Project description:The concept of neurotransmitter identity, once considered singular and immutable for mature neurons, has been broadened by the recognition that many neurons release multiple neuro-active substances. In order to demonstrate the extent of neurotransmitter co-expression, we labelled neuronal nuclei with UNC84-GFP using VAChT and either Vglut or VGAT split-GAL4s and isolated them with the isolation of nuclei tagged in specific ell types (INTACT) method From the isolated nuclei, we isolated and sequenced Poly(A) RNA and examined the relative expression levels of glutamatergic, GABAergic, or cholinergic genes in each population.

Project description:Chromatin profiling of nuclei isolated from genetically defined neuronal subpopulations of the adult Drosophila brain. Cell type-specific histone modification maps were generated from nuclei isolated from all neurons (R57C10-GAL4), Kenyon cells (OK107-GAL4), and octopaminergic (Tdc2-GAL4) neurons using a method similar to INTACT (Deal and Henikoff, 2010; Steinner et al., 2012). Three histone modifications were profiled: H3K4me3, H3K27ac, and H3K27me3. Sequencing was performed with an Illumina HiSeq 2000.

Expression data from WT and VAChT KDHOM ventricles

Project description:VAChT KDHOM mice have a 70% decrease in the vesicular acetylcholine transporter (VAChT) and this leads to a systemic decrease in ACh release and cardiac dysfunction. We used this microarray to determine whether there were transcriptional alterations in the hearts of mutant mice as a result of this long-term decrease in cholinergic tone. We isolated whole hearts from 3-month-old WT and VAChT KD mice and used them for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain homogenous populations for each sample and, as such, pooled together two RNA samples to generate one microarray sample.
