Project description:Budd-Chiari syndrome (BCS) results in sinusoidal congestion and hepatocyte apoptosis, which can further progress to liver fibrosis, cirrhosis, and hepatocellular carcinoma. Endovascular (EV) treatment has been the primary therapeutic method and achieved excellent outcomes. However, whether EV treatment could reverse liver cirrhosis in patients with BCS is still unclear. To investigate the effect of endovascular treatment on liver cirrhosis in patients with Budd-Chiari syndrome, we performed gene expression profiling analysis of the liver from patients with or without EV treatment.

Alterations in fecal metagenomics and serum metabolomics indicate management strategies for patients with Budd-Chiari syndrome

Project description:OBJECTIVES: We investigated the effects of gut microbiota and serum metabolic levels on the patients with Budd-Chiari syndrome (B-CS), and their guidance for clinical management strategies.DESIGN: 214 B-CS subjects including untreated case (n=93) and treated case (n=121), were enrolled in this study. Gut microbiota and serum metabolome were analyzed using shotgun metagenomics and liquid chromatography mass spectrometry (LC-MS). 41 controls were included.RESULTS: The gut microbiota in B-CS patients showed greater abundance of Campylobacter and lower Saccharomyces, Deinococcus and Thiomonas (P < 0.05). 13 differential metabolites such as taurocholate and (R)−3−Hydroxybutyric acid were identified in B-CS patients. The random forest (RF) models showed that serum metabolome could effectively identify B-CS from healthy controls and RF-metabolomics exhibited perfect discrimination (AUC = 100%, 95% CI: 100% – 100%), which was significantly higher than that achieved by RF-metagenomics (AUC =58.48%, 95% CI: 38.46% – 78.5%). The results of association analysis showed that Campylobacter concisus and taurocholate were significantly positively correlated in B-CS patients with clinical manifestations (P < 0.05). Moreover, Actinobacteria was significantly increased in untreated B-CS patients vs. treated B-CS patients (P < 0.05). Campylobacter and Veillonella were significantly increased in treated B-CS patients vs. health controls (P < 0.05). In addition, Fewer discrepant microbes were found between treated and untreated groups for the patients without clinical manifestation.CONCLUSIONS: We identified major alterations in the gut microbiota and serum metabolome of B-CS patients. Alterations in fecal metagenomics and serum metabolomics guided management strategies for the patients with Budd-Chiari syndrome.

With an outer diameter of 0.23 mm it is capable of reaching peripheral vessels for endothelial sampling, and favourably compares to other approaches with stent retrievers or similar tools. Here, we have collected samples from liver and kidney vessels from swine and performed RNA-sequencing to validate the sample quality.

VE-cadherin is essential for trophoblast migration and endovascular invasion

Project description:Vascular endothelial (VE-)cadherin is a homotypic adhesion protein that is expressed selectively by ECs in which it enables formation of tight vessels and regulation of vascular permeability. Since VE-cadherin is also strongly expressed in placental trophoblasts, it is a prime candidate for a molecular mechanism of vascular mimicry by those cells. Here, we show that the VE-cadherin is required for trophoblast migration and endovascular invasion into the maternal decidua. VE-cadherin deficiency results in loss of spiral artery remodeling due to a lack of invasive trophoblasts, leading to decreased flow of maternal blood into the placenta, fetal growth retardation and death. Loss of trophoblast invasion prevents decidualization, extracellular matrix remodeling, and immune cell clearance. These studies identify VE-cadherin as essential for trophoblast migration and coordination of decidual changes during endovascular invasion. They further suggest endothelial proteins such as VE-cadherin that are expressed by trophoblasts may play functionally distinct roles that do not simply mimic those in ECs.
