Project description:We identify and validate 11,039 circular RNAs with high confidence in dopamine and pyramidal neurons --- the preferentially vulnerable prototypical neurons in Parkinson’s and Alzheimer’s --- in 190 human brains using laser-capture RNA sequencing that combines ultra-deep sequencing of polyadenylated and non-polyadenylated RNA with cell-type enrichment afforded by light microscopy.

MiR-137 and GOMAFU form a novel non-coding RNA pathway to regulate human neuron differentiation and molecular networks affected in neuropsychiatric diseases

Project description:Increasing evidence suggests important roles of long noncoding RNAs (lncRNAs) in normal neuron development and neuropsychiatric disorders that disturb early neurodevelopmental processes. One such lncRNA is GOMAFU, which displays aberrant expression in schizophrenia postmortem brains and is known to regulate SCZ-associated splice variants in developing human neurons. However, molecular mechanisms that control expression and biological function of GOMAFU in human neuron development remain elusive. Here we report the identification of a novel miRNA-lncRNA pathway in which miR-137, a well-recognized risk factor for SCZ and autism spectrum disorder (ASD) that play key roles in promoting neuronal differentiation, enhances GOMAFU expression in human neurons through targeting transcription factors that suppress GOMAFU. In addition, loss of GOMAFU in cortical neurons derived from human induced pluripotent stem cells (hiPSCs) by CRISPR-cas9 mediated gene editing significantly reduces dendritic growth during early-stage differentiation. Moreover, GOMAFU deficiency elicited a broad influence on molecular pathways commonly affected in SCZ and ASD postmortem brains, including aberrantly increased expression of numerous genes known to govern neuroimmune responses. Our studies demonstrated essential roles of the lncRNA GOMAFU in human neuron development and connected GOMAFU malfunction with pathological molecular networks in neuropsychiatric disorders.

Project description:A complex train of events unfolds during brain development, guided by activation/repression of gene expression programs. It remains unclear how this process is disrupted in disease. Here we integrate human genetics with transcriptomic data from differentiation of human embryonic stem cells into cortical excitatory neurons. This reveals a cascade of transcriptional programs activated during early corticoneurogenesis in vitro and in vivo. Genetic variation in these programs is robustly associated with neuropsychiatric disorders and cognitive function, with variants concentrated in loss-of-function intolerant genes. Neurogenic programs also capture schizophrenia GWAS enrichment previously identified in mature excitatory neurons, suggesting that pathways activated during pre-natal development underlie disease-relevant deficits in mature neuronal function. Down-regulation of these programs in DLG2-/- lines delays expression of cell-type identity and impairs neuronal migration, morphology and action potential generation, validating predicted deficits. These data implicate specific cellular pathways underlying neurodevelopment in the aetiology of multiple neuropsychiatric disorders and cognition.

It remains unclear how this process is disrupted in neuropsychiatric disorders. Here we integrate human genetics with transcriptomic data from the differentiation of human embryonic stem cells into cortical excitatory neurons.
