Project description:In a cross-over randomised controlled trial,19-night shift workers with abdominal obesity, completed two diet conditions separated by a two week wash out period; a) the control period = four weeks of habitual eating overnight and b) the intervention period = four weeks of avoiding food intake between 1 AM – 6 AM. At the end of each intervention period participants were given a high fat/high carbohydrate challenge meal in the morning and peripheral blood mononuclear cells were isolated from whole bood sampled at fasting, 2 and 4 hours after eating the meal.

Investigating the transition of Apis mellifera summer workers to diutinus workers using proteomic techniques.

Project description:Apis mellifera workers in temperate climates display two castes; short lived summer bees that engage in nursing, hive maintenance and foraging, and long lived winter bees (diutinus bees) which remain within the hive and are essential for thermoregulation. Label free quantitative proteomic analysis was conducted on A. mellifera workers sampled from July to October 2019 to compare the proteomes of workers as the colony progresses through the year. Proteomic analysis revealed a shift in protein expression in workers in September and October in comparison to July and August samples. Workers samples in September and October had a higher abundance of proteins associated with oxidative phosphorylation and storage proteins such as hexamerin. Interestingly, a shift in protein expression was detected in newly emerged bees between July to October, providing evidence that workers have adapted to emerge with a different protein profile in preparation for the winter months.

Project description:We identified and validated characteristic miRNA expression profiles of human whole blood in workers exposed to volatile organic compounds (VOCs) and compared the usefulness of miRNA indicator of VOCs with the effectiveness of the already used urinary biomarkers of occupational exposure. Using a microarray based approach, we screened and detected deregulated miRNAs in their expression in workers exposed to VOCs (toluene [TOL], xylene [XYL] and ethylbenzene [EBZ]). Total 169 workers from four dockyards were enrolled in current study, and 50 subjects of them were used for miRNA microarray analysis.

Project description:Ant societies exhibit a division of labor in which a queen is in charge of reproduction while non-reproductive workers maintain the colony. In Harpegnathos saltator, workers retain reproductive ability, inhibited by the queen pheromones. Following the queen loss, the colony undergoes social unrest with an antenna dueling tournament. Most workers quickly abandon the tournament while a few workers continue the dueling for months and become gamergates (pseudoqueens). However, the temporal dynamics of the molecular mechanisms underlining the caste transition and social dominance remain unclear. To identify genetic factors responsible for this transition, we compared transcriptomes of ovary, fat body, and non-visual brains between dueling and non-dueling workers.

Transcriptional profiling by array of Solenopsis invicta (fire ant) workers to identify genes and gene pathways responsible for division of labor

Project description:We used whole-genome fire ant microarrays to examine the molecular basis for division of labor in fire ant workers by comparing foraging and non-foraging workers from monogyne colonies. Fire ant colonies were collected in the field and transported into the lab were they were reared in standard conditions. We created a nesting chamber containing the queen, the brood and workers performing nursing tasks and a foraging area, separated from the nesting chamber and provided with food and water sources. Foraging workers were collected in the foraging area while non-foraging workers were collected in the nesting chamber. Total RNA was isolated from pools of whole workers and processed for microarrays.

Molecular mechanisms of task allocation in workers of the red imported fire ant, Solenopsis invicta

Project description:The red imported fire ant Solenopsis invicta is an invasive pest in the USA, eastern Asia and Australia that causes billions worth of damage where it has been introduced. In the insect colony, workers perform tasks based on their age as well as the subcaste, where the younger workers as well as the members of the smaller subcastes of workers tend to remain in the nest and tend to the brood while the older ants as well as the members of the larger subcastes of workers leave the nest to perform activities such as foraging. In eusocial insects, juvenile hormone has been identified to be a catalyst for behavioral changes among the worker caste, but the involvement of this hormone in S. invicta task allocation has not been investigated. Here, we conducted RNAseq analysis to identify genes associated with worker division of labor by comparing the expression profiles of foragers and nurses from the medium subcaste. Additionally, we evaluated the changes in the transcriptome of nurses in response to the application of the juvenile hormone analog, S-hydroprene. We found 816 differentially expressed genes between foragers and nurses and 100 differentially expressed genes between nurses treated with acetone and nurses treated with S-hydroprene. Here, we focused on the differentially expressed genes between foragers and nurses that were associated with different molecular pathways such as energy metabolism, glycolysis, juvenile hormone synthesis and metabolism, and immunity. Our results showed that the juvenile hormone analog induced changes in worker gene expression, some of which were as expected if the juvenile hormone is involved in regulating the shift from nursing to foraging in S. invicta workers. This analysis and data from this experiment can provide a slew of potential target genes to be used for control of this invasive pest.

Molecular basis of reproductive plasticity of termite workers under environmental stresses

Project description:Eusociality is an important step in animal evolution that increases the level of biological complexity. The reproductive plasticity of termite female workers provides colonies with the tremendous flexibility to respond to environmental (social) stresses. Here, we established an “inhibition-activation-reinhibition” research model of the reproductive transformation of female workers using three Reticulitermes species. We identified the functional genes involved in the transformation process, explaining why the transformation of workers to reproductives in R. aculabialis were not triggered under the same isolation conditions. Transposable elements in the brains of workers promote the transformation of workers into reproductives. The metabolic levels were associated with changes in stress caused by female reproduction and the nest environment. Strikingly, we confirmed that when female workers experience severe environmental stresses that affect their survival, they do not initiate reproductive transformation, even if there is no female reproductive in the nests. Furthermore, based on the regulatory network of molecules, we constructed machine learning models to demonstrate how numerous genes co-regulate the reproductive plasticity of workers and to predict the fertility of colonies. Among these, deep neural network models had the highest prediction accuracy and generalization, indicating that workers carry a wealth of information about their colony at the molecular level. We suggest that combined molecular function and deep learning model analysis for caste differentiation, reproductive plasticity, colony development and adaptation will provide a methodology for revealing complex gene regulatory networks that respond to environmental changes in social insects. Our findings are essential for understanding the reproductive plasticity of humans and other social animals to respond to environmental changes and social stresses.
