Project description:Bovine Immunodeficiency Virus (BIV) is a member of Retroviruses family which is natural pathogen of cattle, prevalent worldwide. The significance of BIV infection in cattle has not been clearly established and it is still unknown whether BIV induces a specific syndrome or whether it renders animals more susceptible to other infections. To gain insight into host response to BIV infection the pattern of gene expression in bovine macrophage cells (BoMac) was analyzed using BLO Plus microarrays from CAFG, Michigan State University ( GEO GPL9176).

Project description:IntroductionBovine immunodeficiency virus (BIV) is found worldwide in cattle under natural conditions. However, the effect of BIV infection on immune functions has not been fully characterised.Material and methodsTranscriptome analysis of BoMac cells after in vitro infection with BIV was performed using BLOPlus bovine microarrays. Genes identified as differentially expressed were subjected to functional analysis with the Ingenuity Pathway Analysis software (IPA).ResultsOut of 1,743 genes with altered expression, 1,315 were mapped as unique molecules. In total, 718 genes were identified as upregulated and 597 genes as downregulated. Differentially expressed genes were involved in 16 pathways related to immune response. The most enriched canonical pathway was leukocyte extravasation signalling. Interleukin-15 (IL-15) production was indicated as the most activated pathway and the 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 4 (PFKFB4) signalling pathway was the most inhibited one. In addition, the study showed that the inflammatory response was decreased during BIV infection.ConclusionThis is the first report to describe the microarray analysis of changes in gene expression upon BIV infection of bovine macrophages. Our data indicated how BIV influences the expression of genes and signalling pathways engaged in the immune response.

Project description:We used Next-generation sequencing (NGS) to reveal MbovP475-binding sites in BoMac cells. We extracted the genome of BoMac infected with mycoplasma bovis by sonication, then the sheared chromatin in the supernatant was incubated with purified rabbit polyclonal IgG against MbovP475 and protein A/G magnetic beads. The DNA was eluted from the beads and subject to making sequencing library. We totally got 2648 potential MbovP475-binding sites by ChIP-seq. Among them, CRYAB and MCF2L2 gene were further verifed to be down-reguated by MbovP475 through binding to the promoter region of two genes.

Project description:Resistance traits of parasitic infestation and diseases in bovine breeds display various unknown differences in the immune response indicating to be related to the host`s genomic background. We studied the differential gene expression of monocyte-derived macrophages (MDMs) stimulated or unstimulated with LPS in vitro, from two bovine breeds by RNA sequence methodology. MDMs were grown in supplemented RPMI medium for 11 days. Then, 100 ng/ml LPS was added to medium for 48h in treated samples. Total RNA was extracted from macrophages and sequenced by RNA-Seq technology. Resulting sequences were mapped to the bovine reference genome and gene expression was compared between breeds. We found several differences in immune response activation, as leucocyte recruitment, immune response, antigen processing and presentation, positive regulation of NFκB and cell cycle biological processes in unstimulated cells and lipid metabolism, inflammatory response, regulation of cell proliferation and cycle and arginine catabolism in LPS treated cells. Finally, LPS seems to trigger inflammation by distinct pathways in these two bovine breeds.

Project description:Mycotoxins are secondary metabolites produced by a variety of fungi that contaminate animal food and feeds and are capable of inducing a wide range of toxicities. Predictive in vitro models represent valuable substitutes for animal experiments to assess the toxicity of mycotoxins. The complexities of the interactions between epithelial and innate immune cells, vital for upholding barrier integrity and averting infections, remain inadequately understood. In the current study, a co-culture model of bovine epithelial cells (MAC-T) and macrophages (BoMac) was used to investigate the impact of exposure toFusariummycotoxins, namely deoxynivalenol (DON), zearalenone (ZEN), enniatin B (ENB), and beauvericin (BEA), on the inflammatory response elicited by the bacterial lipopolysaccharide (LPS) endotoxin. The MAC-T cells and BoMac were seeded on the apical side of a Transwell membrane and in the lower chamber, respectively, and mycotoxin exposure on the apical side of the membrane was carried out with the different mycotoxins (LC20; concentrations that elicited 20% cytotoxicity) for 48 h followed by an LPS immunity challenge for 24 h. The culture supernatants were collected from the basolateral compartment and these samples were submitted for cytokine/chemokine multiplex analysis. RNA-Seq analysis was performed using total RNA extracted from the MAC-T cells to acquire a more detailed insight into their cellular functions. The multiplex analysis indicated that IFN-γ, IL-1α, IL-8, and MCP-1 were significantly induced post-DON treatment when compared to control cells, and levels of IL-1α and IL-8 were enhanced significantly in all mycotoxin-treated groups post-LPS challenge. Analysis of the sequencing results showed that there were 341, 357, and 318 differentially expressed MAC-T cell genes that were up-regulated in the DON, ENB, and BEA groups, respectively. Gene ontology and pathway analysis revealed that these DEGs were significantly enriched in various biological processes and pathways related to inflammation, apoptosis signaling, and Wnt signaling. These results provide a comprehensive analysis of the co-culture cytokine/chemokine production and MAC-T cells’ gene expression profiles elicited byFusariummycotoxins, which further contributes to the understanding of early endotoxemia post-mycotoxin exposure.

Project description:In this study, we determined the miRNA expression profile of bovine alveolar macrophages, using next-generation sequencing strategy. On an Illumina HiSeq 2000 machine, we sequenced 8 miRNA libraries, prepared from small RNA fractions of alveolar macrophages isolated from 8 different healthy animals (Bos taurus). From the data, the potential novel miRNAs were predicted, and the expression levels of the known miRNAs were determined. We report that 80 known bovine miRNAs are expressed in bovine alveolar macrophages with >100 reads per million. The most highly expressed miRNA was bta-miR-21, followed by bta-miR-27a. Additionally, one putatively novel bovine miRNA was identified. To our knowledge, this is the first RNA-seq study to profile miRNA expression in bovine alveolar macrophages and provides an important reference dataset for investigating the regulatory roles miRNAs play in this important immune cell type.
