Project description:Children with relapsed or refractory T-cell acute lymphoblastic leukemia (T-ALL) have a poor prognosis with few therapeutic options. Multiple lines of evidence point towards the enzyme dihydroorotate dehydrogenase (DHODH) as a metabolic vulnerability in dysregulated and malignant T‑cells. DHODH catalyzes the fourth step of de novo pyrimidine nucleotide synthesis (e.g., uridine, cytidine). Therefore, inhibiting DHODH via small molecule inhibitors (DHODHi) rapidly leads to the depletion of intracellular pyrimidine pools and forces cells to rely on extracellular salvage. In the absence of sufficient salvage, this intracellular nucleotide starvation results in the inhibition of DNA and RNA synthesis, cell cycle arrest, and ultimately death. T‑lymphoblasts appear to be specifically and exquisitely sensitive to nucleotide starvation following DHODHi. We have confirmed this sensitivity in vitro as well as in vivo in three murine models of T‑ALL. Additionally, we have identified that certain subsets of T-ALL seem to have an increased reliance on oxidative phosphorylation when treated with DHODHi. Through a series of metabolic assays, we show that leukemia cells, in the setting of nucleotide starvation, have changes in their mitochondrial membrane potential and may be more highly dependent on alternative fuel sources. We hope these changes point to a metabolic vulnerability that may distinguish them from normal T-cells and other normal hematopoietic cells and offer an exploitable therapeutic opportunity. The availability of clinical-grade DHODH inhibitors currently in human clinical trials speaks to the potential for rapidly advancing this work into the clinic.

Project description:A hallmark of Parkinson’s disease is the specific degeneration of dopaminergic neurons in the substantia nigra pars compacta. Interestingly, not all of these neurons are equally affected. Studies revealed that neurons located more ventrally within the substantia nigra pars compacta have a higher prevalence to degenerate than those located in the dorsal tier. Reasons for this selective neuronal vulnerability are still unknown. The objective of the present study was to gain a better understanding of molecular differences between these two neuronal subpopulations that can explain the selective neuronal vulnerability within the substantia nigra. For this, both neuronal subpopulations were specifically isolated with laser microdissection. Following, their proteome was analyzed via data independent acquisition mass spectrometry. The results of this study revealed a minor number of proteins that were either specific or differentially expressed in one of the examined neuronal subpopulations within the substantia nigra. These proteins are associated with the cytoskeleton, neuronal plasticity, or calcium homeostasis. With these findings a deeper understanding can be gained of the selective neuronal vulnerability within the substantia nigra and of protective mechanisms against neurodegeneration in specific neuronal subpopulations.

Project description:Despite intensive therapy, children with high-risk neuroblastoma are at risk of treatment failure. We applied a multi-omic system approach to evaluate metabolic vulnerabilities in human neuroblastoma. We combined metabolomics, CRISPR screening and transcriptomic data across >700 solid tumor cell lines and identified dihydroorotate dehydrogenase (DHODH), a critical enzyme in pyrimidine synthesis, as a potential treatment target. Of note, DHODH inhibition is currently under clinical investigation in patients with hematologic malignancies. In neuroblastoma, DHODH expression was identified as an independent risk factor for aggressive disease, and high DHODH levels correlated to worse overall and event-free survival. A subset of tumors with the highest DHODH expression was associated with a dismal prognosis, with a 5-year survival of <10%. In xenograft and transgenic neuroblastoma mouse models treated with the DHODH inhibitor brequinar, tumor growth was dramatically reduced, and survival was extended. Furthermore, brequinar treatment was shown to reduce the expression of MYC targets in three different neuroblastoma models in vivo. A combination of brequinar and temozolomide was curative in the majority of transgenic TH-MYCN neuroblastoma mice, indicating a highly active clinical combination therapy. Overall, DHODH inhibition combined with temozolomide has therapeutic potential in neuroblastoma and we propose this combination for clinical testing.

Project description:Despite the rapid progress in dissecting neural circuits for social behaviors, it remains unknown whether specific neural cell types are selectively vulnerable in social dysfunction cases often associated with neurodevelopmental disorders. Here, employing a single-cell transcriptome analysis in mice, we show that an embryonic disturbance known to induce social dysfunction preferentially impairs gene expressions crucial for neural functions in parvocellular oxytocin (OT) neurons—a subtype linked to social rewards—while neighboring cell types experience a lesser impact. Chemogenetic stimulation of OT neurons at the neonatal stage ameliorated social deficits, concomitant with a cell-type-specific sustained recovery of the pivotal gene expressions. Our data illuminates the transcriptomic selective vulnerability within the hypothalamic social behavioral center, offering a potential therapeutic target through specific neonatal neurostimulation.

Project description:Patients with Marinesco-Sjögren syndrome and gene targeting in mice revealed an essential role for the SIL1/Sil1 gene in maintenance of central nervous tissue and skeletal muscle. SIL1/Sil1 expression is not restricted to a certain tissue, and the gene product, SIL1/Sil1, localizes to the (sarco)endoplasmic reticulum. Due to the ADP/ATP exchange activity of SIL1/Sil1 for the major chaperone BiP (GRP78) it functions as a co-chaperone. It is still an enigma how loss of functional SIL1/Sil1 can lead to selective vulnerability of the nervous system and skeletal muscle whereas other cellular populations like lymphoblastoid cells (LCs) are protected against clinical manifestation of SIL1/Sil1 deficiency. In this study we address this issue in order to identify compensatory strategies and to gain a deeper understanding of the selective vulnerability in SIL1 pathology. By studying the morphology of SIL1-deficient LCs, we demonstrate that they present with altered organelle structures indicating subclinical vulnerability and once more the assumed presence of cellular strategies antagonizing phenotypical manifestation of loss of this co-chaperone. Proteomic profiling of MSS-derived LCs revealed affection of nuclear and mitochondrial integrity, cytoskeleton and cellular viability. Apart from that proteins, known to be causative for hereditary neuromuscular disorders or to be involved in development and function of the nervous system were altered in expression. Paradigmatic findings were confirmed in spleen of Sil1 deficient mice in comparison to appropriate controls. These findings along with the results of viability assays and the identification of activation of pro-survival mechanisms provide insights into cellular strategies antagonizing SIL1/Sil1 deficiency and on a more general note into the molecular cause of selective vulnerability defining the SIL1/Sil1-phenotype, in both man and mouse.
