Project description:Mesenchymal stem cells (MSCs) isolated from human gingival tissue and periodontal ligament (PDL) have shown potential for periodontal regeneration. Although PDL-MSCs have been recognized as the best choic for cell-based periodontal regeneration, isolation of PDL-MSCs required tooth extraction which is impractical for clinical use. Supracrestal gingival connective tissue (SG) is a part of gingival tissue which located closely to PDL and can be harvested without tooth extraction. Here, the characteristics of MSCs derived from SG was evaluated. Differential gene expression profiles of hSG-MSCs, hPDL-MSCs, and human marginal gingival tissue (MG)-derived MSCs were compared. Results demonstrated that hSG-MSCs had a more similar gene expression profile to the hPDL-MSCs than the hMG-MSCs, suggested a potential candidate cell source of SG for future peridontal tissue engineering.

Analysis of the gingival tissue transcriptome in states of periodontal health and disease may reveal novel insights of the pathobiology of periodontitis. Project description:EXPERIMENT: Microarray expression profiles derived from the human primary gingival epithelial cells 24.0h after exposure to heat inactivated P. gingivalis ANIMAL MODEL: NON EXPOSURE: Human primary gingival epithelial cells (at 3rd passage) were exposed to heat inactivated P. gingivalis (MOI:100) at 90% confluence. Two types of gingival epithelial cells were used. One with Normal cytokine inducer type (at least 2 fold IL-6/TNF-alpha/IL-1ß when challenged with TLR2/4 agonists) and the other with diminished cytokine inducer type (no change in IL-6/TNF-alpha/IL-1ß when challenged with TLR2/4 agonists). Project description:We investigated the association between subgingival bacterial profiles and gene expression patterns in gingival tissues of patients with periodontitis. A total of 120 patients undergoing periodontal surgery contributed with a minimum of two interproximal gingival papillae (range 2-4) from a maxillary posterior. Prior to tissue harvesting, subgingival plaque samples were collected from the mesial and distal aspects of each papilla. Gingival tissue RNA was extracted, reverse-transcribed, labeled, and hybridized with whole-genome microarrays (310 in total)
