Project description:MicroRNAs (miRNAs) are short non-coding RNAs that are involved in post-transcriptional control of gene expression. The objective of this case-control study was to explore circulating miRNAs in young individuals with long-duration type 1 diabetes (T1D) and healthy controls. Twelve adolescents, age 15.0–17.9 years, with T1D duration of more than 8 years (mean 11.1 years), were enrolled from the Swedish diabetes quality registry. An age-matched control group (n = 12) was recruited. Circulating miRNAs (n = 187) were analyzed by quantitative PCR. We observed that 27 miRNAs were upregulated and one was downregulated in T1D. Six of these miRNAs were tissue-enriched (blood cells, gastrointestinal, nerve, and thyroid tissues). Six miRNAs with the largest difference in plasma, five up-regulated (hsa-miR-101-3p, hsa-miR-135a-5p, hsa-miR-143-3p, hsa-miR-223-3p and hsa-miR-410-3p (novel for T1D)) and one down-regulated (hsa-miR-495-3p), with P-values below 0.01, were selected for further in-silico analyses. AKT1, VEGFA and IGF-1 were identified as common targets. In conclusion, 28 of the investigated miRNAs were differently regulated in long-duration T1D in comparison with controls. Several associations with cancer were found for the six miRNAs with the largest difference in plasma. Circulating miRNAs are potential biomarkers and treatment targets with clinical implications for diabetes-related complications.

LC-MS/MS proteomics on glomeruli from individuals with extreme duration of type 1 diabetes

Project description:Diabetic nephropathy (DN) is a major cause of end-stage renal disease and has limited therapeutic options to prevent its progression. To identify novel therapeutic strategies, protective factors for DN were studied using proteomics on glomeruli from 50-year medalists (individuals with extreme duration, ≥50 years, of diabetes in Joslin Medalist Study) without DN and those with histologic signs of DN. Many enzymes in the glycolytic, sorbitol, methylglyoxal and mitochondrial pathways were elevated in individuals without DN. We also identified that pyruvate kinase M2 (PKM2) expression and activity were upregulated. We then looked into the mechanisms and further validated the findings in vivo (Podocyte-specific Pkm2 KO mice model) and in vitro (cultured podocytes).

Project description:Circulating microRNAs are linked to the onset and progression of type 1 diabetes mellitus (T1DM), making them potential biomarkers for the disease. In this study, we employ a multiplatform sequencing approach to analyze circulating microRNAs in an extended cohort of individuals with T1DM from the INNODIA consortium. Our findings reveal that a set of microRNAs located within the T1DM susceptibility chromosomal locus 14q32 distinguishes two subgroups of individuals. To validate our results, we conduct additional analyses on a second cohort of T1DM individuals, confirming the identification of these subgroups, which we have named Cluster A and Cluster B. Remarkably, Cluster B T1DM individuals, who exhibit increased expression of a set of 14q32 miRNAs, show better glycaemic control and display a different blood immunomics profile. Our findings suggest that this set of circulating microRNAs can identify two different T1DM subgroups with distinct blood immunomics at baseline and clinical outcomes during follow-up.

High altitude illness could happen in individuals who did not get acclimatization. Unraveling the molecular underpinnings of altitude acclimatization would help people to understand the beneficial response of body to high altitude hypoxia and disturbed biological process in un-acclimatized individuals. Here, we measured physiological adjustments and circulating microRNAs (cmiRNAs) profiles of individuals exposed to high altitude to explore the altitude acclimatization in humans.
