Project description:A previously described low-fitness, high stress-resistant, variant of Listeria monocytogenes LO28 WT was subjected to an experimental evolution regime, selecting (in two parallel lines) for increased fitness in unstressed conditions. Evolved variants with increased fitness reverted to WT-like stress resistance. Whole genome sequencing and proteomics were used to identify differences between the ancestral and evolved strains.

Project description:We infected wild type L. monocytogenes EGD-e (1) and its isogenic deltahlydeltaplcA (2) (lacking the ability to breach the vacuolar compartment of host cells following uptake) mutant strain to human intestinal epithelial cell line (Caco-2) with an MOI of 100 and 500 respectively. Bacterial total RNA was isolated at 1 h (deltahlydeltaplcA) and 4 h (EGD-e) post infection, reverse transcribed, hybridised to whole genome microarray and microarray data was analysed as described previously (3) 1. Decoding Listeria by N-terminomics

Project description:This dataset contains spectral information of protein N-terminal peptides isolated from Listeria monocytogenes EGD-e, a bacterial model organism and human pathogen. When mapped onto the Listeria genome these peptides indicate the exact location of translation initiation sites (TIS). The large majority of the identified TIS corresponded to start sites of predicted open reading frames (ORFs), however, a significant fraction of the identified TIS indicated deviations from the current genome annotation. The latter include primarily TIS inside the sequence of predicted ORFs or TIS that delineate the start position of novel ORFs.

Project description:Investigation of whole genome gene expression level changes in Listeria monocytogenes LO28 delta-lhrC1-5 mutant, compared to the wild type strain. The lhrC1-5 genes encode the regulatory sRNAs LhrC1-5. The microarray studied the gene expression of unstressed cells and cells exposed to cefuroxime for 30 min. The lhrC1-5 mutant employed in this study is further described in Sievers et al. (2014) A multicopy sRNA of Listeria monocytogenes regulates expression of the virulence adhesin LapB.
