Project description:Small RNA expression from Nicotiana benthamiana leaves was profiled with the primary goal of ascertaining microRNA isoform diversity for known, conserved families. A secondary goal was to provide a baseline small RNA expression atlas for this species and tissue. Two biological replicates of leaves from growth-room grown plants. The two libraries were each run twice on different runs, so there are a total of four datasets. For most analyses, it will be appropriate to combine the run1 and run2 versions for the libraries.

Project description:In order to find the differentially expressed RNAs and signaling pathways related to the dwarf phenotype induced by NlCSP11, 35S-NlCSP11, 35S-XopQ and 35S-GFP control were infiltrated into Nicotiana benthamiana. We analyzed the differentially expressed RNAs in the infiltrated leaves (NlCSP11_Z/XopQ_Z) and the upper uninfiltrated leaves (NlCSP11_U/XopQ_U) of N. benthamiana. KEGG annotation revealed a large number of differentially expressed RNAs involved in plant-pathogen interactions, indicating that NlCSP11 resulted in a more strong systemic resistance in N. benthamiana.
Project description:The 35S::GFP fluorescence was silenced 6-day after infiltration to Nicotiana benthamiana leaves due to the post-transcriptional gene silencing, but became stable by adding the viral suppressor 2b which repressed RNA silencing in plants. We performed the small RNA high throughput sequencing to test whether WUS can inhibit 2b functions in repressing plant RNA silencing.

KEGG annotation revealed a large number of differentially expressed miRNAs involved in plant hormone signal transduction, indicating that NlCSP1 changed the hormone signal pathway in N. benthamiana.
Project description:Verticillium dahliae is a soil-borne fungus with a broad host range, including the model plants Nicotiana benthamiana and Arabidopsis thaliana. The plant immunity can be activated by Vd-derived patterns while V. dahliae secreted effectors. We report here the RNAseq analyses of samples from N. benthamiana infected by V. dahliae strains 592 and 171 at different time points. The data showed dynamic expression patterns of genes not only from plant defense signaling but also plant developmental signaling.
