However, its function in antiviral immunity is controversial, and how immune responses regulate m6A modification is unknown. We here found TBK1, a key kinase of antiviral pathways, phosphorylated the core m6A methyltransferase METTL3 at Serine 67. The phosphorylated METTL3 interacted with translational complex and enhanced proteins translation, including IRF3, and facilitated antiviral responses. TBK1 also promoted METTL3 activation and m6A modification, which is required for stabilizing IRF3 mRNA. Type I IFN induction was severely impaired in METTL3 deficient cells. Mettl3flfl-lyz2-Cre mice were significantly more susceptible to IAV-induced lethality than control mice. Consistently, Ythdf1—/— mice cannot control viral infection and showed higher mortality than control mice due to decreased IRF3 expression. Together, we demonstrated that innate signals activated METTL3 via TBK1, and METTL3 and m6A modification secured antiviral immunity by promoting mRNA stability and protein translation.

Project description:Troxerutin (TXR), a potent antioxidant compound shows anti-inflammatory activity and possesses hepatoprotective effect. In this study, we aimed to exploit its anti-arthritic properties using adjuvant induced arthritic model. Using relative quantitative proteomics approach, we also tried to understand the mode of action of TXR at mechanistic details and its possible usage in the treatment of arthritis. Results provide a set of candidate biomarkers for responses to TXR in the arthritis and suggest new modalities of anti-arthritic activities. A number proteins were identified from the joint homogenates of the experimental rats using isobaric tag for relative and quantitative proteomics (iTRAQ) method. The detailed study and validation of these proteins involved may be useful in finding out newer treatment modalities.

Project description:RIG-I like helicases (RLHs) and cGAS are crucial cytosolic sensors of viral RNA and DNA, respectively, and induce type I IFNs via TBK1/IKKε. hnRNPM was described to possess antimicrobial activities. Here, we show that hnRNPM promotes TBK1/IRF3 phosphorylation and type I IFN induction downstream of cGAS and RIG-I in THP-1 cells and primary human fibroblasts. Interactome analysis revealed that hnRNPM forms a multiprotein complex with ELAVL1, SON, TBK1, IKKε, IKKβ, and NF-κB p65. The purified hnRNPM complex enhanced TBK1 phosphorylation. Vice versa, TBK1 directly phosphorylated ELAVL1. hnRNPM, ELAVL1, and TBK1 predominantly interacted in the cytosol. Of note, patients with mutations in SON suffer from recurrent infections, further underlining its biologic significance. To our knowledge, hnRNPM, ELAVL1, and SON represent the first non-redundant signaling components merging the cGAS-STING and RIG-I–MAVS pathways. Therefore, our findings may have implications for host defense and auto-inflammatory diseases.
