Maternal metabolic stress affects in vitro embryo's transcriptom pattern

Project description:This study aims to investigate the potential implication of epigenetic mechanisms in the transmission of maternal metabolic information to the embryo, in order to better understand how maternal metabolic stress influences fertility. To do this, we exposed the oocytes of 54 cows to different environments during maturation (24 hours). The treatments include normal glucose concentration (5.5 mM), low glucose concentration (2.75 mM) and low glucose concentration with supplementation of BHB (1.8 mM). BHB is a ketone body that accumulates in circulation during clinical ketosis due to an increased rate of fat mobilization. BHB can be metabolized by the embryo in addition to acting as an epigenetic regulator. Epigenetic modifications are a potential mechanism for transmitting maternal metabolic information from the oocyte to the embryo, given their heritability. As a result, we observe normal oocyte maturation within each condition. On the other hand, we see significantly higher blastocyst rates in the BHB supplemented group compared to the low glucose group (p-value = 0.0318). By modulating the expression of several genes, some of which related to development and metabolism, glucose deprivation (low glucose) has a greater influence on the embryo transcriptome than the exposure to BHB during maturation. BHB appears to have a compensatory effect on blastocyst rates by minimizing the impact of glucose deprivation on developmental rates. This project elucidates how certain metabolic conditions, including clinical ketosis in dairy cows or obesity in women influences fertility and the health of offspring.

Maternal weight affects placental DNA methylation of genes invovled in metabolic pathways in the common marmoset monkey (Callithrix jacchus)

Project description:Accumulating evidence suggests that dysregulation of placental DNA methylation (DNAm) is a mechanism linking maternal weight during pregnancy to metabolic programming outcomes. The common marmoset, Callithrix jaccus, is a platyrrhine primate species that has provided much insight into studies of the primate placenta, maternal condition, and metabolic programming, yet the relationships between maternal weight and placental DNAm are unknown. Here, we report genome-wide DNAm from term marmoset placentas using reduced representation bisulfite sequencing. We identified 74 genes whose DNAm pattern is associated with maternal weight during gestation. These genes are predominantly involved in energy metabolism and homeostasis, including regulation of glycolytic and lipid metabolic processes pathways.

Maternal IgG Affects Brain Development of Mouse Pups

Project description:Maternal IgG is an immunoglobulin present in breast milk and has been shown to play a role in the development of the immune system in infants. The aim of this study was to investigate the effects of maternal IgG on brain development in mouse pups. Maternal IgG immunoreactivity was observed in microglial cells of the pup brain, peaking at postnatal 8 day. In particular, strong IgG immunoreactivity was observed in the microglia in the corpus callosum and cerebellar white matter. IgG stimulation of primary cultured microglia resulted in activation of the type I interferon feedback loop by Syk. Among the IgG subclasses, IgG2a was found to have a strong influence on microglia. Analysis of FcRn KO mice that were unable to take up IgG from their mothers revealed that these mice had abnormalities in the proliferation and/or survival of microglia, oligodendrocytes, and some types of interneurons. FcRn KO mice also exhibited abnormalities in social behavior and adaptation to novel environments. This study suggests that maternal IgG affects brain development in pups.

Transfection with Glucuronidase A affects general proteome pattern in Giardia lamblia trophozoites

Project description:Giardia lamblia is an important causative agent of persistent diarrhea in humans, domestic animals, and cattle. Basic research is usually performed with the strain WBC6 and includes genetic manipulations such as transfections. Here, we investigate how transfection with a plasmid causing stable expression of a foreign gene affects the whole proteome pattern. Using shotgun mass spectrometry, we compare the proteomes of untransfected trophozoites to trophozoites transfected with Escherichia coli glucuronidase A (GusA). Besides GusA, which is detected in the transfected trophozoites only, the proteomes of untransfected and transfected trophozoites differ by 132 differentially expressed proteins. In particular, transfection induces antigenic variation. Since transfection causing stable expression affects the proteome pattern, transfection experiments should take into account this effect. Due to a unique peptide panel, GusA is an example for a suitable internal standard for experiments involving transfected cells.

Project description:We analysed the combined effects of exposure to maternal diabetes and disrupted HIF-1 signaling on the transcriptom in cardiac left ventricles of 12 weeks old male mice. This approach provides the information about the long term changes originating in utero due to maternal diabetes and inefficient response to hypoxia which develops as a result of hyperglycemia. The majority of changes were detected in Hif1a insufficient mice exposed to maternal diabetes.

Maternal antibiotic use in sows affects microbiota and gut development in offspring

Project description:We demonstrated that a maternal antibiotic treatment can change intestinal development of the offspring piglets permanently by showing that maternal gestational antibiotic treatment affects intestinal development in offspring piglets for a period of at least seven weeks after the antibiotic treatment in the sows was finished. It was shown that immediately after birth the piglets from amoxicillin treated sows, showed upregulation of genes involved in processes related to ‘tight junctions’ and ‘immunoglobulins’. In addition, these piglets had significantly lower number of goblet cells. Together, this may lead to a gut wall that is more rapidly closed in piglets from amoxicillin treated sows, affecting the uptake of immunoglobulins and the intestinal development. Later in life, around weaning, gene expression and morphological data indicate that the crypts of piglets from amoxicillin treated sows deepen around weaning as an effect of the amoxicillin treatment which in combination with the upregulation of genes involved in cell cycle processes, ribosomal activity and protein degradation might imply that the intestinal development, the subsequent differentiation of cells or the timing of these processes was delayed by the maternal antibiotic treatment.
