Project description:Smoking cigarettes is harmful to the cardiovascular system. Considerable attention has been paid to the reduced harm potential of alternative nicotine-containing inhalable products such as e-cigarettes. We investigated the effects of E-vapor aerosols or cigarette smoke (CS) on atherosclerosis progression, cardiovascular function, and molecular changes in the heart and aorta of female ApoE−/− mice. The mice were exposed to aerosols from three different E-vapor formulations: (1) carrier (propylene glycol and vegetable glycerol), (2) base (carrier and nicotine) or (3) test (base and flavor) or to CS from 3R4F reference cigarettes for up to 6 months. Concentrations of CS and base or test aerosols were matched at 35 µg nicotine/L. Exposure to CS, compared with sham-exposed fresh air controls, accelerated atherosclerotic plaque formation, while no such effect was seen for any of the three E-vapor aerosols. Molecular changes indicated disease mechanisms related to oxidative stress and inflammation in general, plus changes in calcium regulation, and altered cytoskeletal organization and microtubule dynamics in the left ventricle. While ejection fraction, fractional shortening, cardiac output, and isovolumic contraction time remained unchanged following E-vapor aerosols exposure, the nicotine-containing base and test aerosols caused an increase in isovolumic relaxation time similar to CS. A nicotine-related increase in pulse wave velocity and arterial stiffness was also observed, but it was significantly lower for base and test aerosols than for CS. These results demonstrate that in comparison with CS, E-vapor aerosols induce substantially lower biological responses associated with smoking-related cardiovascular diseases.

Project description:Volume concentrations of secondary organic aerosol (SOA) are measured in 139 steady-state, single precursor hydrocarbon oxidation experiments after passing through a temperature controlled inlet. The response to change in temperature is well predicted through a feedforward Artificial Neural Network. The most parsimonious model, as indicated by Akaike's Information Criterion, Corrected (AIC,C), utilizes 11 input variables, a single hidden layer of 4 tanh activation function nodes, and a single linear output function. This model predicts thermal behavior of single precursor aerosols to less than ±5%, which is within the measurement uncertainty, while limiting the problem of overfitting. Prediction of thermal behavior of SOA can be achieved by a concise number of descriptors of the precursor hydrocarbon including the number of internal and external double bonds, number of methyl- and ethyl- functional groups, molecular weight, and number of ring structures, in addition to the volume of SOA formed, and an indicator of which of four oxidant precursors was used to initiate reactions (NOx photo-oxidation, photolysis of H2O2, ozonolysis, or thermal decomposition of N2O5). Additional input variables, such as chamber volumetric residence time, relative humidity, initial concentration of oxides of nitrogen, reacted hydrocarbon concentration, and further descriptors of the precursor hydrocarbon, including carbon number, number of oxygen atoms, and number of aromatic ring structures, lead to over fit models, and are unnecessary for an efficient, accurate predictive model of thermal behavior of SOA. This work indicates that predictive statistical modeling methods may be complementary to descriptive techniques for use in parametrization of air quality models.

Project description:This study aims to to compare the gene transcription profiles of endothelial cells and stem cells, when they are cultured alone or when they are cultured together. Thus there are two major questions - how do the cells differ, and how do the cells influence each other's gene expression. Thus there are 4 types of sample: endothelial cell monoculture, endothelial cell coculture, stem cells monoculture, stem cell coculture. There are also 4 biological replicates (independent experiments) leading to 16 array data files.

Project description:In this study, we evaluated photosensitized chemistry at the air-sea interface as a source of secondary organic aerosols (SOA). Our results show that, in addition to biogenic emissions, abiotic processes could also be important in the marine boundary layer. Photosensitized production of marine secondary organic aerosol was studied in a custom-built multiphase atmospheric simulation chamber. The experimental chamber contained water, humic acid (1-10 mg L(-1)) as a proxy for dissolved organic matter, and nonanoic acid (0.1-10 mM), a fatty acid proxy which formed an organic film at the air-water interface. Dark secondary reaction with ozone after illumination resulted in SOA particle concentrations in excess of 1000 cm(-3), illustrating the production of unsaturated compounds by chemical reactions at the air-water interface. SOA numbers via photosensitization alone and in the absence of ozone did not exceed background levels. From these results, we derived a dependence of SOA numbers on nonanoic acid surface coverage and dissolved organic matter concentration. We present a discussion on the potential role of the air-sea interface in the production of atmospheric organic aerosol from photosensitized origins.

Project description:Secondary organic aerosols (SOA) are a large source of uncertainty in our current understanding of climate change and air pollution. The phase state of SOA is important for quantifying their effects on climate and air quality, but its global distribution is poorly characterized. We developed a method to estimate glass transition temperatures based on the molar mass and molecular O:C ratio of SOA components, and we used the global chemistry climate model EMAC with the organic aerosol module ORACLE to predict the phase state of atmospheric SOA. For the planetary boundary layer, global simulations indicate that SOA are mostly liquid in tropical and polar air with high relative humidity, semi-solid in the mid-latitudes and solid over dry lands. We find that in the middle and upper troposphere SOA should be mostly in a glassy solid phase state. Thus, slow diffusion of water, oxidants and organic molecules could kinetically limit gas-particle interactions of SOA in the free and upper troposphere, promote ice nucleation and facilitate long-range transport of reactive and toxic organic pollutants embedded in SOA.
