Project description:We have employed whole genome microarray expression profiling to identify genes that are differentially expressed after a single gas plasma exposure in two different head and neck cancer cell lines. Additionally, we aimed to identify genes correlating with cellular adaptation upon repetitive oxidative stress wherefore two different head and neck cancer celllines were exposed to gas plasma weekly in eight treatment cycles.
Project description:Exosomes were isolated from plasma of n = 12 healthy donors (HD) and n = 16 head and neck cancer (HNSCC) patients. miRNA profiling of exosomes was performed using nCounter SPRINT system. miRNAs being predicted to target EMT-related genes (CDH1, VIM, TWIST1 and SNAI1/2) were selected and compared between HD and HNSCC patients.

The Illumina Infinium 27k Human DNA methylation BeadChip v1.2 was used to obtain DNA methylation profiles across approximately 27,000 CpGs in fresh-frozen tumor tissues. Samples included 91 tumors from the oralpharynx and larynx, reflecting all stages of disease, 18 normal head and neck National Disease Research Interchange (NDRI) samples, and 213 normal control blood samples. Bisulphite-converted DNA from the 18 normal head and neck samples were hybridized to the Illumina Infinium 27k Human Methylation Beadchip v1.2.
