate Conditions.

Project description:Phosphorus is a crucial macronutrient for plants playing a critical role in many cellular signaling and energy cycling processes. In light of this, phosphorus acquisition efficiency is an important target trait for crop improvement, but it also provides an ecological adaptation for growth of plants in low nutrient environments. Increased root hair density has been shown to improve phosphorus uptake and plant health in a number of species. In several plant families, including Brassicaceae, root hair bearing cells are positioned on the epidermis according to their position in relation to cortex cells, with hair cells positioned in the cleft between two underlying cortex cells. Thus the number of cortex cells determines the number of epidermal cells in the root hair position. Previous research has associated phosphorus-limiting conditions with an increase in the number of cortex cell files in Arabidopsis thaliana roots, but they have not investigated the spatial or temporal domains in which these extra divisions occur or explored the consequences this has had on root hair formation. In this study, we use 3D reconstructions of root meristems to demonstrate that the radial anticlinal cell divisions seen under low phosphate are exclusive to the cortex. When grown on media containing replete levels of phosphorous, A. thaliana plants almost invariably show eight cortex cells; however when grown in phosphate limited conditions, seedlings develop up to 16 cortex cells (with 10-14 being the most typical). This results in a significant increase in the number of epidermal cells at hair forming positions. These radial anticlinal divisions occur within the initial cells and can be seen within 24 h of transfer of plants to low phosphorous conditions. We show that these changes in the underlying cortical cells feed into epidermal patterning by altering the regular spacing of root hairs.

Vascular transcription factors control epidermal responses to limiting phosphate conditions

Project description:Optimal plant growth is hampered by limiting amounts of the essential macronutrient phosphate in most soils. As a response, plant roots produce root hairs to capture this immobile nutrient. Although vital to high-yielding crops, this response remains poorly understood. By generating and exploiting a high spatial and temporal resolution single cell atlas of the Arabidopsis root, we show a remarkable enrichment of TMO5/LHW target genes in root hair epidermal cells. Moreover, these vascular bHLH factors are sufficient to induce a root hair formation response in the epidermal cells, similar as observed during low phosphate conditions, via the downstream cytokinin signaling. We further show that root hair formation under low phosphate conditions is almost absent when TMO5 function or cytokinin signaling is perturbed. In conclusion, cytokinin signaling links the adaptation of roots under low phosphate conditions in the epidermis to perception in vascular cells.

Under phosphate-limiting conditions plants activate genetic, biochemical and morphological modifications to cope with phosphate starvation. One of the morphological modifications that plants induce under phosphate limitation is the arrest of primary root growth and it is induced by the root tip contact with low phosphate media. The sensitive to proton rhizotoxicity (stop1) and aluminium activate malate transporter 1 (almt1) mutants of Arabidopsis thaliana continue primary root growth under in vitro Pi-limiting conditions, thus, to get insight into the molecular components that control primary root growth inhibition under low phosphate conditions we extracted and sequenced mRNA from the root tips (2-3 mm from the root apex) of wild-type plants (Col-0 accession) and low-phosphate-insensitive mutants almt1 and stop1 grown under low and high phosphate conditions 5 days after germination using an RNA-seq methodology.
